[Efficacy of percutaneous transluminal coronary angioplasty for cardiac event after coronary bypass surgery].

In 31 cases with previous coronary artery bypass graft (CABG) surgery, 46 percutaneous transluminal coronary angioplasty (PTCA) attempts were performed, including 21 angioplasty in vein grafts (success rate 95.7%). Overall success rate was 87.0%. No major complication occurred, but one reperfusion arrhythmia (ventricular tachycardia) in a case of AMI. PTCA was so effective and useful for cardiac events in cases with previous CABG surgery with low risk and minor complication.

[Closure of median sternotomy incision with EPTFE sutures].

EPTFE (CV-0) suture was utilized for closure of median sternotomy incision in 100 patients under 6 years of age who underwent open heart surgery. During 3 years follow up, except for one dislocation of sternum, all patients have healed without complication. EPTFE suture is swanged to a needle that has the same diameter as the thread, which reduces bleeding from the needle hole. It is radiologically lucent, and therefore does not disrupt chest X-ray, angiography or MRI investigations, in contrast to sternal wires. In addition, in the case of life threatening postoperative complication, EPTFE suture allows rapid and simple access to the heart, without the need to cut wires. We concluded that EPTFE suture can be utilized for closure of median sternotomy in patients under the age of six, without an increased risk of postoperative or long-term complications.

Alpha-tocopherol supports the survival and neurite extension of neurons cultured from various regions of fetal rat brain.

The effects of alpha-tocopherol on neurons of fetal rat brain were studied in tissue culture. alpha-Tocopherol supported the survival and enhanced neurite extension of neurons cultured from cerebral cortex, hippocampus, striatum, and septum. The survival effect was observed at concentrations of 10(-9) to 10(-6) M. More than 60% of the cortical neurons could survive for 9 days at 10(-7) or 10(-6) M alpha-tocopherol, whereas almost all neurons died within 2 days in its absence. These results suggest that alpha-tocopherol may be involved in mechanisms regulating neuronal survival and neurite formation in the central nervous system, and indicate that alpha-tocopherol may be an excellent reagent to facilitate neuronal survival in culture.

Characterization of macrophage colony-stimulating factor in body fluids by immunoblot analysis.

We characterized the molecular species of human macrophage colony-stimulating factor (hM-CSF) found in serum and urine, using immunoblot analysis after partial purification on an antibody-bound affinity column. Although antibodies were prepared using the recombinant product of the large form of hM-CSF with a molecular weight (MW) of 85 Kd as the antigen, this immunoblot system was also capable of detecting the small form of hM-CSF with a MW of 40 to 60 Kd. A single band with a MW of 43 Kd, which reacted with anti-recombinant hM-CSF IgG but not with control IgG, was found when serum and urine from normal adults underwent electrophoresis on reduced sodium dodecyl sulfate-polyacrylamide gel and subsequent immunoblotting. This band represented a subunit of the large form of hM-CSF, because the large form of hM-CSF is a homodimer of a subunit with a MW of 43 Kd and the small form of hM-CSF is a homodimer of a subunit with a MW of 20 to 30 Kd. Analysis of serum and urine from leukemic patients and pregnant women, who had higher serum levels of hM-CSF than normal adults, showed only a single band with a MW of 43 Kd as a hM-CSF-specific molecule. These results suggest that the large form of hM-CSF is the major species in human body fluids.

Heparin-releasable platelet factor 4 in patients with coronary artery disease.

Recently, platelet factor 4 (PF4) release by heparin (heparin-releasable PF4) has been examined as a useful marker of the interaction between the substances liberated from circulating platelets and the vascular endothelium. We compared the plasma levels of PF4 and beta-thromboglobulin (beta-TG) after intravenous heparin injection in patients with coronary artery disease (CAD) and normal control subjects. We also studied the effects of low-dose aspirin (81 mg/day) on the plasma level of heparin-releasable PF4 in the CAD patients. Blood samples were obtained before and 5 min after the intravenous injection of heparin (1,000 IU) from 23 patients with CAD and 15 normal control subjects. Although the plasma beta-TG level remained unchanged after heparin injection, the plasma PF4 level markedly increased in both groups. There was a significant difference in plasma PF4 levels at 5 min after heparin injection between the CAD group (100.1 +/- 38.1) and the control group (61.0 +/- 24.0) (p less than 0.01). The PF4/beta-TG ratio after heparin injection was also higher in the CAD group than in the control group (p less than 0.01). There was a correlation between the PF4/beta-TG ratio after heparin and the Gensini CAD score, which defines the severity of coronary atherosclerosis (r = 0.489, n = 23, p less than 0.01). Low-dose aspirin was administered to 11 CAD patients for 246.0 +/- 28.8 days. Blood samples for the assay of PF4 and beta-TG were obtained as stated above, and platelet aggregation, thromboxane B2 (TxB2), and 6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha) levels were also measured before and during aspirin administration.(ABSTRACT TRUNCATED AT 250 WORDS)

