[Determination of electron density in Ar-air mixed coolant gas ICP].

The lateral distribution of electron number density at 0, 2, 5, 10 mm above the load coil in Ar-air mixed coolant gas ICP was measured with Stark broadening method of Hbeta line (486.1 nm) and some of the lateral distribution was converted into radial distribution by Abel inversion under the condition similar to that in the low power Ar-ICP. The results show that the distribution of electron number in Ar-air mixed coolant gas ICP is similar to that in Ar-ICP and exhibit off axis maxima. When a lot of air was added to the coolant, the positions of the off axis maxima would near the axis. The electron number density reached maximum and was higher than that in Air-ICP as 5%-10% air was added to the coolant.

[Indirect determination of industrial chlordane by ultraviolet spectrometry].

Toxicity in chlordane is in proportion to the content of hexachlorocyclopentadiane. Choosing acetone as the solvent, we determined that content by ultraviolet spectrometry, and then calculated roughly the content of this industrial chlordane. The method is cheaper, more rapid than gas chromatography, and suited to rapid testing industrial chlordane.

[The effect of surface structure on the photoluminescence of SnO2 nanoparticles in hydrosols and organosols].

In this paper, we report the optical properties of SnO2 semiconductor nanoparticles in hydrosols and those of SnO2 semiconductor nanoparticles in organosols in which the surfaces of the particles are coated by a layer of organic surfactant molecules. The photoluminescence spectra of SnO2 semiconductor nanoparticles in the hydrosols and organosols in different conditions were measured and discussed. We conclude that the surface structure of the SnO2 semiconductor nanoparticles affects their optical properties strongly. The oxygen deficiencies on the surface of SnO2 semiconductor nanoparticles play an important role in the optical properties. The surface modification of the particles effectively removes the surface defects of the particles and enhances the intensity of luminescence.

Diagnosis of mucosa-associated lymphoid tissue (MALT) lymphoma in gastroendoscopic biopsy specimens.

UNLABELLED: We evaluated the diagnostic criteria of gastric lymphoma in endoscopic biopsies. METHODS: Our study included cases diagnosed as lymphoma by gastroendoscopic biopsy between 1984 and 1994 that were available in the files of the Department of Gastroenterology, Chinese PLA General Hospital. RESULTS: Among 53,400 cases of gastroendoscopy, we found 1672 (3%) cases of malignant neoplasm that were diagnosed by endoscopic features and histological findings. Thirty-eight cases of primary gastric lymphoma included 22 men and 16 women 16 to 82 years old, with a median age of 47.7. The endoscopy showed the ulcerative type in 12 cases, diffuse infiltrating type in 11 cases, massive infiltrating type in 6 cases, and large mucosal folds type in 4 cases. Five cases had a mixed type pattern of involvement. Histologically, 34 cases showed mucosa-associated lymphoid tissue lymphoma (89.5%), 2 cases had lymphoblastic lymphoma, and 2 cases were unclassifiable because of crush artifact of neoplastic cells. The majority of cases of primary gastric lymphoma have morphologic and clinical features that justify their inclusion in the category of low-grade lymphoma of mucosa-associated lymphoid tissue (MALT).

Targeted disruption of the mouse phospholipase C beta3 gene results in early embryonic lethality.

In order to investigate the biological function of phosphatidylinositol-specific phospholipase C (PLC) we generated mutant mice by gene targeting. Homozygous inactivation of PLCbeta3 is lethal at embryonic day 2.5. These mutants show poor embryonic organization as well as reduced numbers of cells. Identical phenotypes were recorded in homozygous mutants generated from two independently targeted embryonic stem cell clones. Heterozygous mutant mice, however, are viable and fertile for at least two generations. We also showed that mouse PLCbeta3 is expressed in unfertilized eggs, 3-cell and egg cylinder stages of embryos. In conclusion, these results indicate that PLCbeta3 expression is essential for early mouse embryonic development.

The light chain of CD98 is identified as E16/TA1 protein.

