Dynamics of cytoplasmic organelles in the cell cycle of the fission yeast Schizosaccharomyces pombe: three-dimensional reconstruction from serial sections.

Changes in the ultrastructure of the fission yeast Schizosaccharomyces pombe during the cell division cycle were analyzed by three-dimensional reconstruction of serial section electron micrographs of freeze-substituted cells. Cytoplasmic vesicles were found at the cell ends during interphase and at the equatorial zone of cells undergoing cytokinesis. Filasomes behaved in a similar but temporally retarded way to vesicles. Microfilament(mf)-associated granules were found attached to the plasma membrane at the growing ends. Microfilaments were identified against the plasma membrane and adjacent to developing septa. From these observations it is suggested that mf-associated structures such as filasomes constitute dense knots of actin network that function in localized cell wall growth by controlling the deposition of cytoplasmic vesicles. Dictyosomes occur as tubular and fenestrated cisternae with associated cytoplasmic vesicles. They were distributed uniformly in the cytoplasm and did not change significantly during the cell cycle. Changes in the three-dimensional localization of cytoplasmic microtubules and mitochondria are also described.

Studies on proton pump inhibitors. I. Synthesis of 8-[(2-benzimidazolyl)sulfinyl]-5,6,7,8-tetrahydroquinolines and related compounds.

Many 8-[(2-benzimidazolyl)sulfinyl]-5,6,7,8-tetrahydroquinolines were synthesized and examined for their (H+ + K+) adenosine triphosphatase ATPase-inhibitory and antisecretory activities. These sulfinyl compounds could be considered to be rigid analogues of the 2-[(2-pyridyl)methylsulfinyl]benzimidazole class of antisecretory agents. All the compounds tested were potent inhibitors of (H+ + K+)ATPase. Most of the compounds also inhibited histamine-induced gastric acid secretion in rats. Among them, 8-[(5-fluoro-2-benzimidazolyl)sulfinyl]-3-methyl-5,6,7,8-tetrahydroqu inoline (XIVm) was found to have the most potent activity. The structure-activity relationships are discussed.

Studies on proton pump inhibitors. II. Synthesis and antiulcer activity of 8-[(2-benzimidazolyl)sulfinylmethyl]-1,2,3,4-tetrahydroquinolines and related compounds.

Many 8-[(2-benzimidazolyl)sulfinylmethyl]-1,2,3,4-tetrahydroquinolin e derivatives were synthesized and tested for their (H+ + K+)adenosine triphosphatase (ATPase)-inhibitory and antisecretory activities against histamine-induced gastric acid secretion in rats. These sulfinyl compounds were synthesized by the oxidation of the corresponding sulfides, which were obtained from the reaction of 8-chloromethyl-1,2,3,4-tetrahydroquinolines and 2-mercaptobenzimidazoles in the presence of potassium carbonate. All compounds tested were potent inhibitors of (H+ + K+)ATPase. Most of the compounds showed antisecretory activity. Among them, 8-[(2-benzimidazolyl)sulfinylmethyl]-1-ethyl-1,2,3,4-tetrahydro quinoline (IXa) was found to have the most potent activity. The structure-activity relationships are discussed.

Studies on gastric antiulcer active agents. II. Synthesis of tetrazole alkanamides and related compounds.

A series of tetrazole alkanamides was synthesized and tested for antiulcer activity against acetic acid-induced gastric ulcer in rats. These compounds were prepared by the reaction of tetrazole alkanoic acids and various amines by the mixed anhydride method or acid chloride method. Among them, 3-[(1-ethyl-5-tetrazolyl)methylthio]propionamide (IIn) was found to have the most potent activity. The structure-activity relationships are discussed.

Studies on gastric antiulcer active agents. III. Synthesis of 1-substituted 4-(5-tetrazolyl)thio-1-butanones and related compounds.

Many 1-substituted 4-(5-tetrazolyl)thio-1-butanones were synthesized and tested for antiulcer activity against acetic acid-induced gastric ulcer in rats. These compounds were prepared by the reaction of 5-mercaptotetrazoles and 4-halogeno-1-butanones. Among them, 1-cyclohexyl-4-(1-phenyl-5-tetrazolyl)thio-1-butanone (VIIIp) was found to have the most potent activity. The structure-activity relationships are discussed.

Effect of OPC-12759, a novel antiulcer agent, on chronic and acute experimental gastric ulcer, and gastric secretion in rats.

