[Seven years' summary report on the treatment of acute promyelocytic leukemia with arsenic trioxide--an analysis of 242 cases].

OBJECTIVE: To observe the effectiveness of arsenic trioxide (As(2)O(3)) in the treatment of acute promyelocytic leukemia (APL). METHODS: Data of clinical observation, seven year follow-up survey were analysed with American SAS system software, concentration of arsenic in cerebrospinal fluid (CSF) was determined by atomic absorbing spectrum. RESULTS: The complete remission (CR) and effectiveness rates were 87.9% and 94.4% for the previously untreated group; 60.0% and 65.0% for the relapsed in As(2)O(3) maintenance treatment group; 69.5% and 84.8% for the relapsed in other than As(2)O(3) maintenance treatment group; and 48.7% and 94.1% for the refractory group, respectively. In the previously untreated group, the relapse rate at 3 years after CR was 26.7%. The probability of survival in 136 follow-up survey patients was 92.0% and 76.7% at 5 years and 7 years, respectively. There were mild side effects. Central nervous system leukemia (CNSL) occurred in 138 out of 412 cases in 6 years, including some refractory leukemia, extramedullary residual leukemia and extramedullary relapse. Prophylactic intrathecal injection could reduce the incidence rate of CNSL. Determination of arsenic concentration indicated that it was difficult for arsenic to penetrate the blood-brain barrier. CONCLUSION: The CR rate and long-term survival are high and the relapse rate is low in APL patients treated with As(2)O(3). There is no cross-resistance between ATRA and As(2)O(3) and other chemotherapeutic agents. As(2)O(3)-resistance occurs scarcely. It can also be used in maintenance treatment.

[Umbilical cord blood stem cells transplantation from an unrelated donor into an adult with severe aplastic anemia].

OBJECTIVE: To evaluate the availability of the treatment of adult severe aplastic anemia with unrelated allo-cord blood stem cell transplantation (CBSCT). METHODS: HLA-matched unrelated cord blood transplantation has been successfully performed for an adult severe aplastic anemia patient. A unit of cord blood provided by Guangzhou Cord Blood Bank containing 1.89 x 10(7)/kg mononucleated cells, 1.8 x 10(4)/kg CFU-GM and of CD(34) positive cells was 0.009. The patient was conditioned with CTX (60 mg/kg) and anti-lymphocyte globulin (ALG, 120 mg/kg). GVHD prophylaxis consisted of both MTX and CsA. The CsA had been given for 8 months. RESULTS: The lowest ANC was 0.6 x 10(9)/L post-transplantation. The patient achieved an ANC of greater than 0.5 x 10(9)/L at 10 days, and the platelet of greater than 50.0 x 10(9)/L at day 20 after transplantation. He developed Grade 1 GVHD in the tenth month after grafting. Microsatellite DNA fingerprinting indicated a stable donor-recipient mixed chimerism, whilst the circulating red cells remained host origin. CONCLUSION: It is the first report in China on successful treatment of adult severe aplastic anemia by unrelated allo-CBSCT.

[Regulatory effect of Erbao granules on brain-gut peptide in juvenile animal model of anorexia].

OBJECTIVE: To study the regulatory effect of Erbao granules (EBG) on central and peripheral brain-gut peptide in juvenile animal model of anorexia. METHODS: Juvenile rat model of anorexia was established by imitating the major cause of infantile anorexia and treated with EBG. The cholocystokinin-octapeptide (CCK-8) and beta-endorphin (beta-EP) concentration in hypothalamus, antrum pyloricum and peripheral blood were examined by radioimmunoassay. RESULTS: CCK-8 concentration in hypothalamus and plasma in the model rats increased (P < 0.05), while blood beta-EP concentration decreased (P < 0.05). After EBG treatment, the CCK-8 concentration normalized and beta-EP increased significantly. CONCLUSION: EBG could reduce the central and peripheral CCK-8 and increase beta-EP secretion significantly in the juvenile anorexia model.

[Clinical study on effect of combined treatment with shenlong oral liquid and radiotherapy in treating nasopharyngeal carcinoma].

OBJECTIVE: To observe the effect of Shenlong Oral Liquid (SLOL) combined with radiotherapy in treating nasopharyngeal carcinoma (NPC). METHODS: Effects of the combination therapy, including clinical effect, changes of cellular immunity and side effects, in treating 60 NPC patients (treated group) were observed and compared with those of the other 60 patients treated with radiotherapy alone (control group). RESULTS: (1) The side effect of radiation in the treated group was lower than that in the control group significantly (P < 0.05); (2) The immediate regression rate of nasopharyngeal and neck tumor in the two groups was not different significantly (P > 0.05); (3) The doses for complete remission of nasopharyngeal and neck tumor in the treated group was lower than that in the control group (P < 0.01); (4) No change of T-lymphocyte subsets was found in the treated group after treatment, but in the control group, OKT3, OKT4 and OKT4/OKT8 ratio were markedly decreased (P < 0.05); (5) The survival rate in the treated group was higher than that in the control group, but without statistical significance (P = 0.0518). CONCLUSION: The combination therapy of NPC by SLOL and radiotherapy is able to reduce side effect of radiotherapy, improve the cellular immunity, reduce the dose of radiation for tumor remission and enhance the therapeutic effect of radiotherapy, it might raise the survival rate of NPC patients.