The 80/40-kDa CD98 protein complex was purified using an anti-CD98 heavy chain monoclonal antibody coupled to Sepharose beads. Eluted proteins were subjected to preparative SDS-polyacrylamide gel electrophoresis, and protein corresponding to the 40-kDa CD98 light chain was excised. Following proteolysis with trypsin, a peptide fragment was sequenced by mass spectrometry. The nine residues obtained were identical to established C-terminal sequences of the human E16 and rat TA1 proteins, suggesting that TA1/E16 protein is the CD98 light chain. Consistent with this, anti-TA1/E16 antibodies specifically immunoblotted the approximately 35-40-kDa light chain present upon immunoprecipitation of the human CD98 complex. Furthermore, anti-CD98 heavy chain antibody specifically co-immunoprecipitated hemagglutinin-tagged light chain from cells transfected with hemagglutinin-tagged E16 cDNA. In conclusion, the CD98 light chain is identical to the TA1/E16 protein, based on partial amino acid sequence identity, antibody cross-reactivity, genetic reconstitution evidence, similar molecular size, and comparable cell distribution.

Adenoassociated virus-mediated transfer of a functional water channel into salivary epithelial cells in vitro and in vivo.

Aquaporin 1 (AQP1) is the archetypal member of a family of integral membrane proteins that function as water channels. Previously we have shown that this protein can be expressed transiently from a recombinant adenovirus (AdhAQP1) in vitro in different epithelial cell lines, and in vivo in rat submandibular glands. In the present study we have constructed a recombinant adenoassociated virus (rAAV) containing the human aquaporin 1 gene (rAAVhAQP1). rAAVhAQP1 was produced at relatively high titers. Approximately 10(11)-10(12) particles/ml and approximately 10(8)-10(9) transducing units/ml. We show that the rAAVhAQP1 can transduce in vitro four epithelial cell lines of different origins, at a level sufficient to detect the recombinant hAQP1 protein by either Western blot or confocal microscopic analysis. The recombinant hAQP1 was correctly targeted to the plasma membranes in all cell lines. Function of the recombinant hAQP1 was measured as fluid flow, in response to an osmotic gradient, across a monolayer of transduced epithelial cells. The data show that even at a low level of transduction, typically approximately 10% of the cells in the monolayer, transepithelial fluid movement is enhanced about threefold above basal levels. In addition, we report that rAAVhAQP1 can transduce epithelial cells in the salivary glands and liver of mice in vivo. These results suggest that rAAVs may be useful gene transfer vectors to direct the production of functional transgenes in salivary epithelial cell types.

Conformationally constrained analogues of diacylglycerol (DAG). 15. The indispensable role of the sn-1 and sn-2 carbonyls in the binding of DAG-lactones to protein kinase C (PK-C).

The binding mode of DAG-lactones to PK-C was investigated using the C1b domain from the X-ray structure of the phorbol ester/C1b complex of PK-C delta as a template. Modeling experiments revealed two binding alternatives in which one of the carbonyls of the DAG lactones remained uninvolved with the protein. Experimentally, however, the removal of either sn-1 or sn-2 carbonyls caused a dramatic drop in binding affinity towards PK-C. Although it was not possible to discriminate between the two binding alternatives of the DAG-lactones, the study demonstrates an important role for the additional carbonyl group. The function of this group could be equivalent to that of the C-9(OH)/C-13 (C = O) motif in phorbol esters, which also appears free of interactions in the phorbol ester/C1b complex. This role presumably reflects interaction with the phosholipid head groups required for high affinity binding under the conditions of the biological assays.

Chemical and Photochemical Dual Polymerization in a Mixed Langmuir Monolayer of Diacetylene Derivatives and Octadecyltrimethoxysilane.

A chemical-and-photochemical-induced dual polymerization at the air-water interface was performed for the first time, by using a ternary mixture of a octadecyltrimethoxysilane (C18TMS), 10,12-pentacosadiynoic acid (PDA), and N-(11-O-alpha-d-mannopyranosyl-3,6,9-trioxa) undecyl 10,12-pentacosa-diynamide (PDTM). Different dual polymerization behaviors of the mixed monolayers of PDA/PDTM/C18TMS (7:1:8 molar ratio) at different subphase pH were monitored by surface pressure-area isotherms and UV-vis spectra. At pH 1, the C18TMS formed a long linear polymer network by strong intermolecular condensation. The PDA/PDTM was isolated to small domains by this network and can only be polymerized weakly upon UV irradiation. At pH 2.6, the weak condensation of C18TMS formed short linear polymers, which caused phase separation between C18TMS and PDA/PDTM. The strong photochemical polymerization of the PDA/PDTM rich domain was similar to the PDA/PDTM binary mixed monolayer. At pH 4.0, the partially hydrolyzed C18TMS was miscible with PDA/PDTM and slowed the polymerization process of PDA/PDTM upon UV irradiation. These results show geometry and kinetic controls of the PDA/PDTM polymer in 2D by using the C18TMS polymer network. Copyright 1998 Academic Press.