Pattern of bone loss of the proximal femur: a radiologic, densitometric, and histomorphometric study.

The pattern of bone loss in the proximal femur was studied in 141 cadaveric femora from 36 women and 39 men ranging from 27 to 89 years of age. Bone mineral density (BMD) of the femoral neck, greater trochanter, Ward's triangle, and tensile and compressive stress regions were measured by dual photon absorptiometry. Radiographs were graded by Singh's method. Histomorphometry of tensile and compressive trabecular areas was performed on ground midsection of the methylmethacrylate-embedded whole proximal femur. Although Singh index had some correlation with BMD of the femoral neck, the various BMD measurements showed that all regions lost bone to the same extent; in particular, no selective bone loss was found at low stress regions (tensile trabeculae or Ward's triangle). Histomorphometry revealed that most of the trabecular bone loss of the proximal femur is attributable to a decrease in thickness of individual trabeculae.

Rhodamine B inhibition of glycosaminoglycan production by cultured human lip fibroblasts.

Rhodamine B (Rh B) is a dye which is used in cosmetics such as lipsticks. We investigated the effect of the dye on the metabolism of [3H]glucosamine-labeled glycosaminoglycans ([3H]-GAG) in confluent cultures of human lip fibroblast KD cells. It was found that Rh B at 10 micrograms/ml and above significantly decreased the accumulation of [3H]-GAG in both the cell layer and the medium after a 24-hr culture. This Rh B (50 micrograms/ml) effect was recognized in the cell layer and the medium after 8 and 24 hr, respectively, and longer. The Rh B at the 25 micrograms/ml-induced decrease in the [3H]-GAG accumulation was reversible in both the cell layer and the medium. On the other hand, the release of [3H]-GAG from the cell layer was unaffected by Rh B. A characterization of [3H]-GAG revealed that all components of the GAG including hyaluronic acid, heparan sulfate, chondroitin 4- and/or 6-sulfate, and dermatan sulfate were significantly decreased by 50 micrograms/ml Rh B in both the cell layer and the medium. Rh B significantly decreased the accumulation of [3H]-GAG in the presence of either 10 microM cycloheximide or 1 mM p-nitrophenyl-beta-D-xyloside, suggesting that Rh B inhibited the sugar chain formation rather than core protein synthesis. Although the number of confluent KD cells was significantly decreased by Rh B at 10 micrograms/ml and above, this Rh B effect was much weaker than that on the [3H]-GAG accumulation. The release of lactate dehydrogenase from the cell layer was significantly increased by 100 micrograms/ml Rh B but not by the dye at 75 micrograms/ml and below. From these results, it was suggested that Rh B decreases the GAG content of human lip fibroblasts through a functional suppression of polysaccharide chain formation in the process of the GAG production.

Inhibitory effect of rhodamine B on the proliferation of human lip fibroblasts in culture.

The effect of the cosmetic dye rhodamine B on the proliferation of human lip fibroblasts (KD cells) was investigated in a culture system. Rhodamine B at 25 micrograms/ml and above significantly decreased the number of the cells after a 72 h culture. A time course study revealed that 50 micrograms/ml of rhodamine B-induced decrease in the cell number occurred after 48 h and longer, suggesting that the dye inhibited cell proliferation without a decrease in cell attachment. The detachment of [3H]thymidine-labeled cells from the monolayer was unaffected by rhodamine B at 100 micrograms/ml and below. The incorporation of [3H]thymidine and [14C]leucine into the acid-insoluble fraction of the cell layer was significantly inhibited by 50 micrograms/ml rhodamine B treatment. Histologically, the damage of KD cells was not marked, however, a degenerative change of nuclei and an irregular shape of the cells as well as a decrease in the cell number were caused by 50 micrograms/ml rhodamine B. Rhodamine 6G caused a severe damage of the cells, and rhodamine B significantly decreased the cell number; rhodamine 123 had no significant effect; rhodamine 116 significantly increased the cell number. Furthermore, rhodamine B decreased the number of both vascular endothelial cells from bovine aorta and vascular smooth muscle cells from murine aorta after a 72 h culture. It is concluded that rhodamine B inhibits the proliferation of human lip fibroblasts. This rhodamine B effect may be a warning sign for the dye toxicity.

