The stainability of proliferating cells with monoclonal antibodies to both bromodeoxyuridine (BrdU) and Ki-67 were compared in 22 malignant lymphomas and six related conditions. A significant correlation was observed (r = 0.78) between the positivities of proliferating cells determined using these two monoclonal antibodies. The growth fractions determined with the antibodies were usually related to the histologic classification, but labelling index was more varied in diffuse large cell type lymphoma. BrdU labelling was well correlated with the Working Formulation, and more useful as a prognostic indicator. On the other hand, Ki-67 was useful for broad recognition of cell proliferative ability.
One day after surgery groups of pinealectomized and sham-pinealectomized animals were placed in continuous darkness, and killed 0, 20, 40, 60, 80 and 100 days later. The data revealed that the sham-pinealectomized group exposed to constant darkness differs from pinealectomized group by showing lower dihydrotestosterone (DHT) concentrations, bw, accessory sex organ wt and levator ani muscle wt. No statistically significant differences were found in plasma LH, FSH and testosterone concentrations or pituitary or testes wt. The results indicated that the accessory sex organ wt were not correlated with the plasma gonadotropin and testosterone levels but they were with DHT. The data are consistent with the idea that some pineal principle acts directly on accessory sex organs in the rat.
The effect of pinealectomy on the circadian periodicity in plasma gonadotropins (LH and FSH), testosterone (T) and dihydrotestosterone (DHT) was investigated in adult male rats maintained under conditions of 12 h light/12 h dark (light on 06:00-18:00 h). Animals were killed at 3 hour intervals over 24 hours by decapitation. Plasma concentrations of LH, T and DTH (but not FSH) were found to exhibit significant circadian variations as determined by analysis of variance. No distinct LH or T nadir was recorded but peaks were observed at 09:00 h. Peak DHT values were recorded near the middle of the dark period. Pinealectomy resulted in an altered daily LH and T rhythmicity, and also in an abolished DHT rhythmicity. The fact that there was no significant increase in mean gonadotropin levels at 50 days after pinealectomy suggests that the pineal regulates the timing of gonadotropin secretion rather than the quantity. Since pinealectomy was found to abolish the circadian rhythm of DHT but not of T, we deduced that the pineal may also be responsible for the rhythmic conversion of T to DHT.
Adenosquamous carcinoma (ASqC) of the pancreas is a rare tumor. We analyzed the radiologic findings and clinical manifestations in two such cases. In both cases (a 51-year-old woman and a 67-year-old man), the portal system was selectively and largely invaded, the superior mesenteric vein in one and the splenic vein in the other, without arterial invasion. Thus, peripancreatic vessels should be carefully observed in patients with ASqC of the pancreas. One case showed an unusual mode of spread. Only a huge metastatic lesion was initially detected, leading to the misdiagnosis of primary malignant mesenteric tumor. Such an unusual growth pattern is also worth noting.
We present two cases with tumor seeding along the needle tract occurring after a large-core needle liver tumor biopsy performed at other hospitals. Color Doppler sonography showed the hypervascular nature of the lesion and increased diagnostic confidence.
Groups of three or four male and female beagle dogs were maintained for 6 months on diets containing BHA at concentrations of 0 (control group), 0.25, 0.5 and 1.0% (w/w). The average daily intake of BHA was about 60 mg/kg body weight in the 0.25% group, 110 mg/kg in the 0.5% group and 220 mg/kg in the 1.0% group. All animals survived the entire experimental period without showing signs of toxicity other than a dose-related retardation of growth. Serum biochemical examinations conducted at 1, 3 and 6 months revealed a slight decrease in albumin concentration and an elevation of alkaline phosphatase and leucine aminopeptidase activity in the higher dose groups. Histopathological and histometrical examination showed no evidence of mucosal alteration in the stomach, oesophagus or duodenum attributable to the administration of BHA. The mitotic index in the squamous epithelium of the distal oesophagus was comparable in the control and BHA-treated groups. Liver weights were increased in BHA-treated groups, but there were no related histopathological changes. From these data it is concluded that feeding of BHA-supplemented diets at palatable concentrations for 6 months has no pathological effects on the stomach, oesophagus, duodenum or liver of beagle dogs.
We investigated the surface markers as well as the expression of beta 2-integrin (LFA-1 beta/CD 18), in T cells migrating through human umbilical vein endothelial cells (EC) in patients with HTLV-I-associated myelopathy (HAM). No significant differences were found in the percentages of both HLA-DR+ T cells and total CD4+ cells and in the expression of beta 2-integrin between the EC-transmigrated and the EC adherent T cells. However, the percentages of HLA-DR+CD4+ cells in the transmigrated cells were significantly higher than those in the adherent cells. These results suggest that activated or HLA-DR+CD4+ T cells play an important role as the first trigger in the immunopathogenesis of HAM.
Adult rabbits were inoculated with liver homogenate from a rabbit that died in a Japanese outbreak of rabbit haemorrhagic disease (RHD). All experimentally infected rabbits died with typical clinical, gross and histological findings of RHD. Distribution of RHD virus in tissues of the infected rabbits was studied by non-isotopic in-situ hybridization. Both viral plus- and minus-strand RNAs were detected within the cytoplasm of hepatocytes, Kupffer cells and splenic and alveolar macrophages, mainly in morphologically intact cells. Strand-specific reverse transcriptase-polymerase chain reaction also demonstrated viral minus-strand RNA as well as plus-strand RNA in the liver, lung and spleen of infected rabbits. These results suggest that viral replication occurs not only in hepatocytes but also in macrophages. The infected macrophages may contribute to viral dissemination in RHD.