The antiulcer effects of OPC-12759, a novel antiulcer agent were compared with those of cetraxate in various experimental ulcer models and on gastric secretion in rats. OPC-12759 (0.3-30 mg/kg, b.i.d., p.o.) significantly accelerated the healing rate of acetic acid-induced gastric ulcer in a dose-dependent manner, while cetraxate did not. When administered orally at 0.3-30 mg/kg, b.i.d., for 7 days, pretreatment with OPC-12759 (0.3-30 mg/kg, b.i.d., p.o.) prevented the formation of acute gastric ulcers, induced by: restraint water immersion stress, aspirin, indomethacin, histamine, serotonin, platelet activating factor (PAF) and DDC. Cetraxate showed antiulcer activity against a part of the OPC-12759-positive gastric ulcer models. Given intraperitoneally at the single dosing range of 10-100 mg/kg, OPC-12759 inhibited the formation of these acute gastric ulcer models. OPC-12759 administered orally at 0.3-30 mg/kg, b.i.d., for 7 days did not inhibit basal gastric secretion in pylorus ligated rats. The results indicated that OPC-12759 possesses wide spectrum antiulcer activity as compared with cetraxate.

Nucleotide sequence of the macromomycin apoprotein gene and its expression in Streptomyces macromomyceticus.

A 1.6 kb Sph I-Sac I DNA fragment from Streptomyces macromomyceticus, expected to include the macromomycin (MCM) apoprotein gene, was sequenced. The fragment (1,556 bp) was found to include a putative promoter, an ORF directing pre-apoprotein which should be split into the leader peptide (Met1 to Gly32) and the MCM apoprotein (Ala33 to Ala144), and a putative terminator. The amino acid sequence deduced from the base sequence of the DNA is consistent with the amino acid sequence previously determined by the Edman degradation and other procedures applied to the protein, except base sequence AAC coding for Asn was found rather than Asp111 previously reported. The GC content of the 3rd letters throughout the ORF was 92% in contrast to the sum of the first and the second letters, 62%. There was a low GC content stretch of 20 bp (30% GC) at about 120 bp upstream of the ORF. The Pst I-Sph I 620 bp fragment including the low GC content stretch showed promoter activity when subcloned in a promoter probe vector. About 700 nucleotides long mRNA, which is long enough to span the ORF and the bordering regions, was identified using the Northern blot analysis. A primer extension experiment showed that the transcriptional starting point was A at 89 bp upstream of the ORF. Dot blot analysis of expression of MCM apoprotein gene indicated that the gene was expressed nearly constitutively, while production of holo MCM (the complex consisting of MCM apoprotein and a specific chromophore) depended greatly on culture conditions.

Enhanced growth rate of cultured smooth muscle cells from spontaneously hypertensive rats.

Smooth muscle cells were isolated and cultured from the aortic media of age-matched, stroke-prone and stroke-resistant spontaneously hypertensive rats (SHRSP, SHRSR:SHRs) and Wistar-Kyoto rats (WKY), and the growth rate of cells from the three strains was compared. Under electron microscopical observation the cells were identified as modified (or synthetic-type) smooth muscle cells. Cells occurring at low densities showed the same morphology for both SHRs and WKY, but cells occurring at higher densities were observed to be smaller in SHRs than in WKY. An analysis of the growth curves of cells showed a significantly enhanced replication rate in cells from SHRs compared with those from WKY, especially in the early passages. In later passages (repeated until the 9th passage), however, this distinction was not clear. These growth characteristics were also confirmed in cells from both 12- and 24-week-old, age-matched SHRs and WKY. We could not find any difference between the growth characteristics of cells from SHRSP and SHRSR. It is possible to hypothesize from these findings that the abnormality relating to hypertension (hypertension being a common characteristic of SHRSR and SHRSP) is found in the smooth muscle cells, and is reflected as an enhanced growth rate when they are exposed to mitogenic stimuli, such as in atherosclerosis.

[Healing promoting effect of proamipide, a novel drug that increases gastric defense mechanisms, on acetic acid-induced gastric ulcers in the rat].

The healing process of acetic acid-induced gastric ulcers was studied for over 180 days after ulcerogen injection, and the effect of a new antiulcer agent, proamipide, was also assessed by histological measurements. In the control group, rapid reduction in the macroscopic ulcer index and increment in histological indices such as healing, regeneration of defective mucosa and proliferation of granulation tissue were observed from the 5th day to the 60th day after the ulcer induction followed by significant aggravation on the 120th and 140th day, suggesting the recurrence or relapse of gastric ulcers may occur at about 120 to 140 days after acetic acid administration. Proamipide (20 mg/kg/day, p.o.) decreased all of these indices significantly from those in the control group. This indicates that proamipide remarkably promotes the healing process in acetic acid-induced gastric ulcers, the regeneration of the injured mucosa and also the formation of granulation tissue. The glandular index in the surrounding areas of the regenerated mucosa was lower than that in the central part in the control animals and this difference was decreased by proamipide intake, suggesting an imbalance in the regenerated mucosa of the scar may have significant roles in the induction of the recurrence or relapse of gastric ulcers.

