Structural analysis and specific expression of microsomal cytochrome P-450(M-1) mRNA in male rat livers.

cDNA clones for the P-450(M-1) mRNA, which exhibits a male-specific expression in rat livers, were isolated by using synthetic oligonucleotides as the probes. Sequence analysis of the cDNAs showed that P-450(M-1) mRNA contains 1,853 nucleotides in addition to a poly(A) chain, and a single open reading frame of 1,500 nucleotides encodes a polypeptide of 500 amino acids with a Mr = 57,187. The predicted NH2-terminal sequence of 30 amino acids agrees well with that of the purified protein determined by Edman degradation, and the predicted primary structure included all the partial sequences of six internal peptides of P-450(M-1) obtained by the proteolytic digestion and a conserved amino acid sequence containing a putative heme-binding cysteine, proximate to the COOH terminus of the molecules. P-450(M-1) showed relatively high sequence similarity with P-450b (Fujii-Kuriyama, Y., Mizukami, Y., Kawajiri, K., Sogawa, K., and Muramatsu, M. (1982) Proc. Natl. Acad. Sci. U.S.A. 79, 2793-2797) (52% similarity), P-450-3b (Ozols, J., Heinemann, F. S., and Johnson, E. F. (1985) J. Biol. Chem. 260, 5427-5434) (64%), P-450-1 (Tukey, R. H., Okino, S., Barnes, H., Griffin, K. J., and Johnson, E. F. (1985) J. Biol. Chem. 260, 13347-13354) (74%), P-450PBc1 (Leighton, J. K., DeBrunner-Vossbrinck, B. A., and Kemper, B. (1984) Biochemistry 23, 4598-4603) (71%), while its sequence similarity with 3-methylcholanthrene-inducible P-450c and P-450d is rather low. Consequently, P-450(M-1) could be structurally classified into the phenobarbital-inducible type of P-450 gene family. RNA blot analysis using a synthetic oligonucleotide specific for P-450(M-1) revealed that P-450(M-1) mRNA was expressed exclusively in the livers of mature male rats in a sex-specific manner, but not in other tissues so far examined.

Differential abnormality in cell-cycle stage of peripheral B cells from patients with systemic lupus erythematosus.

In order to clarify the stage of abnormal activation of systemic lupus erythematosus (SLE) B cells, we investigated the cell-cycle phase of SLE B cells by flow-cytometric analysis. This study uses the simultaneous flow-cytometric analysis of cellular DNA content and incorporated bromodeoxyuridine, an analogue of thymidine, as indicators for DNA synthesis to detect activated B cells in peripheral blood of patients with SLE. In active SLE, the percentage of B cells increased in the S and G2M phase and decreased in the G0G1 phase as compared with normal control subjects. In inactive SLE, the percentage of B cells in the S phase also increased. Active SLE B cells did not progress through the cell cycle with the stimulation of anti-IgM plus PMA or SAC plus BSF, although normal B cells transit into S and G2M phase with such stimulation. These data suggest that SLE B cells are already activated and shifted to a matured state and that for this reason the B cells were poorly responsive to mitogen.

The Stickler syndrome: evidence for close linkage to the structural gene for type II collagen.

The Stickler syndrome is an autosomal dominant hereditary disorder of connective tissue with pleiotropic features including premature osteoarthropathy, mild spondyloepiphyseal dysplasia, vitreoretinal degeneration, and the Pierre-Robin sequence. Genetic linkage studies in two families with the Stickler syndrome have been performed using restriction fragment length polymorphisms associated with the structural gene for type II collagen, COL2A1. No recombinants between the Stickler phenotype and COL2A1 were observed. The total LOD score for linkage of the Stickler syndrome and COL2A1 at a recombination fraction (theta) of zero is 3.59. These findings suggest that, at least in some families, the mutation causing Stickler syndrome affects the structural locus for type II collagen.

Stage 5 retinopathy of prematurity. Prognostic value of morphologic findings.

Anterior and posterior segment findings were evaluated in 184 consecutive cases of stage 5 retinopathy of prematurity (ROP) (368 eyes). One hundred seventy patients had one or more examinations under anesthesia and 150 had open-sky vitrectomy (260 eyes) with or without additional surgery. Ocular findings were inspected and special attention was given to verify the morphology of the retinal detachment (RD) as predicted by ultrasonography. Immature iris and/or pupil, posteriorly or totally closed detachment funnels, vascularized epiretinal membranes, retinal folds, tractional retinoschisis, persistent hyaloid system, and subretinal blood and/or organization were the most significant indicators of bad prognosis. Evaluating stage 5 ROP eyes with these variables in mind is therefore essential.

With time changes of T-lymphocytes after Babesia infection in mice pretreated with Toxoplasma lysate antigen.

