A study on functioning for independent living among the elderly in the community.

The primary purposes of this study were to understand the level of functioning for independent living (FIL) as well as to analyze related factors among the elderly. A questionnaire was used to collect data in a face-to-face interview. A total of 247 subjects over 65 years of age completed the questionnaire. The subjects, who lived at home in Keelung City in Taiwan, Republic of China, were interviewed by trained interviewers. Data analysis was performed using a personal computer with SPSS/PC + 5.0. The results indicated the following: The most common diagnoses of the elderly were hypertension, visual impairment, stomach disease, and arthritis. The level of FIL for most subjects ranged from total independence to mild impairment. According to stepwise multiple regression analysis, five significant variables (physical activity status, the number of chronic diseases, health care needs, age and care provider) were the optimal predictors of physical FIL and total FIL. Three variables (physical activity status, number of chronic illnesses, and care provider factors) were the optimal predictors of mental FIL. Finally, recommendations are made regarding health policy, community nursing practice, and further study.

Sambutoxin-producing isolates of fusarium species and occurrence of sambutoxin in rotten potato tubers.

A total of 50 Fusarium isolates representing 13 species from various sources were surveyed to determine their potential to produce sambutoxin. Sambutoxin production was restricted to Fusarium sambucinum and F. oxysporum, with the exception of one isolate of F. semitectum. Sambutoxin was produced by high percentages of F. sambucinum (80.0%) and F. oxysporum (84.6%) isolates at levels of 1.1 to 101.0 (mu)g/g. In addition, 9 (42.9%) of 21 rotten potato samples were contaminated with sambutoxin at levels of 15.8 to 78.1 ng/g.

FRAXE and mental retardation.

Mental impairment and instability of the CCG repeat at FRAXE is described in six kindreds. Cosegregation of FRAXA and FRAXE was found within one of these kindreds. Cytogenetic expression of FRAXE was shown to skip a generation when associated with a reduction in size of the CCG expansion when transmitted through a male; however, in general, transmission occurred through females and a copy number increased from one generation to the next. In these respects the behaviour of FRAXE paralleled that of FRAXA. A relationship between FRAXE and non-specific mental impairment is strongly suggested by the occurrence in these families of more mentally impaired male and female carriers, after removal of index cases, than could reasonably be expected by chance.

Transformation of cultured human monocytes by peroxidized low-density lipoprotein.

To study the roles of peroxidized low-density lipoprotein (pox-LDL) during the formation of foam cells from human monocytes, monocytes isolated from normal human blood were incubated with RPMI alone, normal low-density lipoprotein (n-LDL) and copper pox-LDL. The concentrations of total cholesterol (TCH) and lipid peroxide in the human monocyte medium did not change significantly after incubation with RPMI medium alone or n-LDL, but those of TCH decreased slightly after incubation with pox-LDL for 24 h. Triglyceride (TG) concentrations in the culture medium of human monocytes decreased after incubation with all above substances. Ultrastructural studies showed that the monocytes changed to macrophages after incubation with RPMI alone or n-LDL and to foam cells after incubation with pox-LDL for 48 h. We conclude that TGs may be metabolized for ATP production by human monocytes. The energy may play a role in the transformation of monocytes to macrophages, and pox-LDL can induce transformation of monocyte-derived macrophages to foam cells.

CBP-140, a novel endoplasmic reticulum resident Ca(2+)-binding protein with a carboxy-terminal NDEL sequence showed partial homology with 70-kDa heat shock protein (hsp70).

