Effect of ethanol on hepatocarcinogenesis initiated in rats with 3'-methyl-4-dimethylaminoazobenzene in the absence of liver injuries.

In this study special care was taken to discriminate between the direct effect of ethanol on hepatocarcinogenesis and secondary effects such as choline deficiency or fatty liver. Rats were divided into 2 groups, D and N. Group D was initiated using 3'-Me-DAB, while the control group N was not initiated. Groups N and D were divided into 4 sub-groups. Each subgroup was given, in the drinking water, one of the following ethanol solutions: 0, 5, 10 or 15% for 45 weeks. Liver tumors were induced only in D groups. Evidence obtained indicates that: (a) in the N group, ethanol did not cause any apparent disorders in histology and lipid metabolism, and (b) in the D group no significant differences were observed in the incidence of HCCs and other liver lesions. Ethanol thus does not appear to enhance hepatocarcinogenesis, at least in the absence of liver injury.

Barium granuloma involving urinary bladder.

The first case of barium granuloma involving urinary bladder is reported. Surgical removal was the treatment of choice. Histological diagnosis was foreign body granuloma with xanthomatous inflammation. The foreign body was identified as barium by analytical electron microscopy.

[Transurethral treatment of bladder diverticulum].

13 patients with bladder diverticula underwent transurethral diverticulectomy. Follow-up cystograms were obtained six months after surgery. The diverticulum had shrunk in all cases and had totally disappeared in 11 (85%). Transurethral diverticulectomy is a simple, effective, and safe procedure. It takes only a short time and can easily be combined with transurethral operations for obstructions of the lower urinary tract.

[The significance of serum prostate-specific antigen, gamma-seminoprotein and prostatic acid phosphatase as prostate cancer markers].

Prostate-specific antigen (PA), gamma-seminoprotein (gamma-Sm) and prostatic acid phosphatase (PAP) have been evaluated in patients with prostate cancer, benign prostatic hyperplasia (BPH), chronic prostatitis and acute prostatitis. PA has proved to be diagnostically more sensitive than PAP and gamma-Sm for the detection of prostatic cancer. Although PA may be elevated more frequently than PAP and gamma-Sm in patients with BPH, there are possibilities that these patients with elevated PA and normal PAP and gamma-Sm may have prostatic cancer or precancerous conditions not detectable in our routine diagnostic procedures. We report two cases of prostatic cancer with persistently elevated PA and diagnosed after repeated biopsies. Our data suggest that PA is a sensitive and useful tumor marker for the diagnosis of prostatic cancer. PAP and gamma-Sm in combination with PA may serve as more useful for differential diagnosis and confirmation of prostatic cancer.

Purification and characterization of cytosolic protein-tyrosine kinase from bovine platelets.

A cytosolic protein-tyrosine kinase has been highly purified from bovine platelets using [Val5]angiotensin II as a substrate. The purification procedure involves sequential column chromatography on phosphocellulose, Sephacryl S-200, poly(L-lysine)-agarose, casein-Sepharose 4B and 2',5'-ADP-Sepharose 4B. Analysis of the most highly purified preparations by SDS/polyacrylamide gel electrophoresis revealed a major silver-stained band of molecular mass 71 kDa. This molecular mass was consistent with results obtained from sucrose density gradient centrifugation, indicating that the enzyme exists as a monomer. The purified kinase, called CPTK 71, efficiently phosphorylated tubulin and p36 (calpactin 1 heavy chain). However, it did not phosphorylate H1 histone. Half-maximal enzyme activity was observed at 2.2 microM ATP, and Mn2+, Co2+ and Mg2+ were effective divalent metal ions for the expression of activity. Insulin, epidermal growth factor, and platelet-derived growth factor had little or no effect on the kinase activity of CPTK 71. CPTK 71 had no immunological cross-reactivity with pp60src. These results suggest that CPTK 71 is a novel type of protein-tyrosine kinases among the enzymes so far reported.

Protein phosphorylation and activation of human platelets by sodium fluoride.

