Specific inhibition of glyceraldehyde-3-phosphate dehydrogenase by koningic acid (heptelidic acid).

A sesquiterpene named koningic acid has been isolated from a strain of Trichoderma koningii as a potent inhibitor of ATP generation in the glycolytic pathway. From experiments with both cultured mouse carcinoma FM3A cells and isolated enzymes, it was shown that koningic acid is a specific inhibitor of glyceraldehyde 3-phosphate dehydrogenase that catalyzes the conversion of glyceraldehyde 3-phosphate to 3-phosphoglycerate.

Effect of cilostazol on platelet aggregation and experimental thrombosis.

A new antithrombotic drug, cilostazol (6-[4-(1-cyclohexyl-1 H-tetrazol-5-yl)butoxy]-3,4-dihydro-2(1H)-quinolinone, OPC-13013) was studied for its inhibitory effect on platelet aggregation in vitro in various experimental animals and man and in dogs ex vivo, for its effect to disperse platelet aggregates in vitro in rabbits and man and for its antithrombotic effect in vivo using its effect to prevent death due to the formation of pulmonary thrombi in mice. Cilostazol produced a potent inhibition of platelet aggregation both in vitro and ex vivo and a dispersion of platelet aggregates in vitro. The mode of action of cilostazol was different from that of acetylsalicylic acid (ASA) in that cilostazol inhibits not only secondary platelet aggregation but also primary platelet aggregation induced by aggregating agents such as adenosine diphosphate (ADP). The drug potently prevented death due to pulmonary thrombosis by platelet aggregates in mice in vivo. Unlike ASA which prevented only death due to collagen-induced platelet aggregation, cilostazol prevented both collagen- and ADP-induced platelet aggregation. These results suggest that cilostazol is a promising antithrombotic drug.

How to identify the Yakuza, Japanese racketeers--their sociology, criminology and physical characteristics.

International crimes such as smuggling and business fraud are increasing. It is possible that members of the Yakuza (Japanese racketeers or gangsters) may be involved in these criminal activities. The Yakuza is one of the best organized criminal groups in the world. It assumes a family-like form with fictitious kinship ties while making a living through organized crime. The sociological and criminological background of the Yakuza and their physical characteristics namely tattooing, digital amputation, and penile spheres are described and discussed so that the Yakuza can be identified even by foreign forensic pathologists.

Ultrastructure of the invasion of human hair in vitro by the keratinophilic fungus Microsporum gypseum.

The pattern of invasion of human hair in vitro by the dermatophyte Microsporum gypseum was studied by transmission and scanning electron microscopy. Mycelia that invaded the hair cortex through the edge of cuticles showed a flattened "frond" growth in contrast to the filamentous form seen on ordinary laboratory media. The frond cells were characterized by the presence of vesicles formed by invaginations of plasmalemma, and lomasomes were prominent in the region adjacent to the hard keratinized tissue of the hair cortex being degraded as well. The initial perforating organ, which originated from the frond mycelium, appeared as an enlarged spherical cell which integrated with the laterally branched hyphae, as revealed by analysis of a three-dimensional model reconstructed from a series of sections. The fully developed perforating organ consisted of a column of wide and short cells which penetrated perpendicularly through the hair cortex. Through the medulla the filamentous hyphae had grown profusely in a longitudinal direction. Our studies confirm earlier light microscope observations and provide new ultrastructural details on the development of the eroding frond and the perforating organ.

Variance of ploidy in Candida albicans.

Determination of ploidy was performed on isolates of Candida albicans from clinical sources by measuring nuclear DNA content with fluorescent microscope photometry. By this criterion and UV irradiation survival experiments, haploid, diploid, and tetraploid strains were identified in this organism. The dimensions of nucleus-associated organelles (equivalent to spindle pole bodies) in these strains increased as a function of ploidy number.

Mechanisms of structural vascular changes in genetic hypertension: analyses on cultured vascular smooth muscle cells from spontaneously hypertensive rats.

1. The basic mechanism underlying the structural vascular changes occurring in hypertension was studied in cultured aortic smooth muscle cells (SMC) obtained by an explant method from spontaneously hypertensive rats (SHR) and stroke-prone SHR (SHRSP) and compared with that in normotensive Wistar--Kyoto (WKY) rats. 2. The growth rate of SMC from SHR and SHRSP at the age of 2.5--11 months was greater than that of SMC from WKY rats even after repeated passages. 3. [3H]Thymidine and [14C]leucine incorporation, and ornithine decarboxylase (ODC) activity of SMC were increased in SHR and SHRSP in comparison with WKY rats. 4. The application of isoprenaline but not noradrenaline to the culture media increased ODC activity acutely in SMC from WKY rats and this increase was blocked by propranolol. 5. These results indicate that SMC from SHR and SHRSP are more prone to proliferate than those from WKY rats and that a beta-adrenergic neurohumoral mechanism accelerates SMC growth independently of blood pressure.

