Mechanism of inactivation of a Fasciola proteolytic enzyme by peptide aldehydes and alkylating agents.

A proteolytic enzyme of the liver fluke Fasciola sp. was purified as described previously by ammonium sulfate fractionation, gel filtration on Sephadex G-200 column and L-phenylalanine-agarose chromatography. Leupeptin, a peptide aldehyde of microbial origin, competitively inhibited the enzyme activity with respect to the substrate alpha-N-benzoyl-L-argininamide; the apparent Ki value for leupeptin is 45 000-fold less than the apparent Km for the substrate. Incubation of the enzyme with leupeptin resulted in time-dependent inactivation of the globinolytic activity, with an inactivation constant (Kinact) of 0.4 microM giving the half-maximum inactivation velocity, and with a minimum inactivation half-time (T) of 2.7 min at infinite concentration of this compound. The inactivated enzyme was not reactivated by extensive dialysis. These results imply that leupeptin yields an affinity labelling of an active site of the enzyme. The activity of the Fasciola proteolytic enzyme was also inactivated by other peptide aldehydes and alkylating agents and inactivation constants observed were 0.5 microM for chymostatin, 13 microM for antipain, 2 microM for p-toluenesulfonyl-L-lysine chloromethyl ketone, 140 microM for p-toluenesulfonyl-L-phenylalanine chloromethyl ketone and 40 microM for iodoacetate under the conditions used.

Tuberous sclerosis: proline and hydroxyproline contents in serum.

The serum levels of proline and hydroxyproline were determined by high-performance liquid chromatography using the post-labeled method with o-phthalaldehyde and sodium hypochlorite in 30 patients of tuberous sclerosis (TS) and compared with those in 32 pathological control subjects of similar age. No significant difference in the levels of serum free hydroxyproline was observed between TS and controls in any age group. In the age group of 9 to 18 years, TS showed significantly higher mean free proline levels in serum than controls with a difference of about 50 mumol/L. In both groups of TS and controls, total and free levels of hydroxyproline from ethanol-extractable serum showed similar age-dependent curves with peak values at age 12 years, and there was also no difference in the 2 levels between the 2 groups. It is suggested that this higher free proline level in TS with the autosomal dominant trait may be explained as the abnormal regulation of proline metabolism.

High incidence of human type-C retrovirus (HTLV) in family members of a HTLV-positive Japanese T-cell leukemia patient.

Sera and peripheral blood cells of an adult T-cell leukemia patient and several clinically normal members of his family from the northwest coast of Japan were examined for evidence of infection with human T-cell leukemia (lymphoma) virus (HTLV). The sera of the patient and his parents had antibodies to HTLV, whereas these antibodies were absent in the sera of the patient's brother and sister. T-cell lines were established from the peripheral blood lymphocytes of all of the family members, and all except the patient's sister expressed HTLV antigens (p19, p24, and reverse transcriptase) and type-C virus particles. Not only the fresh peripheral blood lymphocytes from the patient but also those from his clinically normal mother showed abnormal morphology of the kind characteristic of some patients with T-cell leukemia. These studies are consistent with previous evidence indicative of a high rate of HTLV infection within families, and they show that people whose sera are negative for antibodies may still be infected by HTLV. In addition, the results indicate that infection of T cells by HTLV can be associated with morphological transformation of the cells without other signs of leukemia.

R plasmids in Corynebacterium xerosis strains.

Plasmids coding for resistance to chloramphenicol, erythromycin, kanamycin, streptomycin, and tetracycline have been found in strains of Corynebacterium xerosis isolated from patients with otitis media.

Mechanical, electrical, and morphological characteristics of skeletal muscle fibers from Xenopus and other species of frogs.

Mechanical, electrical, and morphological properties of iliofibularis or semitendinosus of Xenopus laevis, Rana catesbeiana, and Rana nigromaculata were investigated in an attempt to find out the differences between them which will give the basic knowledge for the study of excitation-contraction coupling. With application of electrical stimulation, a single muscle fiber from Xenopus contracted at a faster rate of rise than did the other muscles tested. The maximum rate of rise (Tmax) of tension was in the order of Xenopus, R. catesbeiana, and R. nigromaculata. Ca2+ sensitivity and Tmax of mechanically skinned fibers of Xenopus resembled those of R. catesbeiana. Xenopus muscle had a small cross-sectional area of T-tubule compared with that in other species and the action potential exhibited a small positive-going hump. The volume density of the terminal cisternae of sarcoplasmic reticulum (SR) to the myofibril was the largest in the Xenopus muscle, with a statistically significant difference. Therefore, the Xenopus muscle appears to be good material for investigation of mechanisms related to Ca2+ release from SR, as elicited by the excitation of T-tubules.

Effect of expectorants on relaxation behavior of sputum viscoelasticity in vivo.

We studied the effects of expectorants (mucolytic agents) in vivo on the relaxation behavior of sputum viscoelasticity. Seven female and thirty-three male patients (56.8 +/- 19.3 yrs, range: 21 to 82 years old) with a chronic pulmonary disease except bronchial asthma were studied. They were randomly put into the control group or a group which would be given oral treatments with an expectorant for a week after a one week washout period. The groups were as follows: Group I (n = 8), control; Group II (n = 7), Bromhexine hydrochloride 24 mg per day; Group III (n = 10), Ambroxol 90 mg per day; Group IV (n = 9) alpha - Chymotrypsin buccle 100 ch.u. per day; Group V (n = 6), Serratiopeptidase 30 mg per day. In Groups IV & V, frequency dependence of sputum viscoelasticity at the range of omega = 10(-3) to 10(0) rad.sec-1 were clearly changed after the treatments, and the magnitude of the relaxation and its main relaxation time were significantly increased. On the other hand, in Groups I, II & III, no significant changes of the frequency dependences were observed. These findings suggest that proteolytic enzymes administered orally work on the molecular structure of sputum, and break down their linkages between subunits of the structure.

Multicentre double-blind comparison of piroxicam and indomethacin in the treatment of lumbar diseases.

In a 2-week, double-blind, parallel, multicenter study, piroxicam (20 mg once-daily) and indomethacin (25 mg three times daily) were compared in the treatment of painful lumbar disorders. A total of 230 patients were evaluated, 116 who received piroxicam and 114 who received indomethacin. While both drugs were highly effective in relieving symptoms, numerical superiority was evident for piroxicam in most efficacy parameters. The difference between treatments was most obvious at the end of the first week, when a greater percentage of patients receiving piroxicam was rated as 'very much improved'. No serious adverse reactions or clinical laboratory abnormalities were noted for either drug, although fewer gastrointestinal side effects were observed with piroxicam.

Insulin resistance with pancreatic pseudocyst relieved by percutaneous drainage.

An insulin-requiring type 2 diabetic patient became insulin-resistant with development of a pancreatic pseudocyst following cholecystitis and cholecystectomy. Drainage of the cyst fluid was followed by rapid return to his prior insulin requirement. As there were no indications of insulin allergy, obesity, or hormonal imbalance, we postulate a subclinical state of pancreatitis that was relieved with drainage of the pseudocyst fluid.