Gastric mucosal protection by OPC-12759, a novel antiulcer compound, in the rat.

OPC-12759, 2-(4-chlorobenzoylamino)-3-[2(1H)-quinolinon-4-yl]-propionic acid, was studied for its efficacy to prevent the gastric mucosal damage induced by several necrotizing agents. Experiments were also performed to elucidate the mechanism of this mucosal protective activity. OPC-12759 dose dependently prevented the formation of mucosal necrosis induced by absolute ethanol, 0.2 N NaOH or 0.6 N HCl. PGE2 was also shown to prevent the gastric mucosal erosion induced by necrotizing agents. The mucosal protective effect of OPC-12759 was completely counteracted by pretreatment with indomethacin while that of PGE2 was not. In addition, OPC-12759 given alone increased the generation of gastric mucosal PGE2-like activity. OPC-12759 dose dependently reduced the volume, acid output and pepsin output of the gastric juice in pylorus-ligated rats. The inhibitory effect of OPC-12759 but not of cimetidine or atropine on gastric secretion was also abolished by concurrent administration of indomethacin. These findings suggest that the mucosal protective effect and antisecretory effect of OPC-12759 presumably result from enhancement of the generation of endogenous PGs.

Occurrence of ploidy shift in a strain of the imperfect yeast Candida albicans.

A clinical isolate of Candida albicans, a member of the Fungi Imperfecti, was polyploid as shown by the fact that it contained two kinds of nuclei, one of diploid and one of tetraploid DNA content. These determinations were made by fluorescence microscopy-photometry. The nucleus-associated organelles (NAOs), or spindle pole bodies, of yeast cells in this isolate were classified into two groups, one diploid and the other tetraploid, according to their dimensions as determined by serial thin-sectioning electron microscopy. A ploidy shift from diploid to tetraploid was found in individual cells of a culture of this isolate undergoing diphasic growth in minimal salts medium. A process of shift-down or reduction of ploidy from tetraploid to diploid was also observed by electron microscopy during these growth conditions: this appeared to occur in large cells which showed multiple spindle formation during nuclear division, a phenomenon apparently similar to the process of meiosis II during sporogenesis of Saccharomyces cerevisiae, but differing in that it produces diploid daughter nuclei by the vegetative process.

Mitosis in the fission yeast Schizosaccharomyces pombe as revealed by freeze-substitution electron microscopy.

Nuclear division in Schizosaccharomyces pombe has been studied in transmission electron micrographs of sections of cells fixed by a method of freeze-substitution. We have found cytoplasmic microtubules in the vicinity of the spindle pole bodies and two kinds of microtubules, short discontinuous ones and long, parallel ones in the intranuclear mitotic spindle. For most of the time taken by nuclear division the spindle pole bodies face each other squarely across the nuclear space but early in mitosis they briefly appear twisted out of alignment with each other, thereby imparting a sigmoidal shape to the bundle of spindle microtubules extending between them. This configuration is interpreted as indicating active participation of the spindle in the initial elongation of the dividing nucleus. It is proposed that mitosis is accompanied by the shortening of chromosomal microtubules simultaneously with the elongation of the central pole-to-pole bundle of microtubules of the intranuclear spindle. Daughter nuclei are separated by the sliding apart of interdigitating microtubules of the spindle at telophase. Some of the latter bear dense knobs at their ends.

The distribution and content of ubiquinone in foods.

We measured ubiquinone (UQ)-9 and UQ-10 content in various foods using high performance liquid chromatography. UQ-9 was detected in cereals, some vegetables and their products. Corn oil and wheat germ had large amounts of UQ-9 in particular. UQ-10 was detected in meats, fishes, pulses, nuts, dairy products and various vegetables. Migratory fishes, rapeseed oil and soybean oil had considerably large amounts of UQ-10.

Three-dimensional behaviour of mitochondria during cell division and germ tube formation in the dimorphic yeast Candida albicans.

This study was done to correlate mitochondrial behaviour with nuclear behaviour and cell division as well as with the germ tube formation in the dimorphic yeast Candida albicans. Three-dimensional reconstruction of electron micrographs of serially sectioned cells of the three strains was used to determined the morphological and quantitative relationships between the structures. The results suggested that at the time of entry into the bud a few mitochondria fused into a single giant one, which fragmented during mitosis and resumed a single giant form before cytokinesis, and was then partitioned into two parts. This tendency was also shown during germ tube formation. Quantitative analysis has established that growth of organelles such as the nucleus and mitochondria closely followed total cell growth, the ratio of organelle volume to total cell volume being held relatively constant.
