Streptozotocin diabetes in rats after acclimation to cold environment.

To investigate the effects of acclimation to a cold environment on the alteration of glucose and fat metabolism in diabetes, the levels of blood glucose, triacylglycerol, nonesterified fatty acid, and immunoreactive insulin were measured before and on the 3rd and 14th days after streptozotocin treatment in cold-acclimated (4 degrees C) and warm-acclimated (24 degrees C; control) rats. The cold-acclimated rats showed no hyperglycemia on the 14th day after streptozotocin treatment, whereas the control diabetic rats maintained high blood glucose levels. Triacylglycerol levels did not increase in the cold-acclimated diabetic rats, whereas the control diabetic rats showed markedly high triacylglycerol levels. Immunoreactive insulin levels remained constantly low in both cold-acclimated and control diabetic rats. It was concluded that acclimation to cold changed some of the metabolic consequences of diabetes caused by streptozotocin in rats.

The pathogenesis of vascular changes in erythema nodosum-like lesions of Behçet's syndrome: an electron microscopic study.

The probable sequence of microvascular changes affecting endothelial cells in erythema nodosum-like lesions was examined by electron microscopy in 18 patients with typical Behçet's syndrome. Dark endothelial cells in the superficial and deep dermis exhibited degeneration with contraction changes. Consequently, degenerated endothelial cells showed nuclear and cytoplasmic condensation, vacuolization in the cytoplasm, and rich ribosomes. Moreover, blood vessels which were enveloped by cuffs of lymphocytes simultaneously exhibited both endothelial cell hypertrophy and endothelial cell necrosis. The implied associations between perivascular lymphocytic cuffs and blood vessels exhibiting endothelial cell alterations indicate that microvascular changes in erythema nodosum-like lesions may be closely associated with the delayed-type hypersensitivity reaction.

Electron microscopic observation on dark endothelial cells in erythema nodosum-like lesions of Behçet's disease with ultrastructural X-ray spectroanalysis.

We used electron microscopy and electron microscopic X-ray spectroanalysis to examine the biopsies taken from erythema nodosum-like lesions of 20 patients with Behçet's disease. Electron-dense degenerated endothelial cells were found in the microcirculatory beds of the dermis and subcutaneous fat. However, electron microscopic X-ray spectroanalysis failed to demonstrate the localization of either chlorine or base metals in those degenerated endothelial cells. Moreover, neither in ultrastructurally normal endothelial cells, nor in infiltrating macrophages of erythema nodosum-like lesions the positive deposit of chlorine or base metals could not be detected. The results of this analysis suggest that environmental factors do not play an etiologic role in this disease. Significance of the degeneration of endothelial cells is not clear present.

[Investigation of clinical effect of flomoxef].

Flomoxef (FMOX, 6315-S) is a new antibiotic of oxacephem group with a broad antibacterial spectrum. Seven patients were treated by intravenous instillation of 1 g of FMOX twice daily. Seven-day treatment with FMOX was ineffective against chronic bronchitis caused by Escherichia coli in a patient who had received an operation for esophageal cancer 6 years previously. Seven-day treatment with FMOX was effective against acute pneumonia caused by Staphylococcus aureus in a patient with lung cancer. Nine-day treatment with FMOX was effective against pulmonary suppuration caused by Streptococcus pneumoniae in a patient with lung cancer. Six-day and 7-day treatment with FMOX were excellently or moderately effective against suspected obstructive pneumonia in 2 patients with lung cancer. Five-day and 6-day treatment with FMOX were ineffective against fever of unknown origins in 2 patients with lung cancer. Overall, FMOX was effective in 4 of the 7 patients evaluated.

Electron microscopic observation on the epithelial-connective tissue junction of pemphigus vulgaris. A case report.

An electron microscopic observation on the erosive buccal mucosa of a buccal mucosa of a patient with typical pemphigus vulgaris was carried out. The ultrastructural features of the basal cells and the basal lamina in the advanced oral lesion varied from section to section of the biopsy specimen. The basal lamina showed various morphologic alterations indicating the presence of both destructive and regenerative processes in different sections. The filamentous bodies were also observed both in the basal cell layer and in the upper lamina propria.

Ultrastructure of lymphocyte-mediated fat-cell lysis in erythema nodosum-like lesions of Behçet's syndrome.

In this study, biopsy specimens from 18 patients with Behçet's syndrome were examined by electron microscopy with particular attention to the appearance of the lymphocyte-macrophage infiltrate into the interlobules of subcutaneous fat in erythema nodosum-like lesions. Electron microscopic evaluation revealed vacuolization changes of fat cells with detachment of their cell membrane from the basal lamina that permitted lymphocytes and macrophages to enter into this developed space. We believe that the detachment of fat cells from the basal lamina precedes invasion by lymphocytes which, in turn, attracts macrophages into the space. This eventually leads to fat-cell lysis accompanied by activation of macrophages, which causes further inflammation, completing the picture of panniculitis in the erythema nodosum-like lesions in Behçet's syndrome.

Histomorphometric study of trabecular channels in normal iliac bone.

Cancellous bone channels in the normal iliac crest have been studied histologically and by histomorphometry, and their biological role has been considered. Eighty percent of trabecular channels were typical osteons with the same structural and remodeling features as cortical osteons. The similarity of osteons in these two locations was corroborated by the comparability of morphometric features. The points of difference between osteons in the two types of bone were irregular configurations of trabecular osteons and marrow cells in the central canal in some. Since the number of trabecular osteons decreases with age, and since active trabecular resorption cavities were few in number, it is unlikely that additional osteons are formed in normal bone after the active modeling phase of bone growth. It is improbable that they make a significant contribution to bone remodeling since their number decreases with age, and since the available surface of trabecular channels for remodeling is extremely small. However, it is probable that they aid the intraosseous microcirculation and mineral exchange in thick trabecula and bifurcations, where they are mainly located. The demarcation curves at the 95% confidence interval, which suggest the normal range for numbers of channels, was calculated from the scatter diagram against bone area.

NAD+ glycohydrolase of the plasma membrane prepared from glial and neuronal cells.

NAD+ glycohydrolase (EC 3.2.2.5) activity was detected in the plasma membrane prepared from the primary culture of rat astrocytes. The enzyme has a broad optimum pH range. From the kinetic analysis, a Michaelis constant of 91.2 microM and a maximum velocity of 0.785 mumol/min/mg protein were obtained. ADPribose exhibited a competitive inhibition with respect to NAD. The inhibition by nicotinamide was shown to be of a non-competitive type. ATP and GTP were found to be competitive inhibitors. NAD+ glycohydrolase activity was not detected in the plasma membrane prepared from the primary culture of neuronal cells of chick embryos.
