Estimation of the mutagenic potential of the trematode Opisthorchis felineus in experimentally infected guinea pigs.

We studied the mutagenic potential of the trematode Opisthorchis felineus in experimentally infected guinea pigs by chromosome analysis. The investigation was conducted on 30 treated (experimentally infected) and 20 normal (uninfected) guinea pigs (Cavia porcellus). A sample of living metacercariae of O. felineus was obtained from the muscle of naturally infected crucian carp captured in the Tom River, and 1 ml suspension containing approximately 50 metacercariae in 0.9% NaCl solution was individually given p.o. to guinea pigs that were used as treated specimens, whereas normal guinea pigs from the same stock were given 1 ml 0.9% NaCl solution per individual to serve as controls. The treated and control animals were killed by CO2 asphyxiation and decapitation at 7, 15, 30, 45, and 120 days after contamination. The smears of bone marrow cells for chromosome analysis were prepared by a standard method. Statistical analysis of the data was done using Student's t-test. We found that experimental infection of guinea pigs with O. felineus induced significantly high frequencies of hypoploid cells and, at the 120th day p.i., led to a considerable incidence of chromatid breaks and polyploidy in bone marrow cells.

[An experimental and clinical validation of a method for treating acute inflammatory diseases of the adnexa uteri by using the electrophoresis of a therapeutic mud preparation].

Experimental and clinical investigations have demonstrated that the addition of abdominal-sacral electrophoresis of esobel 1% solution to combined therapy of acute inflammation of the uterine appendages improves treatment results. Bioantioxidants bring about the antiinflammatory action of the mud preparation esobel while its high-polar lipids prevent structural-metabolic impairment of the appendages' tissue elements as shown by a pronounced antiexudative effects and weak secondary alteration.

[Pathomorphological features of proliferative vitreoretinopathy in intravitreal administration of blood mononuclear cells (experimental study)].

The role of blood mononuclears in the pathogenesis of proliferative vitreoretinopathy (PVR) was studied on 25 Wistar rats (200-250 g). Intravitreal injections of 0.05 ml normal saline containing mononuclears (3000/mm@3) into one eye and 0.05 ml normal saline into the other were made through the flat part of the ciliary body under ether narcosis. Mononuclears were isolated using the ficoll-hypaque gradient. Indirect ophthalmoscopy was carried out on days 1, 3, 5, 7, 14, and 21 after injection. After each ophthalmoscopy (except day 1) 5 animals were sacrificed and both eyes were removed. The material was fixed for optic and electron microscopy. Ophthalmoscopic and histological studies showed the development of preretinal adhesions, the main sign of PVR. Bearing in mind the unidirectional flow of liquid in ocular tissues, created by the pressure gradient, the authors suggest that blood mononuclears are involved in the development of vitreoretinal proliferation.

[Experimental validation of a treatment of hypertrophic and keloid postoperative scars with peloidoultraphonophoresis].

Experiments on 107 rats have been made to validate use of ultraphonophoresis of 1% oil extract of polar lipids from silt sulphide muds for treatment of pathological skin scars. Peloidoultraphonophoresis was found able to transform the scars into regenerates of the skin type characterized by the presence of hair, sebaceous glands, elastic fibers, complete recovery of epidermis. The above peloidotherapy increased the number of vessels, tissue basophils in the scar and surrounding skin but decreased the number and severity of pathological alterations in nervous fibers.

[Microwaves and the visual analyzer].

The results of studying an effect of microwaves on the different regions of visual analyzer are investigated. The morphofunctional changes at the light-optic and electron-microscopic levels are revealed. The changes involve not only peripheric area of visual analyzer but also its central regions. Some epidemiologic observations are indicative of: altered sizes of blind spot; decreased magnitudes of optic rheobase and chronaxy; color vision disorder; increased diastolic pressure in central artery of the retina. Study of bioelectrical activity of the retina enabled some investigators to make a conclusion about a reaction disorders occurring in the subcortical visual centers. It is noted that the animal studies not always explain unequivocally the microwave effects on the visual analyzer.

[Effect of peloid therapy on morphofunctional changes of ovaries and oviducts caused by suturing (experimental study)].

A course of ultraphonophoresis of the extract of high-polar lipids of sulfide mud attenuated inflammation of uterine appendages in rats caused by catgut suturing of the ovaries and oviducts. On postoperative day 75-120 peloid therapy resulted in degradation of the suture material and its replacement with connective tissue. The peloid limited postoperative intensification of follicular atresia, raised content of growing follicles and yellow bodies in the ovaries.

[Electrophoresis of sulphide mud extract in combined therapy of chronic inflammatory diseases of uterine appendages (an experimental and clinical study)].

On the original experimental model of chronic inflammation of the ovaries and oviducts in white rats it was established that electrophoresis of 1% esobel solution prevents an increase in the specific volume of interstitial connective tissue, development of fibrosclerotic alterations and adhesive process. Peloidophysiotherapy normalizes growth and atresia of the ovarian follicular system. Thus, electrophoresis of 1% esobel solution is highly effective in combined treatment of chronic inflammatory diseases of the uterine appendages.

[Effects of combined influence of ionizing radiation and light on the retina].

