Parvalbumin in rat cerebrum, cerebellum and retina during postnatal development.

Radioimmunoassay and immunohistochemical studies were performed on the occurrence and distribution of parvalbumin-like immunoreactivity (PA-LI) in developing rat cerebrum, cerebellum and retina. No PA-LI was detected in the nervous tissues of the newborn animals. In the cerebrum, the PA-LI appeared in non-pyramidal neurons at the 2nd postnatal week and increased linearly until the 8th week. In the cerebellum, a rapid increase in the PA-LI took place at the 2nd week, with an enrichment of the antigen to Purkinje neurons. In the retina, amacrine cells contained PA-LI, the levels of which increased from the 2nd to 4th week. Regulation of intracellular Ca2+ concentration may be one of the important factors for the maturation of the central nervous system.

Partial N-terminal sequence analysis of human class II molecules expressing the DQw3 determinant.

HLA-DQ molecules were isolated from DRw9-homozygous and DR4-homozygous cell lines by using a monoclonal antibody HU-18, which recognizes class II molecules carrying the conventional DQw3 determinant. The partial N-terminal sequence analysis of the DQw3 molecules revealed that they have sequences homologous to those of murine I-A molecules. Within the limits of our sequence analysis, the DQw3 molecules from the two cell lines are identical to each other in both the alpha and beta chains. The DQ alpha as well as DQ beta chains were found to have amino acid substitutions when compared to other I-A-like molecules whose sequences have been reported. These differences may contribute to the DQw supertypic specificity. The polymorphic nature of DQ molecules is in marked contrast to that of DR molecules where DR alpha chains are highly conserved while DR beta chains have easily detectable amino acid substitutions.

Parvalbumin exists in rat endocrine glands.

Simple and rapid purification procedures for parvalbumin, one of the Ca2+-binding proteins (extracted from rat skeletal muscle), were developed, and its antiserum was produced in rabbits to measure the parvalbumin content of various rat tissues by RIA. The heat treatment, ammonium sulfate fractionation, and trichloroacetic acid precipitation of soluble fraction from rat skeletal muscle followed by single diethylaminoethyl Sephadex A-50 column chromatography yielded a pure 120 mg protein from 150 g skeletal muscle. Amino acid analysis, together with electrophoretic mobility, indicated that the protein was identical to parvalbumin. The antisera to this rat skeletal muscle parvalbumin (raised in rabbits) did not cross-react with calmodulin or S-100 proteins. The RIA for parvalbumin using this antisera and [125I]parvalbumin revealed that skeletal muscle and brain contained high levels of the antigen; the values of which were 69,486 +/- 4,933.1 and 881 +/- 165.6 ng/mg protein, respectively. However, the parvalbumin antigen in the heart, lung, liver, and spleen could not be detected. On the other hand, the contents of the antigen in the endocrine glands (in nanograms per mg protein) were as follows: pituitary (125 +/- 46.6), thyroid (108 +/- 50.0), adrenal (341 +/- 64.3), testes (227 +/- 37.2), and ovaries (218 +/- 10.1). All of these values were comparable to levels of antigen found in the brain sample. These results suggest an important role for parvalbumin in endocrine glands.

Changes in DOPAC and 5-HIAA after acute cerebral hemorrhage induced by hypertonic glucose solution (i.p.)--in vivo voltammetry study.

Intraperitoneal injection of hypertonic glucose solution induced intracranial hemorrhage in rats. Simultaneously determined plasma norepinephrine exponentially increased followed by death. Using in vivo voltammetry, the present study was undertaken to estimate the relationship between neurochemical parameters and intracranial bleedings induced by hypertonic glucose injection. When placed in a solution of dihydroxyphenylacetic acid (DOPAC) and 5-hydroxyindole acetic acid (5-HIAA), electrically pretreated carbon fiber electrodes produced a 3 distinct peaks. Peak 1 and 2 refer to the extracellular fraction of ascorbic acid and DOPAC. Peak 3 refers to that of 5-HIAA. The peaks of the voltammograms obtained in vivo from different areas of the brain are similar to those observed in vitro using DOPAC and 5-HIAA solutions. Electrodes implanted in the ventricular CSF showed that DOPAC increased immediately after glucose injection, while 5-HIAA did not change significantly. When the electrode was inserted into the putamen, 5-HIAA exponentially increased followed by death, whereas DOPAC showed only a small change. It was presumed that the stroke-induced major oxidizable compounds are different in the lateral ventricle and the putamen. These results suggest that during cerebral bleeding, plasma norepinephrine increases accompanied with a potentiation of the central DOPAC and 5-HIAA system in rats.

Detection of circulating adenocarcinoma-associated antigen in the sera of cancer patients with a monoclonal antibody.