Derivation of pluripotent stem cells from cultured human primordial germ cells.

Human pluripotent stem cells would be invaluable for in vitro studies of aspects of human embryogenesis. With the goal of establishing pluripotent stem cell lines, gonadal ridges and mesenteries containing primordial germ cells (PGCs, 5-9 weeks postfertilization) were cultured on mouse STO fibroblast feeder layers in the presence of human recombinant leukemia inhibitory factor, human recombinant basic fibroblast growth factor, and forskolin. Initially, single PGCs in culture were visualized by alkaline phosphatase activity staining. Over a period of 7-21 days, PGCs gave rise to large multicellular colonies resembling those of mouse pluripotent stem cells termed embryonic stem and embryonic germ (EG) cells. Throughout the culture period most cells within the colonies continued to be alkaline phosphatase-positive and tested positive against a panel of five immunological markers (SSEA-1, SSEA-3, SSEA-4, TRA-1-60, and TRA-1-81) that have been used routinely to characterize embryonic stem and EG cells. The cultured cells have been continuously passaged and found to be karyotypically normal and stable. Both XX and XY cell cultures have been obtained. Immunohistochemical analysis of embryoid bodies collected from these cultures revealed a wide variety of differentiated cell types, including derivatives of all three embryonic germ layers. Based on their origin and demonstrated properties, these human PGC-derived cultures meet the criteria for pluripotent stem cells and most closely resemble EG cells.

Sequence alterations of insulin-like growth factor binding protein 3 in neoplastic and normal gastrointestinal tissues.

Insulin-like growth factor binding protein 3 (IGFBP-3) is an important regulator of normal and malignant cell growth. It modulates the mitogenic effects of insulin-like growth factors (IGFs) by inhibiting growth through mechanisms both dependent on and independent of IGF binding. IGF-I and IGF-II levels are regulated by binding to the IGF-II receptor, which is inactivated by mutation in human gastrointestinal (GI) tumors. We have previously demonstrated elevated IGF-II ligand expression in IGF-II receptor-mutant GI tumors, implicating the IGF signaling system in GI tumorigenesis. Therefore, to investigate the potential involvement of IGFBP-3 in human GI carcinogenesis, direct DNA sequencing of exons 1-4 and intron-exon boundaries of the IGFBP-3 gene was performed in 10 colorectal cancers, 10 gastric cancers, and 10 esophageal cancers. Four distinct sequence alterations were identified: (a) in one gastric and one esophageal tumor, an A to C transversion occurred at nucleotide 5795 (CAC-->CCC), leading to a His-->Pro substitution at codon 179; (b) a second esophageal tumor had a C to T transition at nucleotide 8291 (ACC-->ATC), leading to a Thr-->Ile substitution at codon 277 of IGFBP-3; (c) one alteration comprised a G to C transversion in exon 1 at nucleotide 2132 (GGG-->GCG), leading to a Gly-->Ala substitution at codon 32 in two gastric cancers, seven esophageal cancers, and nine colon cancers; and (d) a C to G transversion located 17 nucleotides from the 3' splice site in intron 1 was observed in three colon cancers and four esophageal cancers. All of these DNA sequence alterations were present in matched normal DNA from the same subjects, which suggests that some or all of them may represent polymorphisms. However, we cannot exclude the possibility that the germ-line nonconservative amino acid substitutions predicted to occur as a result of these alterations result in subtle changes to IGFBP-3 protein function and a predisposition to developing GI malignancy.

Capsaicin-responsive NADH oxidase activities from urine of cancer patients.

