[Surgical treatment of metastatic lung cancer from colorectal cancers].

Based on conclusions obtained after the observation of 61 colorectal cancer patients with a lung metastasis, the resection of lung metastasis as a therapy was evaluated. Among these 61 patients, only 5 patients had been identified as having a lung metastasis at the time of resection of the primary lesion, whereas the other 56 patients developed the lung metastasis after the curative resection of the colorectal cancer. Only one patient with a synchronous lung metastasis and twelve patients (eleven with a solitary metastatic lesion and one with multiple metastatic lesions) with metachronous lung metastasis underwent removal of the lung metastasis. The three-year survival rate was 65.2% in the metachronous group.

Inhibition of alkaline phosphatase activity and D-glucose uptake in rat renal brush-border membrane vesicles by aminoglycosides.

The binding of aminoglycoside antibiotics to, and their effects on, the plasma membrane were studied using isolated rat renal brush-border membrane vesicles. Dibekacin was noted to bind with brush-border membrane vesicles having a single class of many binding sites. 3H-labeled dibekacin binding was inhibited competitively by unlabeled dibekacin, gentamicin or amikacin. The inhibition constants obtained from the Dixon plots followed the order of gentamicin approximately equal to dibekacin greater than amikacin. The alkaline phosphatase activity of brush-border membrane vesicles was inhibited by gentamicin significantly, as was also observed by a histochemical study. Sodium-dependent D-glucose uptake by brush-border membrane vesicles was significantly inhibited by the addition of gentamicin.

Predefined gene transfer for expression of a glycosphingolipid antigen by transfection with a cosmid genomic library prepared from a cell line in which the specific glycosphingolipid is highly expressed.

The deliberate transfer of globotriaosylceramide (Gb3) expression in mouse lymphoma L5178 cells was achieved by transfection with a cosmid DNA library prepared from human Burkitt lymphoma Ramos cells in which Gb3 was highly expressed. The recipient mouse lymphoma cells did not contain Gb3 but did contain its direct precursor, lactosylceramide. The transfected cells expressed Gb3, detected both chemically and immunologically, and contained human DNA detected by an Alu sequence probe. This model demonstrates a general method for studying glycosyltransferase genes and other factors necessary for the expression of glycosphingolipid antigens.

Production of metalloproteinase in gastric tissue with acetic acid-induced ulcer or taurocholic acid-induced gastritis.

A metalloproteinase was extracted from a medium of cultured gastric tissues following acetic acid-induced ulcer or taurocholic acid-induced gastritis, and the enzyme was partially purified by Sephadex G-200 column chromatography. This proteinase was released in active and latent forms with apparent molecular weights of 170,000 and 210,000, respectively. Its activity was abolished by treatment with ethylenediaminetetraacetic acid or 1,10-phenanthroline, but not with phenylmethylsulfonyl fluoride or soybean trypsin inhibitor. This enzyme degraded type I gelatin and type IV collagen but did not attack type I collagen. In contrast, very little of this enzyme was released from gastric tissue following erosion caused by water-immersion stress or from normal gastric tissue. These results suggest that this metalloproteinase plays an important role in gastric lesions by breaking down the basement membrane.

Establishment of five human malignant non-T lymphoid cell lines and mixed lymphocyte-tumor reaction.

Five human malignant non-T lymphoid cell lines have been established. THP-4 and THP-7 were derived from two children with common type acute lymphoblastic leukemia (ALL), THP-6 and THP-8 from two children with null cell type ALL and THP-9 from a child with malignant lymphoma. THP-4 was positive for cytoplasmic mu, B4, common ALL antigen (CALLA) and Ia-like antigen (Ia), THP-7 positive for B4, CALLA and Ia, THP-8 positive for B4 and Ia, and THP-9 positive for surface mu-lambda, B1, B4, CALLA and Ia. No antigenic determinants for monoclonal antibodies used were detected on THP-6. THP-4, THP-7, THP-8 and THP-9 were able to stimulate allogeneic lymphocytes vigorously. THP-6 without Ia stimulated not only allogeneic lymphocytes, but also autologous ones. THP-7 did not stimulate autologous lymphocytes.

[Relationship between biopsy and prognosis of breast cancer].

Clinical data from 131 patients who underwent primary radical operation for breast cancer at the Second Department of Surgery, Shinshu University Hospital during five-years from January 1976 to the end of December 1980 were analyzed to investigate for the relationship between biopsy and prognosis. The incisional biopsy group had a significant higher recurrence rate as compared with the no biopsy group, cytology group and the cancer negative group after excisional biopsy. Although the puncture aspiration cytology group did not show significant difference in recurrence rate, an any more than 3 weeks of interval between puncture aspiration cytology and radical surgery associated with a significant high recurrence rate. This suggests that the interval also exerts an unfavorable influence upon prognosis. Puncture aspiration cytology was recognized to be apt to result in distant metastasis than any other biopsy methods and also it is tended to have a higher recurrence rate in scirrhous and mucinous carcinoma of the breast.