Expression of chicken integrin beta 1 subunit in rat PC12 cells.

We transfected rat pheochromocytoma (PC12) cells with a cDNA encoding chicken integrin beta 1 subunit. The chicken integrin beta 1 subunit produced in stable transfectants associated with two major alpha subunits of rat integrins to form interspecific chimeric receptors. These receptors mediated cell spreading and initial neurite outgrowth on laminin as did corresponding endogenous integrins, although they were slightly less effective in inducing cell adhesion to laminin. These results indicate that chicken integrin beta 1 may functionally substitute for beta 1 subunit of rat integrins in PC12 cells. Apparently, the structure of the integrin beta 1 subunit is highly conserved in the evolution of these species.

Repression of integrin beta 1 subunit expression by antisense RNA.

A quail cell line (QT6-c) was co-transfected with pTEX vector expressing RNA complementary to chicken integrin beta 1 subunit mRNA (Anti-Int) and pRSVneo vector by a calcium phosphate method. Transfectants showing reduced expression of quail integrin beta 1 subunit were selected with an immunoblot assay, and a few positive clones were examined in detail. Northern blot and immunoblot analyses revealed that the Anti-Int caused a clear reduction of the transcript encoding integrin beta 1 subunit depending on culture conditions. The number of cell surface integrins also decreased in proportion to the decrement of the total amount of integrin beta 1 subunits. When one transfectant (QA23) was cultured in a serum-free medium, cell shape changed from fibroblast-like to neuron-like morphology accompanied by a low growth rate, and the cells did not form focal contact on fibronectin. A similar morphological change occurred in QT6-c cells when the cells were infected with Rous Sarcoma virus, which could produce the Anti-Int. The QA23 cells did not attach to fibronectin as efficiently as did the original QT6-c cells. These data suggest that reduced expression of integrin beta 1 subunit affects cell growth as well as cell morphology by disordering the interaction between integrins and matrix proteins and/or cytoplasmic proteins.

Isolation and identification of the new metabolites of 1-[bis(4-fluorophenyl)-methyl]-4-(2,3,4-trimethoxybenzyl)piperazine dihydrochloride (KB-2796) from rat bile, urine and feces.

The metabolites of 1-[bis(4-fluorophenyl)methyl]-4-(2,3,4-trimethoxybenzyl)piperazine dihydrochloride (KB-2796) in the bile, urine and feces in rats were investigated after oral administration of [methine-14C], [benzyl-14C] and unlabelled KB-2796. Their structures were characterized by thin-layer chromatography, mass spectrometry, proton nuclear magnetic resonance and comparison with synthesized authentic compounds. The main pathways of biotransformation of KB-2796 in rats were: (a) O-demethylation at each methoxy group of the trimethoxybenzyl moiety, (b) N-dealkylation at 1 and 4-position of the piperazine ring and (c) hydroxylation at 5-position of the 2,3,4-trimethoxyphenyl ring.

A new haplotype of the beta-globin gene complex, Hbbw1, in Chinese wild mouse.

A new pattern was observed in the electrophoretic survey of the hemoglobin beta chain (Hbb) in Chinese wild mice, Mus musculus. The electrophoretic mobility of the major component of the new Hbb was identical to that of Hbbs on cellulose acetate plate, although it was almost identical to that of Hbbd or Hbbp on acrylamide gel. This suggests that the major component of Chinese Hbb has a unique primary structure. A minor component of the new Hbb was completely different from that of the other three Hbb haplotypes well known. These results indicate that the Hbb-b1 and Hbb-b2 of the new Hbb haplotype, assigned Hbbw1, are unique genes in their molecular structure. So far, Hbbw1 has been observed in northwestern China.

