Transfer ribonucleic acid guanine transglycosylase isolated from rat liver.

Transfer ribonucleic acid (tRNA) guanine transglycosylase (guanine insertion enzyme) was isolated from rat liver and extensively purified. The enzyme catalyzes an exchange of queuine (the base of queuosine, Q) as well as its precursors and guanine for guanine originally located in the first position of the anticodon of "undermodified" tRNATyr, tRNAHis, tRNAAsn, and tRNAAsp from an Escherichia coli mutant or rat ascites hepatoma cells. This is in contrast to the previous observation that E. coli tRNA-guanine transglycosylase catalyzes the exchange of queuine precursors, such as 7-(aminoethyl)-7-deazaguanine and 7-cyano-7-deazaguanine, but not of queuine itself [Okada, N., Noguchi, S. Kasai, H., Shindo-Okada, N., Ohgi, T., Goto, T., & Nishimura, S. (1979) J. Biol. Chem. 254, 3067-3073]. The Km value for queuine of the rat liver enzyme is 9.2 X 10(-7) M, much lower than the values for the bases of queuosine precursors or guanine. Thus, the actual substrate for tRNA-guanine transglycosylase in queuosine biosynthesis in vivo in rat liver may not be 7-(aminomethyl)-7-deazaguanine, which is thought to be an actual substrate guanine, the E. coli system. Queuine or some queuine derivative may be the actual substrate for the tRNA-guanine transglycosylase reaction in the biosynthesis of Q in tRNA of mammalian cells. 6-Thioguanine and 8-azaguanine are also found to be good substrates.

Ureteral ectopia in vestibule without urinary incontinence.

Two cases of ureteral ectopia opening in the vestibule without urinary incontinence are reported. Urinary continence was probably due to the incorporation of the ectopic ureteral ends in the wall of the urethral sphincter.

Implication of endogenous prostaglandin system in the antihypertensive effect of captopril, SQ 14225, in low renin hypertension.

The role of endogenous prostaglandins in the antihypertensive mechanism of orally active angiotensin I-converting enzyme inhibitor in low renin hypertension was investigated. In the SQ 14225-responders with low renin hypertension, blood pressure was elevated to control level and antihypertensive effect of SQ 14225 was attenuated after the inhibition of endogenous prostaglandin synthesis with indomethacin, while no significant change in blood pressure was found in the SQ 14225-responders with normal renin hypertension. Urinary prostaglandin E excretion was not significantly increased after the SQ 14225 administration but significantly decreased after the administration of indomethacin. There was no significant difference in urinary prostaglandin E excretion between normal renin hypertension and low renin hypertension. These results suggest that SQ 14225 may potentiate extrarenal vasodilating prostaglandin system, probably vascular prostaglandin system, contributing to the antihypertensive mechanism in SQ 14225-responders with low renin hypertension. The augmented renin release following SQ 14225 administration was inhibited by indomethacin, suggesting that endogenous prostaglandin system may contribute to the negative short feedback mechanism of renin release.

Indomethacin inhibits the antihypertensive effect of captopril, SQ 14225, in low renin hypertension.

The antihypertensive mechanism of orally active angiotensin I-converting enzyme inhibitor, SQ 14225 (SQ), in low renin hypertension was investigated. In the SQ-responder with low renin hypertension, the hypotensive effect of SQ was abolished after the inhibition of endogenous prostaglandin (PG) synthesis with indomethacin, while no significant change in blood pressure was found in the SQ-responder with normal renin hypertension. These results suggest that SQ may potentiate the vasodilating PG system, contributing to the antihypertensive mechanism in SQ responders with low renin hypertension.

Active and inactive renin after SQ 14225 (Captopril) administration.

The changes of active and inactive renin in plasma after the oral administration of 25 mg or 50 mg of SQ 14225 (Captopril) were studied in 29 hypertensive patients. Inactive renin was calculated as plasma renin activity (PRA) after cold storage minus PRA before cold storage. The patients were divided into 2 groups, responders and non-responders, according to the response of active renin to Captopril. In 9 responders, the active renin increased markedly while the inactive renin decreased. On the other hand, in 20 non-responders, both renin activities increased only slightly. These results suggest that inactive renin may be converted in vivo to active renin by Captopril.

Experimental studies on TDI dermatitis in mice.

Allergic contact dermatitis occurred with 2, 4-toluene diisocyanate (TDI) in BALB/c mice. The results were obtained by the measurement of the ear thickness after challenged to the ear with 1% TDI in olive oil in mice previously sensitized with 5% TDI. The ear swelling rate at 48 hr postchallenge in sensitized mice was more than twice of that in unsensitized mice. In BALB/c-nu/nu (nude) mice, delayed type hypersensitivity could not be induced even when a 30% TDI solution was applied. These results suggest that T cells may play an essential role in contact sensitivity to TDI in BALB/c mice.

FR-900130, a novel amino acid antibiotic. I. Discovery, taxonomy, isolation, and properties.

A new antibiotic, designated FR-900130, has been discovered in the culture filtrate of Streptomyces. The producing organism has been identified as Streptomyces catenulae. The antibiotic was purified by adsorption onto Duolite C-20, passage through Amberlite IRC-50, final purification and desalting on Sephadex G-25. It shows antimicrobial activity against Gram-positive bacteria and synergy with D-cycloserine. The antibiotic is very labile in alkaline solution above pH 8.0 and decomposed during lyophilization or solvent precipitation. It was converted to an acetyl derivative and the biological properties of the derivative were examined.

Studies on new phosphonic acid antibiotics. I. FR-900098, isolation and characterization.

A strain of Streptomyces, isolated from a soil sample and identified as Streptomyces rubellomurinus sp. nov., has been found to produce FR-900098, an interesting new antibiotic containing phosphorus in its molecule. The antibiotic, obtained as colorless crystals, was shown to inhibit a wide variety of Gram-negative bacteria including Pseudomonas, Proteus, and Escherichia coli. Its antibacterial action involves interference with bacterial cell wall synthesis as evidenced by the fact that it causes spheroplast formation by susceptible cells.

Studies on new phosphonic acid antibiotics. III. Isolation and characterization of FR-31564, FR-32863 and FR-33289.

Three phosphonic acid antibiotics were found to be produced in the fermentation broths of Streptomyces. FR-31564 and FR-32863 were produced by Streptomyces lavendulae. FR-33289 was produced by a strain of Streptomyces, identified as Streptomyces rubellomurinus subsp. indigoferus. They are distinct from, but resemble FR-900098 which was reported in our preceding paper, in their chemical and biological characteristics.

FR-900148, a new antibiotic. I. Taxonomy, fermentation, isolation and characterization.

A strain of Streptomyces, isolated from a soil sample and identified as Streptomyces xanthocidicus, has been found to produce FR-900148, a new antibiotic containing chlorine in its molecule. The antibiotic inhibits both Gram-positive and negative bacteria. However, it is not effective against wild type of Pseudomonas aerguinosa. Its antibacterial action is considered to result from cell wall synthesis inhibition since it causes spheroplast formation from susceptible cells.