NADH oxidases of low specific activities from urine of cancer patients were found to be inhibited or stimulated by the vanilloid capsaicin (8-methyl-N-vanillyl-6-noneamide). Similar activities, inhibited or stimulated by capsaicin, were reported previously for sera of cancer patients but not for sera of normal volunteers or for patients with disorders other than cancer. Like those from sera, the activities from urine were resistant to heat and to digestion with proteinase K. Two different fractions with capsaicin-responsive NADH oxidase activities were obtained by FPLC. One fraction in which the 33-kDa band was the major component exhibited NADH oxidase activity stimulated by capsaicin. Another fraction in which 66-kDa and 45-kDa bands were major components exhibited NADH oxidase activities inhibited by capsaicin. A monoclonal antibody generated to a ca 34-kDa form of the NADH oxidase from sera reacted with a urine protein of a ca 33-kDa band in the capsaicin-stimulated fraction. The 33-kDa protein was of low abundance and was estimated to be present in amounts between 5 and 100 microgram/L, depending on the particular patient.

Structural determinants of Ca2+ exchange and affinity in the C terminal of cardiac troponin C.

The C terminal of cardiac troponin C (TnC) has two Ca2+-Mg2+ sites which exhibit approximately 20-fold higher Ca2+ affinity than the two C-terminal Ca2+ specific sites in calmodulin (CaM). Substitution of the third EF-hand of TnC for the corresponding EF-hand of CaM produced a mutant (CaM[3TnC]) with a 10-fold higher C-terminal Ca2+ and Mg2+ affinity. Substitution of loop 3 of TnC for loop 3 of CaM produced a mutant (CaM[loop3TnC]) with a 10-fold faster Ca2+ on rate and a 5-fold faster Ca2+ off rate than CaM. A mutant CaM (CaM[loop3X, Z]) which contained the identical coordinating amino acids and X and Z acid pairs of TnC loop 3 had a 3-fold higher C-terminal Ca2+ affinity without the increased Ca2+ exchange rates exhibited by CaM[loop3TnC]. Thus, loop factors other than the acid pairs must be responsible for the rapid Ca2+ exchange rates of CaM[loop3TnC]. Helix 6 and helix 5 in the third EF-hand of TnC support the rapid Ca2+ on rate of TnC's loop 3 and produce an approximately 4-fold reduction in its Ca2+ off rate, explaining the high Ca2+ affinity of the third EF-hand of TnC. Exchanging loop 3 or helix 5 of TnC into CaM increased the Mg2+ affinity by decreasing the Mg2+ off rate. Our results are consistent with the high Ca2+ and Mg2+ affinity of the third EF-hand of TnC resulting from the two (X and Z) acid pairs in loop 3, coupled with the greater hydrophobicity of helix 6 and helix 5 compared to that of the third EF-hand of CaM.

Evidence of a functional role for the cyclin-dependent kinase inhibitor p21(WAF1/CIP1/MDA6) in the reciprocal regulation of PKC activator-induced apoptosis and differentation in human myelomonocytic leukemia cells.

The functional role of the cyclin-dependent kinase inhibitor p21(WAF1/CIP1) in leukemic cell G1 arrest, differentiation, and apoptosis induced by two PKC activators (PMA and bryostatin 1) was examined using antisense-expressing lines [U937/p21AS(F4) and U937/p21AS(B8)]. Following incubation with 10 nM PMA (24 h), antisense-expressing cells displayed induction of p27(KIP1) but not of p21, whereas empty vector-containing cells (U937/pREP4) exhibited induction of both p21 and p27. Antisense-expressing cells were impaired in G1 arrest, dephosphorylation of the retinoblastoma protein, dephosphorylation and reduction in activity of cyclin-dependent kinase 2, and acquisition of differentiated features (e.g., plastic adherence). Bryostatin 1 induced p27 but not p21 in control cells and was less effective than PMA in initiating G1 arrest and related events. Nevertheless, disruption of p21 expression abrogated the effects of bryostatin 1 on cell cycle arrest and cellular maturation. Dysregulation of p21 did not, however, modify PMA- or bryostatin 1-mediated down-regulation of c-Myc protein. Unexpectedly, disruption of p21 failed to attenuate the net reduction in viable cell number following PMA or bryostatin 1 treatment inasmuch as impaired differentiation was accompanied by a lowered threshold for PMA- and bryostatin 1-induced apoptosis. Inhibition of p21 expression also promoted PMA- and bryostatin 1-mediated loss of mitochondrial transmembrane potential (DeltaPsim ) and release of cytochrome c into the cytosol. Together, these findings demonstrate a critical functional role for p21 in regulating myelomonocytic leukemic cell G1 arrest and differentiation following exposure to two PKC activators exhibiting disparate patterns of activity. They also suggest that following treatment with these agents, dysregulation of p21 prevents leukemic cells from engaging a normal differentiation program through a c-Myc-independent mechanism, and instead directs cells along an apoptotic pathway.

