Purification and properties of two superoxide dismutases from human placenta.

Copper and zinc-containing superoxide dismutase (Cu,Zn-SOD), and manganese-containing superoxide dismutase (Mn-SOD) were purified from human placenta by heat treatment at 70 degrees for 5 min, following precipitation with ammonium sulfate and column chromatographies. The molecular weights of Cu,Zn-SOD and Mn-SOD estimated by gel filtration were 32000 and 81000, respectively. From the result of SDS polyacrylamide gel electrophoresis, Cu,Zn-SOD consisted of two subunits with an equal molecular weight of 16000 and Mn-SOD consisted of four subunits with an equal molecular weight of 21000. One g-atom each of copper and zinc was contained in the subunit of Cu,Zn-SOD, and one g-atom of manganese was contained in the subunit of Mn-SOD. Immunological properties of superoxide dismutases showed that no organ specificity existed but that a species specificity obviously existed. The method of Laurell's rocket immunoelectrophoresis which can determine Cu,Zn-SOD or Mn-SOD on the basis of the immunological difference has been established. The contents of Cu,Zn-SOD and Mn-SOD in various human tissues were measured by this method.

Purification of superoxide dismutases from human placenta using immunoadsorbent columns.

Copper and zinc containing-superoxide dismutase (Cu,Zn-SOD), and manganese containing-superoxide dismutase (Mn-SOD) were purified form human placenta with immunoadsorbent columns using anti-human SOD-antibody conjugated Sepharose 4B. The final preparation has a high purity with a good recovery. The Cu,Zn-SOD purified by an immunoadsorbent column had less zinc content and was more labile than the native Cu,Zn-SOD, but this difference disappeared completely by dialyzing the enzyme obtained by immunoadsorbent column against the buffer containing 1mM ZnCl2. Other enzymological properties of two SODs were identical with that of native SODs.

Abnormalities of chromosome no. 1 related to blood dyscrasias: study of 10 cases.

Partial excess of chromosome 1 (q25-q32) was noted in malignant cells from all of 10 patients who had disorders such as non-African Burkitt's lymphoma, adult T-cell leukemia, myelofibrosis, malignant lymphoma, chronic lymphocytic leukemia or chronic myelocytic leukemia in blast crisis. The break points on chromosome 1 were at centromere, q12, q21, q23, q25 and q32. Variations in the specific region of the long arm of chromosome 1, q25-q32, were thought to be important in the evolution of malignant cell proliferation.

A 14q+ chromosome in adult T-cell leukemia.

Chromosome studies were conducted on two patients with adult T-cell leukemia. In both patients, a marker chromosome 14q+ and a structural change involving chromosome 1 with trisomy of the q arm were found in peripheral blood leukocytes. The 14q+ marker chromosome had resulted from translocation from #5p in one patient and #5q in the other patient. The present and previous studies suggest that the donor chromosomes involved in the 14q+ translocation are variable. This indicates that the 14q+ marker chromosome rather than the donor chromosome is intimately related with adult T-cell leukemia.

Synthesis and antitumor activity of cytosine and adenine nucleosides of unsaturated 5-(aminoacyl)aminopentofuranoses.

Direct synthesis of the 1- and 9-(5-azido-2,3,5-trideoxy-beta-D-glycero-pent-2-enofuranosyl) derivatives (3a and 3b) of cytosine and adenine, respectively, has been accomplished via treatment of the corresponding 2',3'-unsaturated nucleosides (1a and 1b) with triphenylphosphine and carbon tetrabromide in the presence of lithium azide. Members of a new type of (aminoacyl)amino nucleoside, the 1- and 9-[5-(aminoacyl)amino-2,3,5-trideoxy-beta-D-glycero-pent-2-enofuranosyl] derivatives of cytosine and adenine, respectively, have been obtained by condensation of the corresponding, unsaturated amino nucleosides with the active esters of several amino acid derivatives, followed by deprotection. These nucleosides were examined for in vivo antitumor activity against leukemia L-1210 and Sarcoma 180 (solid tumor) in mice; none of them exhibited antitumor activity against L-1210 in mice, but compounds 1a, 3a, and 1-[2,3,5-trideoxy-5-(L-methionyl)amino-beta-D-glycero-pent-2-enofuranosyl]cytosine exhibited weak activity against Sarcoma 180 (solid tumor).

Treatment of refractory acute leukemia with aclacinomycin-A.

Twelve patients with refractory acute leukemia (7 patients with acute myelocytic leukemia and 5 patients with acute lymphocytic leukemia) were treated with a new anthracycline antibiotic, aclacinomycin-A (ACM). ACM was administrated by intravenous drip infusion at a dose of 20 mg/day for 7 or 14 days and this was repeated after at least 7 days. Four of 12 patients (33.3%) achieved a complete remission; 3 of 7 acute myelocytic leukemia (42.8%) and 1 of 5 acute lymphocytic leukemia (20.0%). The days required for achieving the complete remission ranged from 23 to 78 days (median: 61) and the total doses of ACM used from 180 to 500 mg (median: 310), and the durations of complete remission from 11 to 28+ weeks (median: 21+). The untoward effects on digestive organs, such as nausea, vomiting and anorexia, and hematological toxicities were frequently seen; however, they were controlled by supportive treatment. Alopecia was not observed. Arrythmia was recognized in one patient at the initiation of ACM infusion with complete remission without withdrawal of ACM. These results suggest that ACM is a potentially effective anthracycline antibiotic in the clinical management of acute leukemia.
