Evaluation of recovery from local muscle fatigue by voluntary test contractions.

The applicability of surface EMG frequency analysis as a means of evaluating recovery from local muscle fatigue was studied experimentally, fatigue being given as the proportion of the integrated value of frequency components below 60 Hz to that of all components. When the maximum voluntary contractions of elbow flexor were done repeatedly in the recovery period after dynamic or static contractions, the slow wave proportion recovered to the initial value within about 6 min after work. The muscle force of test contraction after 20 min or more was, however, still significantly lower than its initial value. Recovery of integrated EMG value was faster than that of muscle force but slower than that of slow wave components. The effect of blood flow occlusion on the recovery was then tested using different levels of load. When the occlusion was made for 4 min immediately following the sustained contractions, the slow wave component again increased after a phase of decrease but did not reach the final level attained at the end of work, implying that the observed changes in EMG frequency were somewhat related to local muscular fatigue of a peripheral nature. The recovery from local muscular fatigue may thus be evaluated by intermittent brief test contractions after muscular work.

[Studies on the safety of KW-1062 (fifth report). A comparative study on the renal toxicity of KW-1062 and gentamicin in rats by ligh and electron microscopies (author's transl)].

Effects of KW-1062, a new aminoglycoside antibiotic, and gentamicin on the kidney were compared. KW-1062 (5, 10, 20, 40 and 80 mg/kg) and gentamicin (5, 10, 20 and 40 mg/kg) were intramuscularly administered to male Wistar strain rats for 30 days. On the 5th, 15th and 30th day, renal disturbances were examined by light microscopy. On the 30th day, electron microscopy was also carried out. Light microscopic examination revealed that degeneration of the renal tubular epithelium and the round cell infiltration were seen on the 5th day in the group received gentamicin 5 mg/kg. On the 15th and 30th day, these changes were dose-dependent. In the group given KW-1062 40 mg/kg, degeneration of the renal tubular epithelium and the round cell infiltration were recognized on the 15th day. Electron microscopic examination revealed dense body containing myelin-like structures in the epithelial cell of the portio principal in proportion to dose in the group received gentamicin 5 mg/kg. At gentamicin 40 mg/kg, myelin-like structures were collected in the renal tubular cavity and obstructed it. With KW-1062 20 mg/kg or more, similar findings to those of gentamicin 5 mg/kg were obtained, however, severe disturbance such as the obstruction of the renal tubular cavity was not recognized.

[Animal test for evaluation of ototoxicity and safety of KW-1062 (author's transl)].

KW-1062, a new aminoglycoside antibiotic, was tested for the evaluation of audiotoxicity in administration at higher dosage and compared with the audiotoxicity of gentamicin. KW-1062 was given at dosage of 50, 100 and 150 mg/kg intramuscularly for 4 weeks to each group consisting of 10 healthy guinea pigs, respectively. Gentamicin was administered at dosage of 25, 35, 50 and 100 mg/kg in the same way as in KW-1062 to the guinea pigs. In the present study the differential frequency pinna reflex test in the frequency range from 20 kHz to 0.5 kHz was performed before and during administration of the drugs, to evaluate the audiotoxicity. Comparison of number of animal with pinna reflex loss and extent of frequency range of pinna reflex loss revealed that KW-1062 at dose of 150 mg/kg is less audiotoxic than gentamicin at 50 mg/kg and 100 mg/kg, and approximately equivalent in audiotoxicity to gentamicin at dose of 35 mg/kg.

Comparison of efficacy of amoxapine and imipramine in a multi-clinic double-blind study using the WHO schedule for a standard assessment of patients with depressive disorders.

A multi-clinic double-blind controlled study on amoxapine in comparison with imipramine, using the WHO Schedule for a Standard Assessment of Patients with Depressive Disorders, was performed and the data were analyzed with 111 patients. The assessment of severity of illness and overall improvement indicated clearly the superiority of the antidepressive effect of amoxapine to that of imipramine. The onset of antidepressive effect of amoxapine was clearly more rapid than that of imipramine, and in more than half of the patients in the amoxapine group the improvement was seen within four days following the drug administration. Amoxapine was superior to imipramine in terms of safety and usefulness. The side-effects due to amoxapine appeared less frequently and were less serious than with imipramine. The difference between amoxapine and imipramine was especially remarkable for hypotensive effect. The antidepressive effect of amoxaphine was superior to that of imipramine for almost all symptoms and signs. Amoxapine displayed an especially remarkable effect on psychomotor retardation, depressive feeling, anxiety and tension, somatic complaints and sleep disturbance.

Clinical significance of mitochondrial glutamic-oxaloacetic transaminase in serum of patients with liver disease.

Serum mitochondrial glutamic-oxaloacetic transaminase activity was determined in 83 patients with various liver diseases and 10 healthy adults. 1) The average of mitochondrial glutamic-oxaloacetic transaminase value was 1.2 mU in healthy adults, 8.3 mU in patients with acute hepatitis, 13.7 mU in patients with post-transfusion hepatitis, 5.0 mU in patients with persistent hepatitis, 4.5 mU in patients with chronic inactive hepatitis, 9.6 mU in patients with chronic active hepatitis, 5.6 mU in liver cirrhosis, and 295 mU in a patient with fulminant hepatitis. 2) While one patient with acute hepatitis showed the highest value in the group of 29 mU, one patient with fulminant hepatitis showed an extremely high value of 295 mU, revealing an obvious difference between them. 3) One patient with fresh myocardial infarction also showed an extremely high value of 110 mU.