[17beta -estradiol inhibits carotid sinus baroreflex in male rats].

By perfusing isolated carotid sinus, the effect of 17beta-estradiol (E(2)) on carotid sinus baroreflex was observed in anesthetized male rats. The results obtained are as follows. (1) By perfusing with E(2) (10 micromol/L), the functional curve of baroreflex was shifted to the right and upward, with a peak slope (PS) decrease from 0.49+/-0.03 to 0.25+/-0.01 (P<0.01) and a reflex decrease in mean arterial pressure (reflex decrease, RD) from 7.37+/-0.42 kPa to 3.49+/-0.20 kPa (P<0.001), while the threshold pressure (TP) and saturation pressure (SP) were significantly increased from 9.52+/-0.68 kPa to 13.3+/-0.11 kPa (P<0.001) and 24.53+/-0.48 kPa to 27.52+/-0.20 kPa (P<0.01) respectively. Among the functional parameters of carotid baroreflex, the changes of RD, PS, TP and SP were dose-dependent. (2) Pretreatment with different doses of tamoxifen (1, 5, 10, 30 micromol/L), an inhibitor of estrogen receptor, did not block the effect of E(2) on carotid baroreflex. (3) Preperfusion with an inhibitor of NO synthase L-NAME (100 micromol/L) could completely abolish the effect of E(2) on carotid baroreflex. It is concluded that the inhibitory effect of E(2)on carotid sinus baroreflex may be mediated by NO release from endothelial cells, but not by a genomic mechanism.

[Effects of intracerebroventricular injection of angiotensin II on blood pressure and discharges of habenula nucleus cardiovascular neurons].

The effects of icv and iontophoretic application of angiotensin II (A II) and Sar(1),Thr(8) -A II (ST-A II) on blood pressure (BP) and electroneurogram (ENG) of habenula nucleus (Hb) cardiovascular neurons were studied in stress induced hypertensive (SIH) and normotensive rats. Increase and decrease of BP could be induced respectively by icv of A II (20 microgram) and ST A II (20 microgram), to a greater extent in SIH than in the normotensive group. Icv A II also increased the ENG of MHb cardiovascular neurons in both the groups, but the increase was significantly greater in SIH group than in the normotensive one. Iontophoretic application of A II increased the ENG of Hb cardiovascular neurons in both the groups and the increase was also significantly greater in SIH than in the normotensive group. On the other hand, iontophoretic application of ST-A II inhibited the ENG of Hb cardiovascular neurons in the two groups and the inhibition was markedly greater in SIH than in the normotensive rats. It is suggested that central A II is involved in SIH and Hb cardiovascular neurons of SIH show an enhanced sensitivity to A II.

[Regulation effect of curcumin on blood lipids and antioxidation in hyperlipidemia rats].

The regulation effects of curcumin on lowering blood lipids and antioxidation were studied in the hyperlipoidemia model Wistar rats fed with high fat diet for 4 weeks. The hyperlipoidemia model rats were then separated into four groups: normal diet group, normal diet + curcumin (5 g/kg) group, high-fat diet group and high-fat diet + curcumin group. The rats were fed with experimental diets for another 4 weeks respectively. The results indicated that curcumin decreased the serum levels of cholesterol and triglyceride and increased the serum level of apolipoprotein A. Curcumin decreased TBA value and improved total antioxidation capability. The activities of SOD and GSH-PX in liver were enhanced. After taking normal diet for four weeks, the blood fat and cholesterol levels of the hyperlipoidemia rats recovered. But in the normal diet + curcumin group, the same results could be found in two weeks.

[Effect of exposure to low concentration of benzene and its analogues on luteal function of female workers].

The effect of benzene and its analogues(BIA) on the luteal function of female workers was studied in a petrochemical corporation. The female workers from a chemical fiber corporation without benzene exposure were selected as controls. The levels of BIA(toluene and xylene) in the air at breathing zone were sampled and determined with gas chromatography(GC). The menstrual function was followed up. Urine pregnanediol-3-glucuronide (PdG) and follicular stimulating hormone(FSH) were measured by enzyme immunoassay(EIA). The results showed that BIA existed in low concentration in air. The exposure could lead to a shorter length of time of luteal phase(P < 0.05 or P < 0.01) and decrease luteal progesterone level (P < 0.05). It was suggested that the low concentration of BIA exposure could have effect on the luteal function in exposed female workers, and urinary hormones could be used as biomarker for the influences of benzene and BIA on reproduction.

