[Pancreatic acinar cell carcinoma curatively resected by pancreaticoduodenectomy].

The incidence of acinar cell carcinoma among malignant pancreatic tumors is about 1%, and the prognosis is reported to be poor. We resected an acinar cell carcinoma that had arisen on the pancreatic head by pancreaticoduodenectomy in December 1983. This tumor occupied the pancreatic head, and it measured 10 X 9.5 X 9.5 cm. The microscopic findings showed that the tumor cell was well-differentiated and acinar formation was uniform. The growth of the tumor was expansive, and the lesion was clearly capsulated by fibrous tissue. There was no lymph node metastasis, so the curative operation was performed by the enbloc resection of the tumor. Zymogen granules in the tumor cells were confirmed by electron microscopy. Consequently, we concluded that the origin of the tumor was acinar cells.

[Effect of haloperidol decanoate on the dopaminergic system in the rat brain].

The effect of haloperidol decanoate and haloperidol on the dopaminergic system in the rat brain was studied. Each rat was treated with either once daily ip injection of haloperidol (5 mg/kg) for 4 weeks or a single sc injection of haloperidol decanoate followed by once daily ip injection of saline for 4 weeks. Three weeks after cessations of injections, the levels of dopamine (DA) and its metabolites, 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA), were determined in the striatum, limbic area and frontal cortex 4 hours after a single ip injection of haloperidol (1 mg/kg). The haloperidol decanoate-treated rats showed lower levels of DOPAC and/or HVA than the rats of the other two groups. The DA level was higher only in the limbic area of the rats treated with haloperidol decanoate than that of the rats treated with saline. There was no difference of the levels of DA and its metabolites between the saline and haloperidol-treated rats. These results indicate that the haloperidol decanoate-treated rats developed a tolerance to an acute effect of haloperidol on stimulation of DA turnover in the three areas examined, while the haloperidol-treated rats developed no tolerance under these experimental conditions. A single sc injection of haloperidol decanoate to rat is expected to produce a long lasting blocking of the central DAergic system for at least 7 weeks. Such a property of this drug appears favourable for the maintenance therapy of schizophrenia.

Six cases of central serous choroidopathy induced by systemic corticosteroid therapy.

We report 6 cases of central serous choroidopathy (including pigment epithelial detachment in one case) which appeared in the course of systemic corticosteroid administration conducted to cure concurrent general diseases, and in one patient with a steroid-releasing pituitary adenoma. The majority of cases arose within about one month following the administration of more than 200 mg of prednisolon. It is postulated that corticosteroids operate as a kind of stress intervening in the hypophysis-adrenal system, leading eventually to the development of central serous choroidopathy.

Complete nucleotide sequence of a methylcholanthrene-inducible cytochrome P-450 (P-450d) gene in the rat.

The rat cytochrome P-450d gene which is inducibly expressed by the administration of 3-methylcholanthrene (MC) has been cloned and analyzed for the complete nucleotide sequence. The gene is 6.9 kilobases long and is separated into 7 exons by 6 introns. The insertion sites of the introns in this gene are well-conserved as compared with those of another MC-inducible cytochrome P-450c gene, but are completely different from those of a phenobarbital-inducible cytochrome P-450e gene. The overall homologies in the coding nucleotide and deduced amino acid sequences were 75% and 68% between the two MC-inducible cytochrome P-450 genes, respectively. The similarity of the gene organization between cytochrome P-450d and P-450c as well as their homology in the deduced amino acid and the nucleotide sequences suggests that these two genes of MC-inducible cytochromes P-450 constitute a different subfamily than those of the phenobarbital-inducible one in the cytochrome P-450 gene family. In contrast with the notable sequence homology in the coding region of the two MC-inducible cytochromes P-450, all the introns and the 5'- and 3'-flanking regions of the two genes showed virtually no sequence homology between them except for several short DNA segments that are located in the promoter region and the first intron. The nucleotide sequences and the locations of these conserved short DNA segments in the two genes suggest that they may affect the expression of the genes. Middle repetitive sequence reported as ID or identifier sequence were found in and in the vicinity of the cytochrome P-450d gene.

Osteopathia striata with cranial sclerosis affecting three family members.

Skeletal surveys were performed on a 38-year-old Japanese mother, her son and daughter. The radiographs of both children showed characteristic features of osteopathia striata. However, in the mother, the skull, mandible, and lower extremities were homogeneously sclerotic with no evidence of a striated pattern of sclerosis in her skeleton. Additional features of striated sclerosis of the mandible in patients with osteopathia striata are discussed.

Antibody assay of rabbit sera for outer membrane vesicles of Haemophilus influenzae by enzyme-linked immunosorbent assay.

Enzyme-linked immunosorbent assay (ELISA) was applied to the detection of IgG and IgM antibodies against outer membrane vesicles (OMV) antigen of Haemophilus influenzae type b. In this ELISA system, IgG antibody titers were about 40 fold higher than those in indirect hemagglutination assay (IHA). The IgG antibody titers by this ELISA of rabbit sera obtained after immunization were comparable with those by radioimmunoassay (RIA) of the same sera. A significant correlation was established between these two assays (r = 0.973, P less than 0.001).

Microwave surgical treatment of diseases of prostate.