The continuous irradiation of rats with a light (3500 1x) for 48 hours led to destruction of 7.5 +/- 0.43% of neurosensoric cells (the control value was 0.2 +/- (0.02%). Longer photodamage caused progressing decrease in a population of the cells, narrowing followed by disappearance of corresponding layers of retina by the 30th day of the experiment. Preliminary exposure to X-rays with a dose of 5 Gy modified light damages of retina. The combination of two factors caused death of more than 30% of neurosensoric cells and redused the number of rows of nucleis of the external nuclear layer twice for the first 48 hours of illumination. After a longer influence of light a preliminary ionizing irradiation increased the number of destructively changed neurosensoric cells, reaching minimun of 41.7% by 30th day of the experiment. At the same time after a long combined irradiation less changes in row number quantity, numerical density of nucleus of the external nuclear layer and a population of neurosesoric cells in general was observed than after only one light exposure. Changes of choriocapillaries (thrombosis, obliteration, narrowing of a gap) which were more expressed at a combination of factors, led to infringement of blood circulation and played the important role in mechanisms of retina degeneration. Radial glia and neurons of internal layers of retina showed relative resistance to the investigated influences.

[Cerebroprotector activity of Rhaponticum carthamoides extract in rats with brain ischemia].

Electron microscopy and electrophysiological data reveal pronounced structural and functional disturbances in cerebral cortex of rats with model brain ischemia. A 5-day treatment with a dry extract from Rhaponticum carthamoides (Willd.) Iljin. in a daily dose of 150 mg/kg (p.o.) equivalent to 2.7 mg ecdysterone per kg body weight reduced manifestations of the ischemic damage, improved the structure of neurons, and restored brain activity (EEG parameters).

[Plasticity of neuroendocrine transducers under the combined influence of the radiation and light exposure].

The structural changes of neurons of the rat hypothalamic supraoptic (SON) and of paraventricular (PVN) nucleus after 48 h of bright light exposure, of 5 Gy whole-body X-irradiation and of their combination subjected to the analysis by means of light-optic and of electron microscopy for the estimation of radimodificated effect of light exposure lasted 24 h a day and plasticity of neuroendocrine transducers interacted with the optic sensory system. The structural changes of neurons of the SON after combined action are less considerable and more prolonged in comparison with the PVN that loas defermined by their direct connection with the optic sensory system via the retinohypothalamic tract.

[The damage of the neural retina and of the retinal pigment epithelium of eyes of rats exposed to highly intensive visible light under alloxanic diabetes].

The experiments on white mongrel rats showed that the light with the intensity of 6000 1x lasting for 6 hours resulted the destruction of the retinal pigment epithelium, of the internal and the external neurosensory cellular processes with subsequent nuclei piknosis and radial glia replacement. On the 7th day after exposure to light focal effusion of the retina corresponding layers. Under the irradiation in diabetes the amount of cells with karyopyknosis increases in 1.6 times in comparision with the findings in case free of diabetes. Was found, that the alloxanic diabetes in its early stage increases the light impairment of the neurosensory cells of the retina.

[Early ultrastructural reactions in various parts of the visual analyzer in guinea pigs after thermogenic microwave irradiation].

A single total radiation of guinea pigs by means of microwaves of thermogenic intensity (energy density stream 60 mVt/sm2) and exposition for 10 min produces ultrastructural reactions in various parts of the visual analyser, differing in their manifestation degree. The most essential alterative changes have been revealed in the retina, appearing in 6 h after radiation, as degeneration of the membrane disks of the photoreceptors and as enhancement of phagocytic activity of pigmentocytes, and immediately after the radiation--as destructive disturbances of mitochondria in radial glyocytes and as reaction of the synaptic apparatus. In the cerebral visual centers a higher reactivity of neurons of the external geniculate bodies than in the visual cortex is noted, but ultrastructural disturbances of the hematoencephalic barrier and synapses of the visual cortex are more manifested. Early ultrastructural changes in the optic nerve are least manifested.

[Quantitative histologic changes of the glioneuronal complex in the central and intermediate parts of the visual analyzer exposed to microwaves of thermogenic intensity].

Whole-body exposure of guinea pigs to microwaves of 60 mW/cm2 for 10 min causes quantitative changes in glial population of outer geniculate bodies and visual cortex that are manifested by the enlarged perineuronal glia, an increased glioneuronal index, and a relatively stable perivascular glia. The dynamics of the changes observed indicates higher reactivity and repair capacity of the glioneuronal complex of outer geniculate bodies.

[The reaction of glia in visual centers during the whole body effect of combined microwaves and x-rays].

A single whole-body preirradiation with thermogenic microwaves modifies the dynamics of the glial reactions of visual centers of guinea pigs induced by median lethal X-radiation doses. A combination of the two factors produces a synergistic effect, estimated by the degree of alteration of astrocytes and oligodendrocytes at early times after exposure, leads to early activation of microglia, and reduces radiation-induced alterations in glia at later times (25-60 days).

[Dynamics of the morpho-quantitative changes in the central and intermediate regions of the visual analyzer during whole body x-irradiation].

Morpho-quantitative estimation of changes in IV and V layers of visual cortex and outer geniculate bodies of the brain of guinea pigs subjected to whole-body X-irradiation with a median lethal doses shows that neurons from the visual analyzer sections under study are differently damaged. The morphogenesis of changes is connected with the original variations in the quantitative structure of a neuron-glial complex and a selective alterative and productive reaction of the perivascular glia.
