Controlled clinical trial of a rifampicin- trimethoprim combination (Rifaprim) and cotrimoxazole in patients with chronic urinary tract infections.

A controlled clinical trial of Rifaprim (rifampicin 600 mg plus trimethoprim 160 mg, in a single daily dose at 10 p.m.) and cotrimoxazole (two tablets b.i.d.) was carried out in two groups of twenty-one patients, each with chronic or recurrent urinary tract infections. Both treatments lasted 10 days. At the first follow-up, 3-11 days after the end of treatment, bacteriological failure was observed in two Rifaprim patients and in four contrimoxazole patients. Clinical improvement with sterile urine or markedly reduced bacterial count was observed in nineteen. Rifaprim patients and in fifteen cases of the other group. Mild allergic phenomena were observed in three Rifaprim patients, but they did not require discontinuation of treatment; anorexia was complained of by a cotrimoxazole patient. Rifaprim appears a valuable alternative in the treatment of chronic urinary tract infection, mainly because it reduces or abolishes the risk of emergence of resistance.

Chromosomal banding patterns produced by methyl green-pyronin staining after trypsin treatment.

A method is described for producing banding patterns with methyl green-pyronin (MGP) stain in chromosomes of fibrosarcoma cells. 1) The stain was made by mixing equal volumes of 2% aqueous pyronin G, 2% aqueous methyl green, distilled water, and 0.1 M acetate buffer (pH 5.7). 2) Treatment with colcemide and hypotonic KCl (0.075 M) was performed as usual. 3) Metaphase chromosomes were prepared using the flame-drying technique and treated with 0.25% trypsin at 37 C for 45 to 90 seconds. Before staining, the slides were rinsed in PBS, in distilled water, and then were dipped in 0.05 M acetate buffer. 4) Chromosomes were stained for more than 20 minutes, rinsed in distilled water, and hot-air dried. Satisfactory results were obtained in uncontracted metaphase chromosomes. MGP stain has the advantage of permitting much longer trypsin treatment and staining time than the trypsin-Giemsa method while providing satisfactory banding patterns.

EEG evoked response, P300 and CNV during processing of sentence information: clinical application.

Evoked response, P300 and CNV were investigated in the condition in which the subject was required to make a differential response to the acoustically and visually presented sentence with or without meaning, and he was able to determine whether the sentence had a meaning or not by understanding the key information. In healthy persons, P300 amplitude to the beginning of information and to the key information were larger than those to the others. P300 latency to the key character was longer. CNV appeared at the beginning of information and continued. The difference between CNVs produced by meaningful and meaningless sentences was observed after the key information. In aphasia, the difference between CNV's was not observed. In auditory agnosia, the difference between CNVs was not observed to acoustic sentences, but appeared to visual sentences. These results suggest that evoked response, P300 and CNV can be useful for assessing recognition of sentences.

Genetic control of arylsulfatase synthesis in Klebsiella aerogenes.

It was shown that at least four genes are specifically responsible for arylsulfatase synthesis in Klebsiella aerogenes. Mutations at chromosome site atsA result in enzymatically inactive arylsulfatase. Mutants showing constitutive synthesis of arylsulfatase (atsR) were isolated by using inorganic sulfate or cysteine as the sulfur source. Another mutation in which repression of arylsulfatase by inorganic sulfate or cysteine could not be relieved by tyramine was determined by genetic analysis to be on the tyramine oxidase gene (tyn). This site was distinguished from the atsC mutation site, which is probably concerned with the action or synthesis of corepressors of arylsulfatase synthesis. Genetic analysis with transducing phage PW52 showed that the order of mutation sites was atsC-atsR-atsA-tynA-tynB. On the basis of these results and previous physiological findings, we propose a new model for regulation of arylsulfatase synthesis.

Development of mast cells in vitro. II. Biologic function of cultured mast cells.

Mast cells were obtained by long term culture of rat thymus cells on rat embryonic fibroblast monolayers. Pure mast cell preparations obtained culture were incubated with 125I-labeled rat E myeloma protein to study receptors for IgE on their surface. When the cells were obtained after 35 to 45 days culture, the average number of receptors per mast cell was 100,000 to 400,000. An equilibrium constant of the binding reaction between their receptor and rat IgE was in the order of 108 M-1. The histamine content of the cultured mast cells was 0.2 to 5 mug/106 cells. The measurement of histamine content in mast cells recovered after different periods of culture suggested that the histamine content increased with maturation. Even after 45 to 50 days culture, the histamine content of cultured mast cells was significantly lower than that in rat peritoneal mast cells. The cultured mast cells were passively sensitized in vitro with rat IgE antibody against Nippostrongylus brasiliensis. The sensitized cells released histamine upon incubation with the antigen. It was also found that cultured mast cells released histamine upon exposure to compound 48/80. These results indicated that cultured mast cells have physiologic functions similar to those of normal rat mast cells, but they have not reached full maturation.