A new transurethral probe for microwave radiation of the prostate has been developed. As a preliminary experiment, sliced ham was radiated with microwaves using this probe in order to evaluate the extent of thermal effect. Using mongrel male dogs, microwave coagulation of the prostate was examined. These animal experiments showed marked destruction of the prostate gland. Furthermore, the safety of this method was confirmed on the basis of results from the experiments. Prostatic bladder neck obstruction also has been treated in 6 patients with benign prostatic hypertrophy or carcinoma of the prostate by this technique. There has been no mortality and also no complications. The results of this preliminary clinical trial have been excellent.

Effects of vitamin C on tumor induction by diethylnitrosamine in the respiratory tract of hamsters exposed to cigarette smoke.

A large dietary supplement (1%) of vitamin C was given to male Syrian hamsters exposed to cigarette smoke and receiving 12 weekly subcutaneous injections of diethylnitrosamine (DEN) to determine whether or not high doses of vitamin C can prevent the development of tumors by DEN in the respiratory tract. Treatment with DEN developed various types of tumors in the respiratory tract of hamsters and cigarette smoke exposure potentiated the tumor development in the nasal cavity, larynx and trachea. In comparison with the smoke-exposed hamsters treated with DEN, the vitamin C-supplemented hamsters showed significantly lower incidence of nasal cavity tumors and exhibited significantly earlier appearance of tracheal tumors. In addition, the laryngeal tumors also tended to develop earlier in the vitamin C-supplemented hamsters. The results indicate that high doses of vitamin C may inhibit tumor induction by DEN and exposure to cigarette smoke in the nasal cavity, but the development of laryngeal and tracheal tumors appeared to be accelerated by vitamin C supplement.

A possible explanation for a greater discrepant suppression by adenosine and N6-cyclohexyl adenosine of the guinea-pig ileal contraction.

Adenosine and N6-cyclohexyl adenosine (CHA) reduced dose-dependently a twitch contraction of guinea-pig ileum and their IC50 values were 1.0 X 10(-5) and 1.1 X 10(-8) M, respectively. By exposure to adenosine uptake and deaminase inhibitors, dipyridamole and erythro-9-(2-hydroxy-3-nonyl) adenine (EHNA), the adenosine-induced inhibition was enhanced, whereas the CHA-induced one was almost unchanged. Therefore, outstanding potency difference of the suppression by adenosine and CHA was minimized by concurrent addition of both the inhibitors. Uptakes of [3H]CHA into the guinea-pig ileum and taenia-coli were considerably lower than those of [3H]adenosine and were insensitive to dipyridamole. In the rat connective tissue segment, uptake of [3H]adenosine was smaller than that in the guinea-pig taenia-coli and ileal segments (especially, in the latter), and was virtually unaffected by 10(-5) M dipyridamole. These findings suggest that a greater discrepancy of the suppression by adenosine and CHA of the ileal twitch may substantially result from their different uptake and deamination in the synaptic region rather than their dissimilar intrinsic activity for the purinoceptor.

Expression of the gag gene of human T-cell leukemia virus type I in Escherichia coli and its diagnostic use.

An expression plasmid, pHY202, was constructed which directs the synthesis of a fusion protein encoded by the gag sequence of human T-cell leukemia virus type I (HTLV-I) inserted into the lacZ' gene. Escherichia coli cells harboring pHY202 produced the 43-kDal LacZ'-Gag fusion protein with a yield of approx. 0.3% of total soluble proteins. The fusion protein is specifically recognized by monoclonal antibodies against the Gag proteins p19 and p24, and could be applicable for the diagnosis of HTLV-I infection, because almost all sera from HTLV-I carriers gave a positive response in the enzyme-linked immunosorbent assay (ELISA) employing the LacZ'-Gag hybrid protein purified by immunoaffinity column chromatography.

Gastric acid secretory function in preterm infants.

To establish normal values for gastric secretory function in preterm infants, we studied 34 healthy preterm infants once a week during hospitalization. Basal acid output, pentagastrin-stimulated acid output, fasting serum gastrin, and fasting serum pancreatic polypeptide were measured during each study. Basal acid output at 1 week of age was 12 mumol/kg/hr, increasing over the first 4 weeks to 30 mumol/kg/hr. Administration of pentagastrin 6 micrograms/kg subcutaneously increased acid output in all age groups. Pentagastrin-stimulated acid output at 1 week was 21 mumol/kg/hr, increasing over the first 4 weeks to 44 mumol/kg/hr. Acid secretion did not change significantly over the next 4 to 6 weeks. Fasting serum gastrin concentration was stable over the first 6 weeks of life, but doubled during the end of the second month. Pancreatic polypeptide was found at low levels throughout the study. These studies confirm that the majority of healthy preterm infants secrete acid in quantity sufficient to maintain the gastric pH less than or equal to 4, providing a barrier to bacteria and protein antigens.

Role of the sympathetic nerve in bladder and urethral sphincter function during the micturition cycle in the dog evaluated by pressure flow EMG study.

The role of the sympathetic nerve on bladder and urethral sphincter function during the whole micturition cycle, including the collecting and emptying phases, was evaluated on 10 decerebrated dogs by pressure flow EMG study. A series of experiments was performed before and after hypogastric nerve transection. In the control condition and after hypogastric nerve transection, reflex micturition with bladder contraction and spasmodic rhythmic sphincter contractions occurred. Urodynamic parameters of the micturition cycle were statistically compared between control and nerve transection experiments. Threshold volume, threshold pressure and opening pressure showed a small but significant change after nerve transection. A decrease in voided volume seems to be secondarily accompanied with a decrease in threshold volume. It seems that adrenergic nerves play a certain role in bladder function during the collecting phase of the micturition cycle.
