Endotracheal neurilemoma with a lymphoid cuff. An ultrastructural and immunohistochemical study.

A neurilemoma occurred in the trachea of a 63-year-old woman complaining of dyspnea. Microscopically, the excised tumor was composed predominantly of cellular areas of Antoni type A tissue and had a peripheral lymphoid cuff. The schwannian origin was confirmed both immunohistochemically and ultrastructurally. This tumor without nuclear palisading and Verocay bodies differed somewhat from a conventional soft tissue neurilemoma and closely resembled a particular form of neurilemoma of the stomach, previously reported as a unique entity. Thus, this type of neurilemoma does occur in sites other than the gastrointestinal tract.

Proton nuclear magnetic resonance study on the aromatic amino acid-guanine nucleotide system: effect of base methylation on the stacking interaction with tyrosine and phenylalanine.

The stacking interactions of tyrosine methylester (TyrOMe)-guanosine-5'-monophosphate (GMP), TyrOMe-7-methylguanosine-5'-monophosphate (m7GMP), phenylalanine methylester (PheOMe)-GMP and PheOMe-m7GMP pairs in neutral buffer solution have been studied by proton nuclear magnetic resonance (1H-NMR). The H8 proton signal of GMP showed no noticeable temperature dependence, while the signals of other protons showed usual dependences arising from the ring stacking interaction with aromatic amino acids. The results can be interpreted in terms of the intramolecular C-H ... O hydrogen bonding and ring stacking. Complex formations in 1:1 molar ratio were deduced for all pairs from their Job plots. The association constant for each pair was obtained by analysis of the Scatchard plot. Further, the van't Hoff plot provided thermodynamic parameters of the complex structure. The analyses of these data suggested that albeit the N-quaternization of GMP strengthens the stacking interaction with aromatic amino acid, the bulky methyl group in m7GMP facilitates the dissociation from the amino acid with small environmental change. The possible conformations of GMP and m7GMP in the interaction states are discussed on the basis of the coupling constants.

Crystal structure and molecular conformation of E-64, a cysteine protease inhibitor.

In order to elucidate the conformational characteristics of cysteine protease inhibitors contributing to their inhibitory activities, the conformation of E-64 (N-[N-(L-3-trans-carboxyoxiran-2-carbonyl)-L-leucyl]-agmatine), a potent inhibitor of papain, was determined by X-ray crystal structure analysis. The molecules were packed in the crystal through electrostatic forces and hydrogen bonding between the oppositely charged terminal groups and between the amide groups. Two crystallographically independent E-64 molecules both took a flattened and slightly curved structure, which is similar to that of loxistatin, a related cysteine protease inhibitor. Based on the present results, a possible inhibitory mechanism of E-64 is proposed, with reference to the binding mode observed in the crystal structure of papain-substrate analogue complex.

Induction of twinning in dairy or crossbred heifers by ipsilateral frozen embryo transfer.

Induction of twinning by ipsilateral nonsurgical transfer of frozen two or demi-embryos was attempted in 129 virgin dairy or crossbred heifers that had been kept under a stable in our station (group-1), mountain field (group-2) and private farm (group-3) conditions. Ninety seven heifers (75%) were diagnosed pregnant by rectal palpation at 35 to 60 days of gestation; 86 heifers produced 37 sets of twins and 49 single calves. Pregnancy rate of group-1 was lower compared to those of groups-2 and -3 (63%, 88%, 78%, respectively). Abortion and mortality rates of group-3 were significantly lower (P less than 0.05) than those of groups-1 and -2 (8% and 6%, 12% and 16%, 18% and 24%). Twin calves had a lower birth weight (P less than 0.05) than singles. But there was no significant difference in weight between singles and twin calves at 270-330 days of age. Dystocia and difficult delivery were not observed in females producing twins. The incidence of retained placentas in twin calving cows (17%) was higher than that of single calving cows (2%). The interval to conceive postpartum was longer in twin calving cows (95 +/- 41 days) than in single calving cows (87 +/- 26 days). Gestation length was 5.0 days shorter for twin calving cows (P less than 0.05).

Temporal properties of information extraction in reading studied by a text-mask replacement technique.

A text was replaced with a visual mask for a fixed duration every time the reader made a saccade. The threshold duration was measured when the mask was presented at the beginning of a saccade or a fixation, or at a certain delay after the onset of a fixation. The effect of the mask on reading time, as well as on the subjective legibility of the text, was also investigated. It was shown that saccadic suppression exists in reading; subjects are not affected by a mask as long as it is inserted during the saccade. The visual sensitivity is recovered only partially at the initial part of fixation and is recovered fully approximately 70 msec after the beginning of the fixation. The visual information can be extracted during a later part of the fixation period as efficiently as or even more efficiently than during the early part of the fixation.

