A case of prostatic carcinoma with skin metastases.

The 30th case of metastatic skin carcinoma of prostatic origin was presented. The cytosolic R1881 receptor was detected moderately in the metastatic nodule of the skin, however, the measurement of this receptor had a limited value for the purpose of treating the patient with antiandrogenic agents.

Alpha 1- and beta-adrenergic and muscarinic cholinergic receptors in guinea-pig nasal mucosa.

The alpha 1- and beta-adrenergic and muscarinic cholinergic receptors in guinea-pig nasal mucosa were measured for the first time by direct binding techniques using [3H]prazosin, [3H]dihydroalprenolol ([3H]DHA) and [3H]quinuclidinyl benzilate ([3H]QNB). The maximum binding capacities of [3H]prazosin, [3H]DHA and [3H]QNB in guinea-pig nasal mucosa were 20.1, 42.3, 159 fmol/mg protein, respectively. The dissociation constants of [3H]prazosin, [3H]DHA and [3H]QNB in guinea-pig nasal mucosa were 0.37, 0.77 and 1.5 nM, respectively. After the removal of the superior cervical ganglion in the guinea-pig, the number of alpha 1-adrenergic receptors was increased in the nasal mucosa while the number of beta-adrenergic and muscarinic receptors remained unchanged. Thus, alpha 1-adrenergic receptors are probably postsynaptic receptors in the target cells of sympathetic nerves, while beta receptors relate to the circulating catecholamines.

Loss of epidermal growth factor receptor on the renal neoplasm induced in vivo with Xenotropic pseudotype Kirsten murine sarcoma virus.

A characteristic feature of normal rat kidney (NRK) cells in their ability to bind epidermal growth factor (EGF) and loss of the ability to bind EGF in the renal neoplasm induced in vivo with C type RNA virus were demonstrated. Maximal binding capacity and apparent dissociation constant of NRK cells were 3.36 femtomoles of EGF bound per 10(6) cells and 2.85 X 10(-11) M, respectively. The binding of EGF to transformed cell in vivo with Xenotropic pseudotype Kirsten murine sarcoma virus was abolished.

Effect of anticancer drugs on renal neoplasm induced with xenotropic pseudotype kirsten murine sarcoma virus.

The sensitivity of a renal neoplasm induced in vivo with C type RNA virus to anticancer drugs was evaluated. The proliferation rate of the virus induced neoplasm cells depended on the concentrations of antimetabolites, alkylates and antibiotics, but not depends on that of alkaloids. An inhibitory effect of drugs on the virus induced neoplasm cells became evident in the following order, i.e., antimetabolites, alkylates, antibiotics and then alkaloids.

Radioimmunoassay of diethylstilbestrol in plasma of patients with prostatic carcinoma.

A method for the radioimmunoassay of diethylstilbestrol (DES) was established and it was used to measure the plasma concentration of DES in patients with prostatic carcinoma receiving intravenous drip infusion of 500 mg of diethylstilbestrol diphosphate (DES-DP). The sensitivity of this assay system was 40 pg per tube, with a recovery of 98%. The cross reactivity of the anti-DES antibody with estradiol, testosterone, and ethynylestradiol was less than 0.01% of DES, and that with DES-DP was 0.7%. In patients with prostatic carcinoma, plasma DES concentration at 6 and 24 hr after was declined to 10.2% and 4.4% of the level at the termination of intravenous administration of DES-DP, respectively; however, the concentration in patients with renal dysfunction was higher than that in ones with normal renal function.

Radioimmunological quantitation of urinary trypsin inhibitor.

Using monospecific antibody to human urinary trypsin inhibitor, we developed a highly specific and sensitive radioimmunoassay (RIA) for measuring human urinary trypsin inhibitor. No cross-reactivity of the antibody with protein standard serum, which contained albumin, alpha 1-antitrypsin, haptoglobin, alpha 2-macroglobulin, transferrin, IgG and IgA, was observed. The sensitivity of the system was 10 ng of trypsin inhibitor per assay tube, and 5-10 microliters of urine was sufficient to determine the concentration of trypsin inhibitor in urine. The amounts excreted in the urine of 10 healthy men and 10 healthy women were 4.83 +/- 2.46 (mean +/- SD) and 3.86 +/- 1.35 mg/day, respectively. The correlation between estimates by RIA and those by enzymic assay was r = 0.96 (p less than 0.005). The method proposed here can be used to determine the concentration of urinary trypsin inhibitor in a small amount of biological fluids and cells.

Accumulation of hydrocarbon in a patient with adrenoleukodystrophy.

The lipid composition of tissues from a patient with adrenoleukodystrophy was examined. Cholesterol esters of both brain and adrenal tissues contained increased proportions of fatty acids longer than C22 in agreement with previous reports. During this study, thin-layer chromatographic analysis of the neutral lipid fraction from the cerebral white matter and the adrenal gland revealed an unknown spot, which was identified as hydrocarbon of n-alkanes (CnH2n + 2, n = 16-33) by means of gas-liquid chromatography and gas chromatography/mass spectrometry. There may be a relationship between the accumulation of hydrocarbons and the change in fatty acid metabolism that is the fundamental disorder of ALD, as: (1) the fatty acids abnormally accumulated in cerebral white matter and adrenal gland coincided in chain length (i.e. C24-C26) with the main peaks of n-alkane in the same tissues, and (2) an abnormal accumulation of n-alkanes occurred in the diseased tissues where cholesterol esters with very long chain fatty acids were markedly increased.

Adrenergic and cholinergic receptors in human nasal mucosa in cases of nasal allergy.

We have demonstrated for the first time alpha-1 adrenergic, beta-adrenergic, and muscarinic cholinergic receptors in human nasal mucosa obtained from patients with nasal allergy or sinusitis. Using the radioligand 3H-prazosin, 3H-dihydroalprenolol (3H-DHA) and 3H-quinaclidinyl benzilate (3H-QNB), we determined the receptor densities (Bmax) and affinities (KD) of alpha-1, beta, and muscarinic receptors. There was an increased number of muscarinic receptors and a decreased number of alpha-1, beta receptors in patients with nasal allergy but not in those with sinusitis. Binding affinities (KD) of alpha-1, beta, and muscarinic receptors were unchanged in all patients with nasal allergy and sinusitis. Disorders of the autonomic nerve receptors appear to play a definite role in pathology of nasal allergy.

Change of total and prostatic serum acid phosphatase levels following hyperglycemic treatment in patients with prostatic carcinoma.

For the purpose of reevaluating diagnostic usefulness of total and prostatic serum acid phosphatase levels in patients with prostatic carcinoma, change of these levels was studied in 110 randomly selected patients, of which 35 were untreated prostatic carcinoma cases. Following hyperglycemic treatment, both levels in untreated carcinoma patients decreased significantly to their pretreatment levels and this test was a useful tool for diagnosis in the patients with prostatic carcinoma.
