Hemoglobin synthesis of both adult and fetal types in a human CML cell line.

We present evidence for the continuous erythroid differentiation of a cell line, KU-812-F, without the addition of an inducer. Erythroid differentiation of these cells was confirmed according to the following findings: 1) erythroid morphology; 2) existence of glycophorin A and carbonic anhydrase I as a membrane marker protein and an enzyme characteristic of erythroid cells, respectively; 3) synthesis of adult type (HbA and HbA2) and fetal type (HbF) hemoglobins, as detected on isoelectric focusing; and 4) transcription of the alpha-, beta-, and gamma-globin genes, as detected on Northern blot analysis.

Reduction of size of myocardial infarction with nicorandil, a new antianginal drug, after coronary artery occlusion in dogs.

The effects of nicorandil, a new antianginal drug, on size of myocardial infarction were studied in anesthetized, open-chest dogs after left anterior descending coronary artery occlusion. To quantify the extent of the hypoperfused zone, 99mTc-albumin microspheres were injected into the left atrium 1 min after occlusion. Fifteen minutes after occlusion, dogs were randomly assigned to a control group or a nicorandil-treated group that received immediately after assignments 100 micrograms/kg of nicorandil followed by a continuous infusion of 30 micrograms/kg/min for 6 h. Six hours after occlusion, the left ventricle was cut into 3 mm thick slices for triphenyltetrazolium chloride staining and autoradiography. The extent of the hypoperfused zone (26.1% +/- 3.1% of the left ventricle in the control vs. 23.2% +/- 3.7% in the treated group, mean +/- SEM) was not different between the two groups. The ratio of the extent of myocardial necrosis to the extent of the hypoperfused zone was significantly smaller in the treated group (64.3% +/- 7.2%, n = 7, p less than 0.05) than in the control group (92.6% +/- 9.2%, n = 7). Thus, nicorandil administered early after coronary artery occlusion reduced the size of myocardial infarction by 31%.

Immobilized Datura stramonium agglutinin column chromatography, a novel method to discriminate the urinary hCGs of patients with invasive mole and choriocarcinoma from those of normal pregnant women and patients with hydatidiform mole.

Datura stramonium agglutinin (DSA) binds the asparagine-linked sugar chains commonly found in the human chorionic gonadotropin (hCG) purified from urine of patients with invasive mole or choriocarcinoma, but not in that of normal pregnant women or patients with hydatidiform mole. With the use of an immobilized DSA column, a novel method to discriminate urinary hCGs from various trophoblastic diseases was developed.

Mechanisms of absorption of inorganic mercury from rat small intestine. I. Solvent drag effect on absorption of inorganic mercury.

A correlation of mercury absorption with water absorption was investigated by using the perfusion of rat small intestine with buffers containing 10(-4) M HgCl2. With a decrease in the osmolarity of the buffers or with increases in the sodium ion and urea concentrations in the buffers, absorption of water and mercury and accumulation of mercury in the intestinal tissue increased, and the increases in mercury absorption and accumulation were found to correlate with the increase in water absorption. Both the decrease in osmolarity and the increase in sodium ion concentration increased mercury accumulation in the epithelial cell after the perfusion. But these changes did not alter the mercury distribution in subcellular fractions. The results suggest that the increase in water absorption due to the hyptonicity or the increase in concentration of sodium ion or urea increases the mercury absorption and accumulation in the epithelial cell without change in the distributional pattern of mercury in the cell.

Mechanisms of absorption of inorganic mercury from rat small intestine. II. Composite effects of pH and halide ions on transport of mercuric chloride into isolated brush border membrane vesicles in rats.

Composite effects of pH and halide ions on the transport of HgCl2 into brush border membrane vesicles (BBMV) were investigated in rats. BBMV were incubated for 10 min. in buffer solution at different pH containing 10(-4) M HgCl2. The increase in pH increased the uptake of Hg by BBMV as a result of the increase in transport of Hg into intravesicular space and decrease in binding of Hg to BBMV. The isotonic displacement of NaCl in the buffer solution by LiCl or KCl did not change the uptake of Hg at each pH. The displacement of NaCl by mannitol increased the uptake of Hg at each pH, while the displacement by NaBr or NaI decreased the uptake and diminished the increasing effect of pH on the uptake of Hg. These changes in uptake of Hg due to the displacement were mainly ascribed to the changes in transport of Hg. These results suggest that the increase in pH mainly increases the transport of HgCl2 as a result of the conversion to hydroxide forms of Hg such as Hg(OH)Cl and Hg(OH)2, and Cl- Br- and I- act as the competing ions with OH- and decrease the transport of Hg.

Immunoelectron microscopic demonstration of S-100b protein-like in centriole, cilia, and basal body.

We report here the ultrastructural localization of S-100b protein-like immunoreactivity in the centriole, cilia, and basal body. Duodenum and trachea of guinea pigs and rats were fixed and immunostained by the protein A-gold method. All centrioles, cilia, and basal bodies observed showed clear S-100b protein-like immunoreactivity. Specific colloidal gold particles were located over the microtubules in these cell organelles. However, other microtubules scattered throughout the cytoplasm were devoid of immunoreactivity. Although the functional significance of S-100b protein-like immunoreactivity in the centriole, cilia, and basal bodies remains to be elucidated, the present results introduce new perspectives into the investigation of localization and function of S-100 proteins.