[Effect of chromium gluconate on body weight, serum leptin and insulin in rats].

In order to observe the effect of chromium on rat body weight, serum leptin and insulin, eight groups of rats were randomly fed diets (basal diet or high-fat diet) with different levels of chromium gluconate [proximally Cr(III) 1,2 and 3 mg/kg BW]. All rats were weighted once a week. By the end of the 8th week, all rats were decapitalized and the levels of serum leptin and insulin were tested by RIA method. The ratio of liver, spleen, kidney and testickle to body weight were calculated. The average body weight and the levels of leptin and insulin of high-fat-diet groups were significantly higher than those of the basal-diet group (P < 0.05), but the insulin and leptin of groups with chromium were significantly lower than their corresponding control groups without chromium(P < 0.05). It was concluded that chromium could cause weight loss slightly and reduce the levels of insulin and leptin.

[An experimental study on the effect of positional asphyxia on cardiac and pulmonary functions].

The purpose of the study is to explore the effect of positional asphyxia on cardiac and pulmonary functions and its role in causing death. respiration, pulmonary artery pressure and cardiac output were measured by inserting the swan-ganz into the pulmonary artery from the femoral vein. Results showed that: (1) the respiration was stable before hanging, getting faster during hanging, and becoming irregular at the terminal stage of hanging; (2) the pulmonary artery pressure increased significantly during and at the terminal stage of hanging as compared with that before hanging; (3) the cardiac output was stable before and during hanging, but decreased significantly at the terminal stage of hanging. It is concluded that hanging may cause respiratory imbalance and hypoxia, resulting in cardiac and pulmonary dysfunction. The death caused by hanging belongs to respiratory death, the characteristic of which is primary respiratory arrest followed by secondary cardiac arrest.

[In vitro antiviral activity of antisense oligonucleotides against influenza virus].

For developing of antisense oligonucleotides as potential antiviral therapeutic agents against influenza A virus, phosphorothioate oligodeoxynucleotides(PS-ODN) targeted to part to the 3' and 5' end sequences which are common to the eight RNAs of type A influenza virus were synthesized. The in vitro cytotoxicity of these PS-ODNs was assayed and then antiviral activity of these PS-ODNs was evaluated by using cultured MDCK cells infected by A/JingFang/86-1(H1N1). It was found that(1) No in vitro cytotoxicity was seen when concentration of PS-ODN was up to 50 mumol/L. (2) Oligonucleotides(IV4 #) complementary to 5' terminal conserved sequences and ODN(IV6 #, IV7 #)composed by 3'/5' terminal conserved sense sequences or antisense sequences all exhibited the most potent antiviral activity. IV4 # ODN exhibited significant antiviral activity with reduced hemegglutination titer about 50% at concentration of 1 mumol/L, and over 70% at 10 mumol/L. The antiviral activity of IV4 # was in a dose- and sequence-dependent manner. (3) IV4 # ODN showed significant antiviral activity not only against H1N1 subtype strains of influenza virus, but also against H3N2 subtype strains. (4) The multiplicity of infection(MOI) of influenza virus had effects on antiviral activity of IV4 # ODN. IV4 # ODN showed better dose-dependent antiviral activity when there was lower MOI. These results suggested the the 3'/5' terminal sequences of influenza A virus could be used as a target for designing antiviral oligonucleotides against influenza A virus.

[Genetical analysis on cross incubation of Beauveria bassiana].

Heterokaryon of Beauveria bassiana was formed during the cross incubation of two vegetative compatible strains with genetic markers of actidione resistance and 34 degrees C tolerance. The chromosome(s) or its fragment successive losses, recombination and segreation led to haploidization during the conidia formation period. After at least 4 generations of parasexual cycle, the genetic character of heterokayon could get to relative stable. Genetic marker and RAPD analysis indicated that the combinants showed the phenomenon of preferential selection of one parental type by unrandom chromosome lossing and the gene of the other parental strain was suppressed or lost completely. Different culture medium, in vivo or in vitro, and different mixture ratio of original strain spores could affect parasexual process and then the preferential selection. The results also demonstrated the heterosis effect of cross culture.

[Study on the plasmid instability of Bdellovibrio BDG-9].

A new method system was established in this paper to study the plasmid in stability of Bdellovibrio BDG-9. Using this system, it was found that when BDG-9 was cultured singly on the SMB plate, plasmid pST I was unstable although pST I still replicated and distributed to progeny cell normally. The results showed that the pST I copy number in single cell of BDG-9 decreased gradually to zero with the propagation of BDG-9. Additionally, plasmid pST I was very important for the growth of BDG-9, and with the lacking of pST I s, the growth and propagation of BDG-9 ceased.