Antibodies against pokeweed agglutinin binding proteins isolated from F9 embryonal carcinoma cells were used to screen a lambda gt11 expression library constructed from the cells. A cDNA clone thus obtained encoded a novel calcium binding protein of 140 kDa (CBP-140). Antibodies raised against the CBP-140 fusion protein stained a 140 kDa band in extracts not only from F9 cells but also from various mouse organs. A calcium blot experiment using CBP-140 fusion protein verified the calcium binding property of the protein. In the partial amino acid sequence so far clarified (652 amino acid residues) we could not detect EF-hand, but could detect contiguous acidic amino acids, which may serve as a calcium-binding site. CBP-140 showed homology with 70-kDa heat shock protein, though it was not induced by heat shock treatment. Localization of CBP-140 in endoplasmic reticulum was shown by indirect immunofluorescence staining and also by subcellular fractionation. Amino acid sequence of CBP-140 contains a carboxyl-terminal Asn-Asp-Glu-Leu (NDEL) sequence, which resembles Lys-Asp-Glu-Leu (KDEL) sequence, a signal to retain the resident proteins in endoplasmic reticulum; NDEL sequence may indeed play a similar role.

cDNA cloning and sequence analysis of a novel calcium binding protein with oligoproline motif.

Using antibodies against pokeweed agglutinin-binding proteins from F9 embryonal carcinoma cells, we isolated a cDNA clone PW29 reacting with the antibody. mRNA hybridizing with the cDNA was 4.7 kb, and was strongly expressed in the testis, brain, kidney and heart as well as in F9 cells. The size of mRNA in the testis was heterogeneous. Sequencing the cDNA clone revealed a putative polypeptide of 70 kDa, which is rich in hydrophilic amino acids and has a characteristic sequence of (Glu(Lys))5. The protein also has an oligoproline motif, which conforms to the rule of binding capability to Src homology region III. The cDNA was translated as a fusion protein with glutathione-S-transferase, and was verified to have calcium binding activity, upon a calcium blot experiment.

[An immunohistochemical study of HLA-DR expression in salivary glands from patients with Sjogren's syndrome].

Specimens of salivary gland tissue from 21 cases of SS were analyzed by ABC immunohistochemistry technicue with a monoclonal antibody to HLA-DR. The results showed that positive immunostaining for HLA-DR were found in duct cells, acinar cells and a part of infiltrating cells respectively. The proportions of duct and acinar cells expressing HLA-DR were not increased between SS and controls. However, there are differences in their staining intensity. The HLA-DR expression on the epithelial cells in SS was most intense than in controls. Our data suggest that the HLA-DR expression of SS may be regarded as an important factor in local immune rections.

Effect of long-term water storage on dentin bonding.

PURPOSE: To determine the effect of long term water storage on assembly bond strength and interfacial integrity of resin composite bonded to dentin. MATERIALS AND METHODS: Four groups, each containing 10 molar teeth, were established to measure the shear strength of All-Bond 2 and Amalgambond bonded P50 resin composite assemblies at 24 hours and after 6-month water storage at 37 degrees C. An additional four groups of similarly bonded Class V restorations were subjected to the same immersion periods. Marginal leakage was determined using eosin dye and the restoration interface was observed using polyvinyl siloxane impressions to determine gap location. RESULTS: The shear strength values (MPa) for All-Bond 2 were 32.68 +/- 7.12 and 23.29 +/- 5.77 at 24 hours and 6 months respectively. The mean values for Amalgambond were 26.03 +/- 5.94 and 12.90 +/- 6.90. The difference between the values at different immersion times was statistically significant (P < 0.05). There was no evidence of dye leakage in any of the teeth. No gaps were found at the margins of the restorations correlating with the absence of microleakage. Gaps were found commonly at the floor of the Amalgambond restorations.

[Correlated multiple parameters in pleural effusion with pathology of pleurae for differential diagnosis of tuberculous and carcinomatous pleuritis].

The level of ADA, LZM, leukocyte count, lymphocyte in tuberculous effusion is higher than that of carcinomatous effusion (P < 0.05). All results showed that the change of multiple parameters is related to the pathological manifestation of pleurae. The false positive or negative result was influenced by pathological appearance and bloody character of pleural effusion. Using the ADA, LZM cut off value at 35U and 30mg, respectively, and pleural/serum ADA ratio > 1.4, LZM > 1.1, a sensitivity and specificity of 100 percent were achieved.

[Effect of Achyranthes bidentata polysaccharides (ABP) on antitumor activity and immune function of S180-bearing mice].