The ability of sodium fluoride (NaF) and thrombin to stimulate aggregation and protein phosphorylation in intact human platelets was measured and compared. When platelets were stimulated by NaF, phosphorylation of the 20 KDa protein was transient and after 5-10 min returned to the same level as that of unstimulated cells. On the other hand, 47 KDa protein was slowly phosphorylated without obvious dephosphorylation. The slow activation of the 47 KDa protein phosphorylation correlated well with the time required for the aggregation and secretion. Phosphoamino acid analysis showed that the phosphorylated amino acids of the 47 KDa protein from platelets activated by NaF and thrombin were slightly different. These results suggest that different stimuli may lead to the same protein phosphorylation by different biochemical mechanisms of action.

[Incidence of renal diseases in the general population].

The purpose of this study was to determine the incidence of the urological diseases of the kidney. Among 4,094 persons (3,146 men and 948 women) studied, 86 cases of renal cyst, 7 cases of urolithiasis, 3 cases of cystic kidney, and one case of renal cell carcinoma were discovered. The rate of renal cyst was two times higher in men than in women. One sixth of renal cyst patients showed microscopic hematuria. Renal cysts tended to progress in number and size with age. The size of the renal cyst did not correlate with the appearance rate of microscopic hematuria.

Inhibitory effects of niacin and its analogues on induction of ornithine decarboxylase activity by diethylnitrosamine in rat liver.

Pharmacological doses of niacin and its analogues were given intraperitoneally to rats with and without coadministration of a hepatocarcinogenic dose of diethylnitrosamine (DEN), and their effects on the induction of ornithine decarboxylase (ODC, EC 4.1.1.17) activity in the rat liver were studied. The induction of ODC activity by DEN was inhibited by 74.3, 85.5, 94.6, 97.6, 72.6 and 55.2% by nicotinamide, nicotinic acid, 3-hydroxymethylpyridine, beta-picoline, pyridine-3-aldehyde and ethylnicotinate respectively. When given alone, these analogues did not induce ODC activity. All these compounds are known to have a niacin effect. DEN-induced ODC activity was also inhibited by 84.0, 93.3, 52.8 and 75.9% by 6-aminonicotinamide, picolinic acid, pyridine-3-sulfonic acid and thionicotinamide, respectively, but, peculiarly, they induced ODC activity by their administration alone. These niacin analogues are known to have anti-niacin effects. Tryptophan, N'-methylnicotinamide and isonicotinic acid hydrazide did not affect the DEN-induced ODC activity but could induce ODC by themselves. Tryptophan belongs to the former group and isonicotinic acid hydrazide to the latter group. The reason for these discrepancies is discussed.

Promotion of hepatocarcinogenesis by suxibuzone in rats initiated with 3'-methyl-4-dimethylaminoazobenzene.

Among phenylbutazone (PZ) and its related compounds, suxibuzone (SUX) caused the most extensive decrease in pyruvate kinase (PK) activity with lower toxicity. Therefore, we studied the effect of SUX on rat hepatocarcinogenesis to confirm our assumption that an agent which causes a prolonged decrease in PK activity in rat liver promotes hepatocarcinogenesis. For initiation rats were fed a diet containing 0.06% 3'-methyl-4-dimethylaminoazobenzene (3'-Me-DAB) for 4 weeks. At the end of 53 weeks of the experiment the incidences of liver tumors were 14.3 and 70.0% in the rats fed basal diet and in the rats fed 0.5% SUX diet, respectively, after the initiation. No tumors were observed in rats fed the SUX diet without the initiation. The result shows that SUX promotes hepatocarcinogenesis and supports the above assumption.

[Developmental studies on the foramen rotundum, foramen ovale and foramen spinosum of the human sphenoid bone].

For the purpose of these studies 111 cases of the fetus skulls, 25 cases of the juvenile skulls and 220 cases of the adult skulls were used and the following results were obtained; 1. The perfect ring-shaped formation of the foramen rotundum is observed in all cases of the fetuses after the 4th fetal month. The earliest perfect ring-shaped formation of the foramen ovale is observed in the 7th fetal month and the latest in 3 years after birth. The earliest perfect ring-shaped formation of the foramen spinosum is observed in the 8th month after birth and the latest in 7 years after birth. 2. The foramen rotundum is mostly oval-shaped in the fetal period and it is round-shaped in general after birth. The majority of the foramen ovale is oval-shaped and it is irregular in shape compared with the rest of two types of foramina. The majority of the foramen spinosum is round in shape. 3. The average diameter of the foramen rotundum is 3.55 mm in the adult. The average maximal length of the foramen ovale is 7.48 mm and its average minimal length is 4.17 mm in the adult. The average diameter of the foramen spinosum is 2.63 mm in the adult. The differences between the right and left and the sexual differences fail to be observed in all types of foramina.