[A study of correlation between essential hypertension and HLA-DQA1 alleles].

OBJECTIVE: To analyse the hereditary susceptibility by genotyping of HLA-DQA1 alleles in essential hypertensives. METHODS: The allelic types of HLA-DQA1 were detected by PCR-SSP technic in 52 cases of essential hypertensives and 86 normal individuals as the control. RESULTS: The frequency of HLA-DQA1*0302 allele in hypertensive group was markedly higher than that in normal control group (17.9572 vs 3.5531), but the frequency of HLA-DQA1*0103 allele in normal group was higher than that in hypertensive group. CONCLUSION: HLA-DQA1*0302 may be a correlative gene of essential hypertension, whereas HLA-DQA1*0103 may be a defence gene of hypertension.

An acquired sensitivity to H2O2-induced apoptosis during neuronal differentiation of NT2/D1 cells.

Brief exposure of neuronally differentiated human NT2/D1 cells to hydrogen peroxide induced cell death by apoptosis with an ED50 of 30 microM, whereas a 70-fold higher concentration was required to obtain an ED50 effect in undifferentiated NT2/D1 neuronal precursor cells. This enhanced sensitivity in NT2/D1 neurons was correlated with an 8-fold lower level of intracellular glutathione. Pretreatment with N-acetyl-L-cysteine, an agent that is able to raise the levels of intracellular glutathione, promoted the survival of hydrogen peroxide-treated NT2/D1 neurons. Thus, the low glutathione level may contribute to the high sensitivity of NT2/D1 neurones to hydrogen peroxide-induced apoptosis. This study indicates that neuronal susceptibility to oxidative damage is developmentally regulated.

A drug-responsive and protease-resistant peripheral NADH oxidase complex from the surface of HeLa S cells.

Our laboratory described a ca. 34-kDa protein of the HeLa S cell surface that bound an antitumor sulfonylurea N-(4-methylphenylsulfonyl)-N'-(4-chlorophenyl) urea (LY181984) with high affinity and that exhibited NADH oxidase and protein disulfide-thiol interchange activities also inhibited by LY181984. The quinone site inhibitor 8-methyl-N-vanillyl-6-noneamide (capsaicin) also blocked these same enzymatic activities. Using capsaicin inhibition as the criterion, the drug-responsive oxidase was released from the surface of HeLa S cells and purified. The activity of the released capsaicin-inhibited oxidase was resistant to heating at 50 degrees C and to protease digestion. After heating and proteinase K digestion, the activity was isolated in >90% yield by FPLC as an apparent 50- to 60-kDa multimer. Final purification by preparative SDS-PAGE yielded a capsaicin-inhibited NADH oxidase activity of a specific activity indicative of >500-fold purification relative to the plasma membrane. The final activity correlated with a ca. 34-kDa band on SDS-PAGE. Matrix-assisted laser desorption mass spectroscopy as well as reelectrophoresis of the 34-kDa band indicated that the ca. 34-kDa material was a stable mixture of 22-, 17-, and 9.5-kDa components which occasionally migrated as a ca. 52-kDa complex. The purified complex tended to multimerize and formed insoluble 10- to 20-nm-diameter amyloid rods. The components of the purified 34-kDa complex were blocked to N-terminal amino acid sequencing and were resistant to further protease digestion. After multimerization into amyloid rods, the protein remained resistant to proteases even under denaturing conditions and to cyanogen bromide either with or without prior alkylation.