Achyranthes bidentata polysaccharides (ABP), extracted from the root of Achyranthes bidentata Blume, 25-100mg.kg-1.d-1 x 7 could inhibit tumor growth by 31%-40%. Combination of cyclophosphamide (Cy) and ABP increased the rate of tumor growth inhibition to 58%. ABP 50 and 100% mg/kg ip could potentiate LAK cell activity and increase the Con A (5 micrograms/ml)-induced production of tumor necrosis factor (TNF-beta) from murine splenocytes. The optimal time for TNF production was on d 8. We also found that ABP 1-2 micrograms/ml strongly inhibited the proliferation of S180 and K562 cells in vitro. The S180 cell membrane content of sialic acid was increased and phospholipid decreased after ABP acting on cells for 24 hours. The changes were significantly different from the control group (P < 0.05 or P < 0.01), but the membrane cholesterol content and membrane mobility indices (Ch/PI) were not affected. The results suggest that the antitumor mechanism of ABP may be related to potentiation of the host immunosurveillance mechanism and the changes in cell membrane features.

[Flow cytometry analysis of 67 squamous cell carcinoma of the tongue].

Flowcytometry was used to measure the DNA content in 67 cases with squamous cell carcinoma of the tongue. The relationship of FCM resulte are compared with clinical and pathological parameters. The results show that the recurrences are always appeared within 2 years after operation. Therefore, it is important to follow up patient for the first 2 years after operation. 75% of recurrences are aneuploidy. With multiple statistic analysis, only the clinical stage and aneuploidy are related to lymph node metastasis. The close relative factors to recurrence of SCCT are clinical stage, lymph node metastases, DI value and DNA ploidy.

Effect of warm ischemic injury on morphology and viability of aortic valves in rats.

Warm ischemia is one of the most important causes of valvular damage from death of donor to its valve harvesting. This study aimed at using qualitative and quantitative methods to characterize warm ischemic injury through models of SD rat's aortic valves and design to show the relationship among ultrastructural, biological and biochemical changes concerning with the length of warm ischemia time (WIT). 102 harvested SD rat's aortic valves were divided into 6 groups of different ischemic time for this study. 432 photomicrographs of transmission electron microscopy (TEM) were put into computer for analysis. The volume ratio of nucleus to plasma in cells (Vnp) and the ratio of extramembrane area to volume of mitochondria (S/V) were used to characterize the degree of valvular cell injuries. Valvular cells culture and biochemical metabolism including glucose degradation and 3H-TdR absorption rate were adopted. The valvular cells depicted a significant decrease and 3H-TdR taking-up also being inhibited under the influence of prolonged WIT.

[Observation on left ventricular diastolic function of patients with heart-deficiency syndrome of cardiovascular diseases].

In this paper, left ventricular diastolic function (LVDF) of 99 Heart-Deficiency Syndrome (HDS) patients was observed with pulse Doppler echocardiogram and was compared with normal group. The results showed that different LVDF abnormalities occurred in the HDS patients. When E, Ei DC were decreased and A, Ai, A/E, Ai/Ei, IRT increased, the degree of HDS deteriorated, the pattern was Heart Qi-Yin Deficiency > Heart Yang Deficiency > Heart Qi Deficiency > Heart Yin Deficiency. In conclusion, the above parameters of LVDF could be important indices for evaluation of HDS.

[Nationwide survey of the distribution of human parasites in China--infection with parasite species in human population].

The infection rate of main species of parasites and their character by first nationwide survey of human parasites was made. The overall infection rate of human intestinal parasite and the infection rate of most species of parasites were higher in females than that in males. The infection rate according to the age group, the highest infection rate was found in the group aged 5-14 years. With regard to the relation between parasitic infections and occupations of the infected persons, the highest infection rates of Ascaris lumbricoides were exhibited in school children, of hookworm and Clonorchis sinensis were in halfpeasants and halfmerchant and vegetable grower; of Trichuris trichiura were in fishers, of Giardia lamblia, Entamoeba histolytica and Taenia were in herdsmen and halfherdsmen and halfpeasants. In this survey it is also shown that each nationality has their main species of parasites. The family clustering of some main parasites were proved by some province/autonomous region/municipality.