UH series of monoclonal antibodies recognizing major histocompatibility complex class II antigen(s) of Japanese monkeys (Macaca fuscata).

Six mouse monoclonal antibodies were developed by immunization with a Japanese monkey cell line. These monoclonal antibodies, designated the UH series, reacted with large populations of peripheral B cells and monocytes, but not with T cells. The distribution of reactivities and the molecular weight of the membrane antigens recognized were similar to those of the HLA-DR monoclonal antibody; one inhibited the binding of HLA-DR. Human interferon-gamma induced increased expressions of all the UH antigen epitopes.

[A case of quartet cancer--a carcinoma of the breast followed by three synchronous cancers (kidney, thyroid and colon)].

A case of a surgically treated primary quartet cancer is reported, along with a review of the Japanese literature. A 68-year-old woman, curatively operated for a right breast cancer 6 years previously, was then diagnosed as having a left renal cell carcinoma, a thyroid cancer, and a sigmoid colon cancer as a result of a subsequent screening examination within one year. These synchronous, triple cancers were resected in a curative operation for each. Histopathologically, each neoplasm was proven to be a primary cancer lesion, and two different types (papillary and trabecular) were found in the thyroid gland.

[Clinical significance of serum NCC-ST 439 as a tumor marker for colorectal cancer].

Clinical significance of preoperative serum NCC-ST 439 level was studied in 119 cases of colorectal cancer (90 of primary, 29 cases of recurrent). The positive rates for serum NCC-ST 439 were 28.9% in primary and 65.5% in recurrent cases. The false positive rates for benign disease were 5.6%. These rates were low when compared with those for other tumor markers. The positive rates for serum NCC-ST 439 exhibited a strong correlation with wall invasion, lymph node and liver metastases. A combination assay of NCC-ST 439, CEA and CA 19-9 produced a high positive rates as 43.3% in primary and 86.2% in recurrent cases. These results demonstrate that measurement of serum NCC-ST 439 may be useful for cancer staging and improves the diagnostic rate in combination with CEA and CA 19-9.

[Comparison of high resolution computed tomography and pulmonary function tests in diagnosis of mild emphysema].

To assess the ability of high resolution CT scan and pulmonary function tests in detecting and grading mild emphysema, we correlated the high resolution CT scan and pulmonary function tests with the pathologic grade of emphysema and the destructive index of lung specimens from 42 patients undergoing thoracotomy for solitary pulmonary nodules. Using the high resolution CT scan, we could identify the pathologic grade of mild and moderate emphysema. By measuring diffusing capacity per unit alveolar gas volume (DLco/VA), it seemed to be possible to detect the mildest degree of alveolar destruction assessed by the destructive index, which was not detected by high resolution CT scan. The reason for these results seemed to be that we assessed the severity of emphysema by detecting the air space enlargement on high resolution CT scan images caused by the destruction of alveolar walls, which were detectable by measuring DLco/VA. We conclude that it is possible to detect mild emphysema using the combination of high resolution CT scan and pulmonary function tests.

Stacking interactions between protonated nucleic acid bases and aromatic amino acids: spectroscopic and structural analyses of m3CMP-tryptophan derivative complex.

As a stacking model between nucleic acid bases and aromatic amino acids, the interaction on m3 CMP-tryptophan derivative has been studied by 1H-NMR and X-ray crystal analyses. From the comparative 1H-NMR experiments using CMP and m3CMP, it is suggested that the N(3)-protonation by methylation greatly strengthens the stacking interaction with tryptophan. Parallel alignment with a separation distance of 3.38A is shown by the X-ray analysis of m CMP-tryptamine complex. The stacking mode is very similar to those observed in the complexes of indole ring with m1A and m7G.

Effects of thioamide substitution for the enkephalin conformation. Crystal structure of Boc-Tyr-Gly-Gly-Phe psi [CSNH]Leu-OBzl.

The crystals of Boc-Tyr-Gly-Gly-Phe psi[CSNH]Leu-OBzl monohydrate (C40H51N5O8S.H2O), a monothionated Leu-enkephalin analogue, were obtained with space group P2(1), a = 12.616(3), b = 9.347(2), c = 18.548(5) A, beta = 96.31(4) degrees. The structure was elucidated by X-ray diffraction analysis, and refined to the R value of 0.091 for the observed 3294 reflections. Two antiparallel molecules related by a pseudo twofold symmetry were stabilized to each other by four intermolecular hydrogen bonds. The molecular conformation was bent at the Phe residue, and the extended moiety of the Tyr-Gly-Gly fragment was almost perpendicular to that of the Phe-Leu residues. Consequently the molecule, as a whole, formed an L-shape conformation with a slightly left-handed helicity.