Adult female mice of the ICR/JCL strain were injected intraperitoneally with an emulsion of Toxoplasma lysate antigen (TLA; 100 micrograms per head) in light mineral oil (LMO) twice at 2 weeks' interval. Each mouse was inoculated intraperitoneally with 10(2) erythrocytes infected with Babesia rodhaini 2 weeks after the second injection of the emulsion. The rates of decrease in the erythrocyte count, hemoglobin concentration, and hematocrit value 10 days after inoculation, as compared with the counterparts determined 1 day before inoculation, were distinctly lower in these treated mice than in untreated control mice. In the course of Babesia inoculations, substantial reduction in size and weight of the thymus became higher in untreated control than in TLA-treated mice as the infection progressed. In untreated controls, the number of Thy-1 positive (Thy-1 (+) ) cells was about 91.9% of the normal value being 3.7 X 10(7) cells in the whole thymus 1 day before inoculation while those in TLA-treated mice were 6.5 X 10(7) and 0.9 X 10(7) cells 1 day before inoculation and 10 days after inoculation, respectively. The size and weight of the spleen increased in both groups by day 10 after inoculation. The total number of Thy-1 (+) cells in the spleen was 3.9 X 10(7) and 11.7 X 10(7) 1 day before and 10 days after inoculations, respectively in untreated and 12.9 X 10(7) and 15.6 X 10(7), respectively in TLA-treated mice. The number of Thy-1 (+) cells in the liver was 0.5 X 10(7) and 1.2 X 10(7) 1 day before and 10 days after inoculations, respectively in untreated and 0.3 X 10(7) and 3.4 X 10(7), respectively, showing a rate increase of 1033.3% in TLA-treated mice. The rate of increase in count of Thy-1 (+) cells contained in the peripheral blood was 61.4% in the former and 64.1% in the latter. In untreated mice, degeneration and destruction of lymphocytes in the thymus and spleen follicles and aggregation of lymphocytes, which were not found in Thy-1 (+) staining, around pericapillary ducts in the liver were seen histopathologically 10 days after inoculation. In TLA-treated mice, however, perivascular infiltration by Thy-1 (+) lymphocytes and activation of Kupffer cells in the liver and activation of spleen follicles were observed locally 10 days after inoculation.

Growth hormone-releasing hormone (GHRH)-secreting pancreatic tumor in a patient with multiple endocrine neoplasia type I.

A growth hormone-releasing hormone (GHRH)-secreting pancreatic tumor in a 36-year-old man, who had typical, familial, multiple endocrine neoplasia (MEN) type I with hyperparathyroidism and acromegaly, is described. The resected tumor, weighing 30 g, showed unusual histological features characterized by a meningioma-like arrangement of crescent-shaped cells and contained many cells that reacted with C-terminal specific antibody to GHRH-44 and a few somatostatin-immunoreactive (IR) and calcitonin-IR cells, but no GH-IR cells. A high concentration of IR-GHRH (9.8-13.2 micrograms/g wet weight tissue) with the full molecular size of GHRH-44 was detected in a tumor extract. Electron immunocytochemical study by the protein A-gold method revealed GHRH-IR granules with a mean diameter of 147 nm. After removal of the tumor, the plasma IR-GHRH level became normal (decreasing from 299 to 16.1 pg/ml) and the plasma IR-GH level also decreased, but still remained slightly high (decreasing from 42.4 to 9.6 ng/ml), suggesting the presence of an adenomatous lesion in the hypophysis.

Lower eyelid reconstruction with a conchal cartilage graft.

A conchal cartilage graft without skin or mucosa was used as a posterior lining for lower eyelid reconstruction in seven patients. The raw surface of the graft was smooth enough to cause no corneal irritation and was epithelialized from the surrounding mucosa about 3 to 4 weeks after the operation. The lower eyelid reconstructed in this method produced a stable lid margin.

Spindle cell carcinoma of the uterus.

A case of spindle cell carcinoma of the uterus in a 56-year-old woman is reported. Microscopically, it showed an intimate admixture of epithelial and sarcomatous elements. The epithelial nature, especially the squamous cell nature, in sarcomatous areas was indicated by immunoreactivity for keratin and ultrastructural characters, such as bundles of tonofilaments and some cell junctions, while the tumor cells in these areas were also vimentin-positive. Furthermore, ultrastructural study and immunostaining for actin demonstrated myofilaments in tumor cells in both tumor nests and sarcomatous areas. This may impart the high degree of invasiveness of spindle cell carcinoma.

In vitro and in vivo activity of NY-198, a new difluorinated quinolone.

NY-198 [1-ethyl-6,8-difluoro-1,4-dihydro-7-(3-methyl-1-piperazinyl)-4-oxo-3- quinolinecarboxylic acid hydrochloride] is a new difluorinated quinolone characterized by the presence of a C-methyl group at the 3 position of the piperazine moiety. It has a broad antibacterial spectrum. The in vitro antibacterial activity of NY-198 was almost the same as those of ofloxacin and norfloxacin, but far exceeded that of pipemidic acid. NY-198 was more active than norfloxacin against Pseudomonas maltophilia, Acinetobacter calcoaceticus, and anaerobic microorganisms. Cross resistance was not observed between NY-198 and various antibiotics including methicillin, gentamicin, and ampicillin. NY-198 had bactericidal activity at the MIC or slightly higher than the MIC. It showed excellent antibacterial activity against various systemic infections in mice. After oral administration, NY-198 was two times more active than or as active as ofloxacin and two to four times more active than norfloxacin.

Periosteal epithelioid hemangioendothelioma with leptomeric fibrils.

A case of recurrent periosteal epithelioid hemangioendothelioma of the right femur in a 66-year-old woman is reported. Microscopic examination showed epithelioid tumor cells with frequent intracytoplasmic vacuoles arranged in small nests or cords in hyalinized stroma. Immuno- and lectin-histochemical studies for factor VIII-related antigen and ulex europaeus I lectin indicated the endothelial nature of the tumor cells. Ultrastructurally, a few tumor cells contained leptomeric fibrils and crystalline filamentous aggregates in addition to showing certain features of endothelial cells. There seems to be no previous report of a vascular tumor containing leptomeric fibrils, which are often noted in normal striated muscle cells and their tumors. The bland appearance of the epithelioid tumor cells, poor vasoformative nature and prolonged clinical course differentiated this tumor from conventional angiosarcoma and so-called hemangioendothelioma of bone.