Retinopathy of prematurity (1) birth weight, gestational age and maximum PaCO2.

The eyes of neonates who were premature or had been exposed to increased ambient oxygen from May 1979 to February 1985 were examined. Twenty out of 256 infants were found to have retinopathy of prematurity (ROP) of stage 3 or worse in at least one eye. Using uivariate statistics (chi-square analysis or the Wilcoxon test), the variables representing the maximum arterial carbon dioxide tension (PaCO2), birth weight and gestational age were found to be significantly associated with ROP. It is recommended to maintain PaCO2 values under 50 mmHg, especially in less mature newborns (less than 1,300g in birth weight or 210 days of gestational age).

Uptake of aminoglycoside antibiotics into brush-border membrane vesicles and inhibition of (Na+ + K+)-ATPase activity of basolateral membrane.

The effects of aminoglycoside antibiotics on plasma membranes were studied using rat renal basolateral and brush-border membrane vesicles. 3',4'-Dideoxykanamycin was bound to the basolateral membrane and brush-border membrane vesicles. They had a single class of binding sites with nearly the same constant, and the basolateral membrane vesicles had more binding sites than those of the brush-border membrane. Dideoxykanamycin B was transported into the intravesicular space of brush-border membrane vesicles, but not into that of basolateral membrane vesicles. The (Na+ + K+)-ATPase activity of the plasma membrane fraction prepared from the kidney of rat administered with dideoxykanamycin B intravenously decreased significantly. Aminoglycoside antibiotics entrapped in the basolateral membrane vesicles inhibited (Na+ + K+)-ATPase activity, but those added to the basolateral membrane vesicles externally failed to do so. The activity of (Na+ + K+)-ATPase was non-competitively inhibited by gentamicin. It is thus concluded that aminoglycoside antibiotics are taken up into the renal proximal tubular cells across the brush-border membrane and inhibit the (Na+ + K+)-ATPase activity of basolateral membrane. This inhibition may possibly disrupt the balance of cellular electrolytes, leading to a cellular dysfunction, and consequently to the development of aminoglycoside antibiotics' nephrotoxicity.

Binding of 3',4'-dideoxykanamycin B to ATP and its related compounds.

The interactions of aminoglycoside, 3',4'-dideoxykanamycin B(DKB) with ATP and its related compounds were investigated. ATP, ADP, cyclic AMP and FAD bound to the DKB-conjugated Sepharose 4B column. The binding of DKB to ATP was also confirmed by equilibrium gel filtration. In the acidic pH region, the fluorescence of nucleotides was quenched by DKB. The Stern-Volmer plots showed that the molar ratios of the complexes were 1:1. The apparent stability constant was dependent on the number of the phosphate groups of nucleotides and was in the order of ATP greater than ADP greater than AMP.

Electron microscopic study of the developing human vitreous collagen fibrils.

Vitreous fibrils, and collagen fibrils in other ocular tissues, were observed in 5-, 6- and 7-weeks human embryos. The vitreous fibrils in 7-weeks embryos were distinctly different from the fibrils of other ocular tissues. The vitreous fibrils near the rim of the optic cup were similar to the fibrils in the anterior cavity of the lens vesicle in the 6-week embryo. Broad fibrils were demonstrated with a diameter of 30-40 nm and a striated banding pattern of about 60 nm periodicity.

Adaptation of a human monocytic leukemia cell line (THP-1) in a protein-free chemically defined medium.

We have adapted a human monocytic leukemia cell line [THP-1c12(+)] so that it can proliferate in a completely protein-free chemically defined medium of an equal mixture of Dulbecco's modified Eagle's minimum essential medium and Ham's F12. This cell line was designated as THP-1c12(-). When a sufficient number of THP-1c12(-) cells was seeded in culture, the maximum cell density after 6 days of culture was 1 X 10(6)/ml with a doubling time of 30 hr. Transferrin showed a slight stimulative effect on the growth of THP-1c12(-) cells, but insulin did not. The surface profile of THP-1c12(-) was the same as that of THP-1c12(+), which possessed Mol. Mo5, LeuM3, My9 and Ia-like antigens as well as a large number of Fc receptors. Phagocytic ability with respect to IgG-coated sheep red blood cells was evident in both THP-1c12(+) and THP-1c12(-) cells. A small percentage of THP-1c12(-) cells retained the ability to reduce nitroblue tetrazolium when stimulated with 12-tetradecanoyl-phorbol-13-acetate. Culture supernatant from THP-1c12(-) stimulated the incorporation of 3H-thymidine into its own cells, suggesting an autocrine mechanism by which the growth of THP-1c12(-) cells is induced in a protein-free medium.