ECG analysis of golden hamsters exposed to long-term -Gz conditions: ordinary and pathological findings.

Changes of ECG due to long and intense -Gz acceleration was investigated in anesthetized Syrian golden hamsters. The R-R interval decreased (heart rate increased) slightly at the very initial phase of steady -Gz load. However, the R-R interval increased during the rest of the whole steady -Gz period. The P-R interval increased with a smaller -Gz load (-4 and -6 Gz), but it decreased with a larger -Gz (-8 and -10 Gz). The size of the QRS complex was measured as R amplitude. It decreased in all -Gz conditions. Effects were a little different for smaller and larger -Gz, as was observed in the case of P-R changes. Pathological ECGs were frequently observed. They were: (1) Arrhythmia of heart rate with or without atrio-ventricular blocks. (2) Abnormal P wave; e.g., splitting, flattening, or abolition of P wave. (3) Alteration of the QRS complex in amplitude or contour. (4) Changes in ST-T components. In spite of all these changes, however, it should be noted that several Syrian golden hamsters could tolerate up to -10 Gz for as long as 1,000 s.

[Cross reactive antigens of Acholeplasma laidlawii and L-form of Staphylococcus aureus].

We demonstrated that the membrane of Acholeplasma laidlawii PG8 and L-form of Staphylococcus aureus, both of which induce cellular immunity in BALB/c mice, were antigenically related each other. Foodpad responses of the mice immunized with a mixture of either antigen and Freund's complete adjuvant showed clearly a cross reaction when challenged with the other antigen. Cross responses to incorporate 3H-thymidine to the spleen lymphocytes of the mice immunized with either antigen occurred in the presence of the other antigen. Furthermore, the purified T cells, but not B cells, of the spleen were activated in the presence of antigen-presenting cells. These antigens existing in the membrane fractions of both microorganisms were purified by Razin's method. Finally, these membrane components of A. laidlawii and L-form of S. aureus were subjected to gel electrophoresis and transferring to nitrocellulose membrane and used to stimulate the spleen lymphocytes of the mice immunized with A. laidlawii or of non-immunized mice. The fractions representing molecular weights of approximately 45 kD, 25 kD, and 13 kD of both microorganisms consistently stimulated the lymphocytes of the immunized mice but not those of non-immunized mice.

[Significance of determination of the blood antibody level in Chlamydia trachomatis infection of the uterine cervix].

Kits for measurement of C. trachomatis specific antibody are not commercially available, but of the significance of determination of the serum antibody level has not been clarified. In this study, we first determined the C. trachomatis antibody level in patients considered to be free of C. trachomatis infection. Then, we evaluated the relationship between the presence or absence of antigen with the antibody level in patients at our department who were available for antigen examination. Anti-C. trachomatis IgA and IgG antibodies were detected in all age groups with a peak frequency in the 20's expect that IgA antibody was absent in the umbilical blood (IgG antibody was detected also in the umbilical blood). Concerning the relationship between the presence or absence of C. trachomatis antigen and the serum antibody levels, both IgA and IgG antibodies were significantly different (chi 2 test, p less than 0.01) between the antigen-positive group and the antigen-negative group. Because females infected by C. trachomatis were more often asymptomatic than infected males, and because antigen tests may produce false negative results, determination of the antibody level must be included in the clinical tests. Determination of the antibody level is considered to be valuable also in screening for mother-child infection and asymptomatic infection.

[Follow-up study in a case of traumatic epilepsy with increased accumulation of 123I-N-isopropyl-p-iodoamphetamine].

Recent studies suggested the potential use of 123I-N-isopropyl-p-iodoamphetamine (IMP) for the assessment of pathophysiology of epilepsy. We studied a case of epilepsy with increased accumulation of IMP. Brain SPECT scan of IMP was performed in a case of 31-year-old male with traumatic epilepsy. Increased accumulation of IMP was observed in the left temporal area on both early (20 min after injection) and delayed (3 hr) images of studied 6 weeks after the head trauma. X-CT did not show any abnormalities. The accumulation of IMP decreased as the symptom of the patient improved. The SPECT study performed 3 months after the accident showed slightly decreased uptake in the left temporal area on the early image with homogeneous uptake in the delayed image. These findings suggested the altered retention mechanism of IMP in this case.