The antigen (YH206 antigen) corresponding to the monoclonal antibody (MoAb) YH206 (IgM), which was produced against a lung adenocarcinoma cell line A549, was found to be an extremely high-molecular-weight protein of over 330,000 daltons by means of SDS-PAGE and Western blot analysis. The immunohistological distribution of the antigenic determinant recognized by MoAb YH206 was found to be limited to adenocarcinoma tissues. It was shown by reversed passive hemagglutination assay (RPHA) that the antigen could be detected not only in the spent medium from human lung cancer cell line A549 but also in the sera from cancer patients. Only 3 out of 30 (10%) healthy donors and 4 of 31 (12.9%) patients with benign diseases had serum antigen levels of more than 1/64 dilution. In contrast, 42 of 87 (48.3%) patients with lung cancer and 29 of 50 (58.0%) patients with cancers of the digestive organs had elevated levels of the antigen. As regards the relation between antigen levels and clinical stages of lung cancer, values of more than 1/128 dilution were detected only in patients with stage III or IV disease.

[Intra-arterial infusion chemotherapy using a subcutaneously implanted silicone reservoir--with reference to the chemotherapy protocol].

Sixty patients with advanced cancers were treated with intraarterial infusion chemotherapy using a subcutaneously implanted silicone reservoir. Forty-five patients received a low dose-intermittent intraarterial infusion chemotherapy employing either combination MFC (MMC 4 mg, 5-FU 500 mg, Ara-C 40 mg) at intervals of two weeks or MMC 4 mg weekly. The results were as follows. (1) Among 30 cases, where objective evaluation of the initial response was possible, partial responses (PR) were observed in 15 cases (50%). (2) No severe complication was observed using this protocol. Only one case out of the 45 needed to discontinue the medication because of bone marrow suppression. (3) Among 28 cases, where follow-up data were available, the average hospital-free interval was 207 days. The average ratio of hospital-free interval to survival period was 56.4%. The results indicate that our method is beneficial as a maintenance chemotherapy for patients with advanced cancers, because it is effective, safer and improves the quality of life.

[Intra-arterial infusion chemotherapy using a subcutaneously implanted silicone reservoir--with special reference to the method].

Sixty patients with advanced cancers were treated with intraarterial infusion chemotherapy using a subcutaneously implanted silicone reservoir. Forty-nine patients were catheterized from a branch of the left subclavian artery. The results were as follows. (1) In spite of some technical difficulties, catheterization from a branch of the left subclavian artery was minimally invasive, and catheter trouble using this route was less frequent than that occurring when the profound femoral arterial route was employed. (2) Implantation of the catheter end using a silicone reservoir liberated the patients from restrictions in their daily lives, and with this method, intraarterial infusion chemotherapy could be carried out easily when required without hospitalization. We conclude that this procedure is a beneficial method of intraarterial infusion chemotherapy for patients with advanced cancers.

[Intravesical Bacillus Calmette-Guerin for treatment of bladder tumor].

The patients with bladder tumor (11 tumor) were given intravesical Bacillus Calmette-Guerin (BCG) therapy. The concentration of BCG solution varied from 80 to 240 mg in 40 ml of normal saline. The intravesical instillation therapy with this solution was repeated 6 times at a week interval. Seven of the tumors showed excellent response and had completely disappeared upon endoscopic examination. All of the highly responded tumors were small papillary lesions of low grade and low stage of malignancy. Regarding side effects, irritable bladder occurred in 70%, elevation of body temperature of over 37 degrees C in 40% and gross hematuria in 30% of the patients. Among them, one patient was treated with INH (0.3 g/day) for 2 weeks with satisfactory remission of the symptoms. Though the number of cases and follow up period are not satisfactory enough, topical therapy with BCG can be said to be an effective therapeutic measure against early stage urotherial tumors.

[A clinical study of renal cell carcinoma].

Eighty two cases of renal cell carcinoma, treated at our Department between July, 1971 and May, 1984, are reviewed. The highest incidence of this disease was seen in the 5th decade and the average age of the patients was 58.8 years. Male patients predominated over the female patients, the ratio being 1.6: 1. According to Robson's classification, thirty eight cases were in stage I, five cases in stage II, nine cases in stage III and thirty cases in stage IV. The overall survival rate at one, three and five years was 77, 58 and 48%, respectively. The most common symptom was macroscopic hematuria, followed by asymptomatic, lumbago and palpable mass. The site of distant metastases was most frequently in the lung, followed by the bone, liver and lymph node. There were no remarkable differences in the prognosis of stage I patients, between the group treated with preoperative renal arterial embolization and non-treated group. No remarkable difference was seen between the survival rate of the stage IV patients treated by nephrectomy and these not treated by nephrectomy. Renal arterial embolization was an effective therapeutic measure for patients with non-resectable stage IV renal cell carcinoma (p = 0.0647, compared with non-treated group).