[A case of hypereosinophilic syndrome associated with eosinophilic endocarditis].

A 66-year-old woman, who had had bronchial asthma, was admitted to our hospital because she suffered from fever, productive cough, wheezing, dyspnea, and chest pressure sensation. Her chest X-ray showed migrating infiltration and marked cardiomegaly. Her ECG at the admission revealed abnormal Q wave and T wave inversion, though that of 3 years before had been almost normal. Hematology showed leukocytosis and eosinophilia of 8,000/mm3 without abnormal cells. All immunological tests were negative and the specific cause of the eosinophilia was unknown. 2 weeks after admission, she complained of severe chest pain suddenly and her ECG showed ST elevation on V1-4 and serum CPK level was elevated to 290 IU/l. By the thrombolytic agent and anticoagulant therapy, her symptom was lightened immediately. 2 months later, we made her cardiac catheterization and myocardial biopsy. Her LVG showed a small aneurysm of the apex, though her CAG was normal finding. The biopsy revealed moderate fibrosis and cellular infiltration including a few eosinophils. We thought that eosinophilic endocarditis had existed first, and secondary embolism continued led to the small infarction. The hypereosinophilia was spontaneously normalized 2 months after admission, but the patient complained of myalgia and sensory disturbance of extremities. The biopsy of quadriceps muscle could prove neither infiltration of eosinophils nor vasculitis. But we diagnosed mononeuritis multiplex due to hypereosinophilia. Judging from various symptoms and laboratory findings, this case was included to the hypereosinophilic syndrome. We also thought allergic granulomatosis and angitis as one of the differential diagnoses, but histologically vasculitis was not proved. In this case, eosinophilia was disappeared without using corticosteroids.(ABSTRACT TRUNCATED AT 250 WORDS)

[CD33-positive acute lymphoblastic leukemia with breast tumor and cavernous sinus tumor].

A 38-year-old woman was diagnosed as acute lymphoblastic leukemia (L2) in Oct. 1985. After VP and AdVEMP therapy, complete remission was obtained. In Oct. 1987, she noticed bilateral breast tumors and leukemic cell infiltrations were shown in a biopsy specimen of the breast tumor. Bone marrow was occupied with 94 percent blasts. The second complete remission was achieved by the AdVP therapy. In Nov. 1988, she developed double vision and photophobia. The examinations of CT and MRI showed cavernous sinus tumor, and 20 percent blasts were recognized in a bone marrow aspirate. The leukemic cells were negative for peroxidase, but were positive for both lymphoid and myeloid cell surface markers (CD2, CD5, CD7, CD33). The two color flowcytometry showed that CD5 and CD33 were simultaneously expressed on the leukemic cells.

[Pathomorphological study of maxillary invaginated incisor teeth].

Maxillary invaginated incisor teeth have been pathomorphologically investigated and some results have been obtained as follows: 1) The classification used in this study is as follows. That is, class I: Invagination is limited within the enamel. class II: Invagination extends toward the pulp chamber in a manner of pipe. class III: Invagination seen in class II expends at its end in the fashion of drop. class IV: Invagination occludes the whole coronal pulp chamber or extends toward the root apex beyond the cervical line. Frequent enamel-lined cavities of invaginated teeth were found to contain the contents (72.7%). Solid contents were found with considerable frequency in the cavities of the class I invaginated teeth of the author's classification (80.1%). On the other hand, the solid contents were rare in cavities of class III or IV (27.3%). In most cases, contents were found in limited areas of the cavity of class III or IV (72.7%). Any invagination corresponding to class II is not seen in this research. 2) The tissue composing the contents in the cavity of invaginated tooth was not clarified by the light microscopic observations of ground sections and film replicas. In class IV, poor hypomineralization was partly found in the inner enamel, and the dentin around the cavity was sclerotic. 3) As for the measurements of microhardess, the Vickers hardness number of contents in the cavities of invaginated teeth was approximately 1/4 of that of inner enamel. 4) The contents gave a higher radiolucency than the inner enamel by means of contact microradiography. 5) Electron microscopy revealed that the cavity contents of invaginated teeth was composed of poor inorganic elements resulting in a smooth aspect, and that, in most cases, a rod-having enamel was found in the surface layer of the contents and the rest of contents was composed of rodless enamel. An organic continuation of the tissue was observed between inner enamel and the contents. The results of electron microscopy, microhardness measurement, and contact microradiography, may indicate that the contents is composed of a hypomineralized enamel. 6) Caries incidence were found at a rate of 40.9% at cavity walls of invaginated teeth. Thus, it was suggested that cavity walls were susceptible of caries. In the contents of invaginated cavity, there was found primary caries appeared there.