Comparative study on pyruvate kinase activity-reducing action of various promoters of hepatocarcinogenesis.

Chemicals such as PB, DDT, EED, sodium deoxycholate, thiobenzamide, orotic acid and SorFAE which enhance the formation of liver tumors or HN, caused a marked decrease in PK activity in Wistar rat liver that persisted for at least 4 weeks, though the decrease caused by EED was not so marked as that caused by other hepatic promoters. Non-promoting agents such as DH, and some chemicals, such as DEXA, testosterone and AB, which enhance the formation of GGT-positive foci in rat livers, but do not enhance the formation of tumors or HN, caused decreases in PK activity in some cases. Any such decreases, however, were only transient and normal levels were restored within 4 weeks. The results suggest that the continuous decrease in PK activity may be a useful marker in screening tests for the promoters of hepatocarcinogenesis.

Therapeutic hepatic vein angioplasty for Budd-Chiari syndrome.

The authors report a case of Budd-Chiari syndrome treated by percutaneous transluminal angioplasty (PTA). In this case, the occlusion of three major hepatic veins with a big collateral to the inferior vena cava via the right inferior hepatic vein (RIHV) and stenosis of the ostium of RIHV were seen. We performed successful PTA of this stenosis.

Changes in the volume and histology of retractile testes in prepubertal boys.

Testicular development was studied in prepubertal boys with retractile testes. Testicular volume, the diameter of the seminiferous tubules and the number of spermatogonia in the tubules were decreased in cases of unilateral retractile testis, when compared with values for the contralateral normally descended testis. On the other hand, in patients with a unilateral retractile testis and contralateral inguinal testis, there was no difference in the developmental parameters between the two testes. These results suggest that the retractile testis has developmental failures characteristic of a cryptorchid testis and therefore requires orchiopexy.

Enhancement of hepatocarcinogenesis by sorbitan fatty acid ester, a liver pyruvate kinase activity-reducing substance.

Effect on hepatocarcinogenesis of dietary sorbitan fatty acid ester (SorFAE), which had been known to cause decrease in pyruvate kinase (PK) activity, was studied in rats fed a diet containing 0.06% 3'-methyl-4-dimethylaminoazobenzene (3'-Me-DAB) for 6 weeks. The incidence of hyperplastic nodules and/or hepatocellular carcinomas in the rats fed the 3'-Me-DAB diet alone was 45.0% at the end of 51 weeks, whereas the incidence in the rats fed 3'-Me-DAB diet followed by 5 or 10% SorFAE or 0.1% phenobarbital (PB) diet were 76.2, 90.5, and 95.0%, respectively. These incidences were significantly higher compared with the group fed 3'-Me-DAB diet alone (P less than .05). No tumors were observed in rats fed 10% SorFAE diet alone. The results show that SorFAE has an enhancing effect on hepatocarcinogenesis, although the effect was weak compared to that of the effective PB dose. The results seem to confirm our assumption that a chemical that causes decrease in PK activity in rat liver might promote hepatocarcinogenesis.

Effects of margarine on hepatocarcinogenesis by N,N'-2, 7-fluorenylenebisacetamide in mice.

The effect of a safflower margarine on hepatocarcinogenesis by N,N'-2, 7-fluorenylenebisacetamide was compared with that of safflower oil from which the margarine was made. A high-dextrin diet was used as a control for the two high-fat diets. The number of surviving mice in the margarine group at the end of the experiment was significantly lower compared with both the dextrin and the oil groups. A significantly high incidence of hepatic nodules type 2 was observed in the margarine group, whereas most liver tumors in both the dextrin and the safflower oil groups were of HN type 1 (p less than 0.05). Hepatocellular carcinomas were induced in some mice in all the groups. However, no statistical difference was found among the three dietary groups. Thus the difference in the types of the hepatic nodules among the three groups was not reflected in the incidence of hepatocellular carcinoma.