Gastrointestinal absorption of inorganic mercuric compounds in vivo and in situ.

The absorption of inorganic mercury in rats was studied by using ligated gastrointestinal segments and perfusion of small intestine. Poorly soluble mercuric oxide (HgO) as well as mercuric chloride (HgCl2) was absorbed from the ligated segments in the following order: duodenum greater than stomach = jejunum = ileum. The ligation of bile duct decreased the duodenal absorption of HgCl2, while no change was observed in that of HgO. In the bile duct-ligated rats, the coadministration of bile increased the absorption of HgCl2 compared to that in rats without the ligation. The absorption of HgCl2 was increased with an increase of pH of the solution perfused into small intestine. These results suggest that the alkalinity of bile promotes the absorption of HgCl2.

Effects of vinpocetine on cyclic nucleotide metabolism in vascular smooth muscle.

A novel vasodilating agent, vinpocetine (14-ethoxycarbonyl-(3 alpha,16 alpha-ethyl)-14,15-eburnamenine) inhibits Ca2+-dependent phosphodiesterase, selectively, among the three forms of cyclic nucleotide phosphodiesterase identified in the rabbit aorta. The concentration of vinpocetine producing 50% inhibition of Ca2+-dependent phosphodiesterase activity was approximately 21 microM, both in the presence and absence of Ca2+-calmodulin (CaM). Increasing the concentration of CaM in the presence of Ca2+ did not prevent vinpocetine-induced inhibition of Ca2+-dependent phosphodiesterase, thereby indicating that vinpocetine inhibited the enzyme by interacting with the enzyme and not with CaM. To determine the influence of vinpocetine-induced inhibition of Ca2+-dependent phosphodiesterase on cyclic nucleotide metabolism in vascular smooth muscle, cyclic nucleotide levels in isolated rabbit aortic strips were also investigated. Addition of vinpocetine produced dose-dependent increases in only the cyclic GMP levels and there was no significant effects on the cyclic AMP levels. These results provide pharmacological evidence that Ca2+-dependent phosphodiesterase mainly hydrolyzes cyclic GMP in vascular smooth muscle. Vinpocetine may induce vascular relaxation by increasing cyclic GMP contents in vascular smooth muscle through selective inhibition of Ca2+-dependent phosphodiesterase.

Vitamin A acid-induced activation of Ca2+-activated, phospholipid-dependent protein kinase from rabbit retina.

Ca2+-activated, phospholipid-dependent protein kinase from rabbit retina was partially purified. Vitamin A acid (retinoic acid) stimulated this protein kinase in the presence of Ca2+, while other metabolites of vitamin A such as retinol or retinal were less effective. The order of the extent of phosphorylation of the various substrate proteins by this protein kinase was identical in the presence of vitamin A acid or phosphatidylserine. The major spots of the 32P labeled peptide from histone H1 phosphorylated in the presence of vitamin A acid by this protein kinase did not differ from those obtained from histone H1 phosphorylated in the presence of phosphatidylserine. Retinol caused a further enhancement of the enzymatic activity, whereas the addition of retinal inhibited the activation by vitamin A acid. Thus, vitamin A and its metabolites may play an important role in the regulation of Ca2+-activated, phospholipid-dependent protein kinase activity in the retina.

Fish egg polysialoglycoproteins: structures of new sialooligosaccharide chains isolated from the eggs of Oncorhynchus keta (Walbaum). Fucose-containing units with oligosialyl groups.

A series of acidic oligosaccharide alditols having different neutral core oligosaccharides were isolated from salmon egg polysialoglycoproteins by alkali-borohydride treatment followed by anion-exchange chromatography and Iatrobead chromatography. Their structures were determined by methylation analysis, molecular secondary ion mass spectrometry of underivatized oligosaccharides, and enzymatic desialylation. The molecular secondary ion mass spectra of intact sialooligosaccharides exhibit pronounced quasi-molecular-ion peaks, (M + H)+, (M + Na)+, (M + 2Na - H)+, and/or (M + K)+, as well as some diagnostic sequence ion peaks. Of a number of oligosaccharide alditols, the following are novel: Fuc alpha 1 leads to 3GalNAc beta l1 leads to 3Gal beta 1 leads to 4Gal beta 1 leads to 3[(leads to 8NeuGc alpha 2)n leads to 6]GalNAcol (n = 1-6). The proton nuclear magnetic resonance spectra of these oligosaccharides are also reported and discussed.