Phagocytotic activities of latex beads by FRTL cell: possible interaction of cell surface glycoprotein.

We investigated the phagocytic activity of cultured rat thyroid cells (FRTL) by using latex beads. FRTL cells ingested many carboxylate-modified polystyrene latex beads (c-latex beads), which had carboxyl residues on the surface, whereas they ingested few polystyrene latex beads (p-latex beads), which had no electrostatic charge. Phagocytotic activity of c-latex beads was stimulated by TSH in a dose-related manner. Dibutyryl, 8-bromo-cAMP, and forskolin also stimulated this process. Tunicamycin significantly inhibited dibutyryl cAMP-stimulated phagocytotic activity of c-latex beads by FRTL cells. N-acetylglucosaminidase also inhibited dibutyryl cAMP-stimulated phagocytosis. These results suggest that the interaction between negative surface charge of c-latex beads and positive surface charge probably induced by N-acetylglucosamine in the cell surface glycoprotein, might play an important role in the phagocytosis of FRTL cells.

Effects of somatostatin and serotonin on calcitonin secretion from cultured rat parafollicular cells.

A primary culture of mammalian parafollicular cells was established from rat thyroid glands in order to investigate the effects of serotonin and somatostatin on calcitonin secretion. Minced rat thyroid glands were dissociated with collagenase and cultured in a Ham's F-12K medium supplemented with calf serum (5%), insulin (1.3 X 10(-6) mol/l), hydrocortisone (10(-8) mol/l), transferrin (6.1 X 10(-9) mol/l), and glycyl-L-histidyl-L-lysin (2.5 X 10(-8) mol/l). Immunohistochemical peroxidase-antiperoxidase method revealed that the cultured parafollicular cells were immunopositive for human calcitonin, and electron microscopy demonstrated the existence of dense secretory granules in the cultured parafollicular cells. Addition of the Ca2+ to the culture medium stimulated calcitonin secretion from the cells dose-dependently as measured by radioimmunoassay. Pre-incubation of serotonin with the cells produced higher calcitonin levels in a dose-dependent manner. On the other hand, pre-incubation of somatostatin with the cells significantly inhibited calcitonin secretion.

[Changes in CT findings in patients with liver metastases of breast cancer during effective intra-arterial infusion].

A study was done of the changes occurring in the CT findings of 12 cases in which liver metastases were found after radical mastectomy for breast cancer, as a result of intermittent low-dose hepatic arterial infusion chemotherapy using a reservoir, achieving PR. In four cases (33.3%) only shrinkage of the metastatic lesions was seen; in 3 others (25.0%), this was accompanied by irregularities in the margins of the liver surface; in the other 5 (41.7%), a high proportion of total, widespread compensatory deformity of the liver was observed. A tendency was noted for these changes to be more pronounced in cases in which the metastatic lesions occupied a large proportion of the liver. Hepatic degeneration appeared with a high frequency in cases where liver metastasis followed breast cancer, and it is considered that considerable care is necessary in evaluating the efficacy of treatment.

Amidolytic kinetic assay of protein C by selective spectrophotometry in a centrifugal analyzer.

This rapid, simple amidolytic assay of protein C activity in whole plasma involves activation by protein C activator from the venom of Agkistrodon contortrix contortrix (Protac) and use of a Cobas Fara spectrophotometer programmed for kinetic assay. Plasma is incubated with activator venom in the presence or absence of antibody to human protein C in the instrument, chromogenic substrate (S-2366) is added, and the absorbance is measured at 405 nm. The difference between the absorbance of the sample plasma with and without antibody to human protein C correlated well with protein C antigen as assayed by enzyme-linked immunosorbent assay (ELISA) and the Laurell rocket technique in normal subjects, patients being treated with warfarin, and patients with liver cirrhosis or disseminated intravascular coagulation. Our mean value for protein C in normal subjects is 115.9 (SD 16.7)% for amidolytic activity, 103.0 (SD 17.4)% for ELISA, and 97.2 (SD 18.1)% for the rocket technique. The high value for normal subjects presumably includes some nonspecific amidolytic activity activated by the activator venom, as indicated by measurable activity in immuno-depleted protein C-deficient plasma. Within-run and between-run CVs were less than 5% at low, normal, and high concentrations of protein C amidolytic activity.

[Prognostic significance of the nuclear DNA content of a small adenocarcinoma of the lung].

DNA ploidy has been determined by flow cytometry in 28 surgical specimens from patients with adenocarcinoma of the lung (tumor diameter, 3.0 cm or less), and the relationship between DNA ploidy and the clinical stage and survival also has been investigated. The cases were classified according to the DNA index (DI) into group A (DI 1.00 to 1.50), group B (DI 1.51 to 2.00) and group C (DI greater than 2.01). A close relationship was observed between the DI and the survival rates but not between the DI and the clinical stage. Tumor ploidy is considered to be one of the prognostic factors and may be useful in selecting adjuvant therapy.