Genetic markers in spontaneously hypertensive rats.

Establishment of various models for hypertensive diseases such as spontaneously hypertensive rats (SHR) and stroke-prone SHR (SHRSP) clarified the importance of genetic factors in the pathogenesis of these diseases and further accelerated studies on their genetic mechanisms. Although various biochemical abnormalities have been detected and can be used as biochemical markers in these models, they have not been always closely related to blood pressure in F2 generation obtained by cross breeding between SHR and normotensive Wistar-Kyoto rats. Recent studies indicate that biomembrane abnormalities detected in erythrocytes and other membranes of SHR and SHRSP may not only be biochemical markers but also related to the pathogenesis of hypertensive diseases.

[Vaginal fungi in toxemia of pregnancy (author's transl)].

In order to prevent the transfer of the fungi in maternal vagina into a neonate through transvaginal delivery, we examined the presence of vaginal fungi in the patients with toxemia of pregnancy occurring chiefly from the later stages of pregnancy and made an analysis from the quantitative aspect of fungi (number of colonies produced on Mizuno-Takada medium). The results obtained are as follows: 1) The detection rate of vaginal fungi was evidently so high as 37.1% in toxemia group compared with 27.6% in non-toxemia group (p less than 0.01). And the detection rate of vaginal Candida albicans (hereinafter: C. albicans) also proved to be higher tendency in toxemia group. 2) When the toxemia patients were divided into mild cases and severe ones for comparison, the detection rate of vaginal fungi and that of vaginal C. albicans were both higher in the severe case group. Particularly in the group which had the symptoms of toxemia at the examination time of vaginal fungi there was seen the higher rate. 3) From the quantitative aspect of vaginal fungi it is evident that there were more cases with over 51 colonies in the group showing the symptoms of toxemia at the time of fungi examination than in the group showing no such symptoms at the examination and the group of non-toxemia (p less than 0.01).

Potentiation by dipyridamole of the inhibition of guinea-pig ileum twitch response caused by adenine derivatives.

The inhibition actions of adenosine and adenine nucleotides, such as ATP, AMP and cyclic AMP, on contractions of guinea-pig from ileum induced by transmural stimulation were potentiated by dipyridamole, whereas those of inosine, morphine and tetrodotoxin were not affected. Tritium activity, accumulated during incubation of the ileal segments with [3H]adenosine, was reduced by dipyridamole. The adenosine added was degraded to inosine and then hypoxanthine during incubation, with the ileal segments, thereby restoring the twitch response of the ileal segments. In the presence of dipyridamole, the degradation of adenosine and the recovery of the twitch response was retarded. Thus, dipyridamole may potentiate the inhibitory actions of adenosine and adenine nucleotides on the twitch response of the ileum by inhibiting both the accumulation and the degradation of adenosine.

Effects of methylxanthines and imidazole on the contractions of guinea-pig ileum induced by transmural stimulation.

Methylxanthines (10(-5) to 10(-3)M) were found to increase the amplitude of contractions of guinea-pig ileum induced by transmural stimulation but to inhibit those induced by acetylcholine or histamine. The order of the abilities of methylxanthines to augment the contractile responses was theobromine greater than caffeine greater than theophylline. When the contractions were completely suppressed by reduction of the calcium content in the medium or by addition of cyclic AMP, methylxanthines restored the responses effectively, just as does addition of calcium. Methylxanthines also accelerated the release of acetylcholine from the ileum associated with stimulation. Imidazole (3 X 10(-5) to 10(-3) M) had an essentially similar effect to methylxanthines in potentiating the contractile responses and in augmenting the release of acetylcholine. The present results indicate that the potentiating effects of methylxanthines and imidazole are due to an action on the nerve terminals, not on the postsynaptic membranes or contractile elements. Therefore, it si concluded that theit potentiating actions are due to facilitation of the movement of calcium in the nerve terminals on excitation, resulting in increased release of acetylcholine, and are not due to the effect of cyclic AMP formed as a result of their inhibitory actions on phosphodiesterase.
