We have developed a 2-dimensional gel method for identification of RNA sequences crosslinked by the intercalative drug 4'-hydroxymethyl-4,5',8-trimethylpsoralen (HMT). This method is being used to localize such sites in E. coli ribosomal RNA. We report here the identification of a site for HMT crosslinking within positions 434 and 497 of 16S rRNA. We suggest a likely site for HMT intercalation, in which residues U548 and U473 become crosslinked via the drug.
An histochemical technique combining the retrograde transport of horseradish peroxidase (HRP) and catecholamine histofluorescence was used to identify those neurons of the cat's mesencephalic ventromedial tegmentum (VMT) which project to visual cortex. In a region corresponding to the ventral tegmental area-A10 nucleus we identified a group of fluorescent cells which were retrogradely labeled by HRP injections into visual cortex. It is suggested that these cells belong to a hitherto unknown, posterior part of the mesocortical dopaminergic system.

A significant factor affecting the chance for recovery is the ability of the skid row alcoholic to form and sustain a relationship with someone or something. The author describes an open-ended, community residential program that encourages such relationships and which is proving to be an effective treatment model.
Eight newborn babies were tested to see whether they could make discriminating headturn responses to the sound of a 9-second recording of a human voice coming from 15 degrees, 30 degrees and 80 degrees from the midline, from either the right or left side. The babies showed significant differences in response to sounds coming from the right and the left, and to sounds coming from 15 degrees and 80 degrees from the midline. Pethidine ('Meperidine') given to the mother during labour had a significant effect, both on degree of response and on the babies' apparent ability to localize sound.

Most simian virus 40 (SV40)-transformed BALB/c 3T3 clones employed for biochemical studies have been used without regard to passage level. To determine whether virus-induced properties are stable as a function of passage, we have extensively characterized one transformed clone, FNE, which was isolated after SV40 infection BALB/c 3T3 cells in factor-free medium. From the initial testing at passage 5 and for at least 50 subsequent subcultures, the cells stably maintained many transformed growth properties, including high saturation density, morphology, colony formation on contact-inhibited monolayers, tumorigenicity, and synthesis of viral-specific RNA. However, other properties varied as a function of passage. There was a slight decrease in viral genome equivalents per cell from 1.1 copy/cell at passage 5 to 0.7 copies at passage 40. Initially, the cells were negative for all type C virus; however, cells carried at low density for 13 to 20 passages (65 to 100 generations) began to release an endogenous type C virus that then persisted in the culture. Spontaneous release of type C virus did not occur in control BALB/c 3T3 cells carried under identical culture conditions for 90 passages. When the cultures were releasing type C viruses they stained uniformly and brightly positive for SV40 tumor (T) antigen by immunofluorescence, whereas T antigen staining was variable at early passage. These experiments suggest that subtle but perhaps important differences in viral gene expression can occur as a function of passage; they also demonstrate the importance of evaluating the interactions between SV40 and endogenous type C viruses.
The role of radiotherapy in the management of rectal and rectosigmoid cancer is still far from clear. Our experience with 98 patients is presented: eight patients were treated with radical radiotherapy, 13 received postoperative radiation, and 67 were treated for palliative purposes only (bleeding, pain, mass, and fistulation). Satisfactory results were demonstrated in all three categories, and sterilization of disease by radiation was histologically proven in three cases. The necessity of high-dose radiation for the achievement of optimum results is demonstrated, and the techniques and problems are described.

Tadpoles of Xenopus laevis reared in water containing 0-01% propylthiouracil continue to grow but fail to develop or metamorphose. The haemoglobin of such tadpoles has been extracted in buffer, converted to a cyanmet form, and run on polyacrylamide gels. The developmentally retarded tadpoles are found to possess adult-type haemoglobin rather than the tadpole type which normally characterizes their developmental stage.

Clinical trials were undertaken to determine whether lucanthone (miracil D) affects radiation-induced regression in measurable pulmonary metastases and advanced squamous-cell oral and pharyngeal tumors. The time required for 50% tumor regression was decreased by approximately 50% in those patients who received lucanthone in addition to irradiation. These results indicate that lucanthone has a definite adjuvant effect when used together with irradiation.
