Characterization of functional beta-adrenoceptor subtypes in rabbit urinary bladder smooth muscle.

Spontaneous contractile force of muscle strips isolated from male rabbit urinary bladder dome (detrusor) and base (trigonal muscle) was significantly inhibited by isoproterenol (10(-7)-10(-5) M), a non-specific beta-adrenoceptor agonist or by terbutaline (10(-8)-10(-5) M), a selective beta 2-adrenoceptor agonist. The EC50 values for isoproterenol and terbutaline in detrusor were the same as those in trigonal muscle but the maximum relaxant response to isoproterenol or terbutaline was significantly greater in detrusor than in trigonal muscle. Dobutamine (10(-5)-10(-4) M), a relatively specific beta 1-adrenoceptor agonist caused a small but significant relaxant response in trigonal muscle but no change in detrusor. In trigonal muscle the relaxant response to dobutamine was less than that to terbutaline. The relaxant response to 10(-6) M isoproterenol in detrusor was completely blocked by butoxamine (10(-4) M), a selective beta 2-antagonist or by propranolol (10(-6) M), a non-specific beta-antagonist but not by metoprolol (10(-6)-10(-4) M), a selective beta 1-antagonist. Relaxation of trigonal muscle induced by 10(-6) M isoproterenol was inhibited by 10(-5) M metoprolol by 30%, by 10(-4) M butoxamine by 70%, or completely by 10(-6) M propranolol. These findings are consistent with the view that the density of beta-adrenoceptors is higher in the detrusor than in trigonal muscle, and that the relaxant response to beta-adrenoceptor stimulation is mediated by beta 2-subtype in the detrusor and by both of beta 1- and beta 2-subtypes in trigonal muscle of the male rabbit.

Serum Erythropoietin levels and inhibitors of erythropoiesis in patients with chronic renal failure.

The role of various factors in erythropoiesis was studied in 13 predialysis patients and 41 hemodialysis patients. Serum erythropoietin (ESF) was measured by the fetal mouse liver cell bioassay in vitro. The effect of uremic sera on heme synthesis and erythroid progenitor cell (CFU-E) formation was examined using normal human bone marrow cultures. Serum ESF levels in both predialysis (99.9 +/- 45.0 mU/ml) and dialysis (141.2 +/- 109.7 mU/ml) patients were significantly higher than those in normal controls (42.0 +/- 25.8 mU/ml), although the titers were not sufficiently increased to correct the anemia. Serum ESF concentrations did not correlate with hemoglobin level or inhibition of both heme synthesis and CFU-E formation. Bone marrow CFU-Es in uremic patients were normal in the number, the responsiveness to ESF and the percentage in S-phase. Significant decrease in heme synthesis was observed in dialysis patients. The degree of inhibition of CFU-E formation showed a relationship to hemoglobin levels in uremic patients. By the CFU-E formation assay, the difference in inhibitory effects of the sera obtained from dialysis patients immediately before and after a hemodialysis was significant only under a low ESF concentration (0.125 U/ml) but not under a high concentration (1.0 U/ml). In conclusion, inhibition of heme synthesis and CFU-E formation, in addition to inadequate ESF production, plays an important role in renal anemia.

Comparison of the peptide and saccharide moieties of gamma-glutamyltransferase isolated from neoplastic and non-neoplastic human liver tissue.

gamma-Glutamyltransferase from human hepatoma and the surrounding non-neoplastic liver tissue was purified by immunoaffinity column chromatography and characterized with regard to molecular weight, isoelectric point (pI), amino acid composition, hexosamine content and affinity for various lectins. Both enzymes showed the same molecular weight (the heavy subunit 64 000; the light subunit 26 000) and pIs (3.7-3.9) on SDS-polyacrylamide gel electrophoresis and isoelectric focusing in polyacrylamide gel, respectively. After neuraminidase treatment, the pIs for both enzymes shifted to a more alkaline pH (pI 5.7). Both enzyme preparations exhibited similar amino acid compositions; however, the glucosamine content of the hepatoma enzyme was 362 nmol/mg protein, about 3-fold higher than that of the enzyme isolated protein from the non-neoplastic tissue. Binding of the two enzymes to lectins revealed that less of the hepatoma enzyme bound to Sepharose-conjugated wheat germ agglutinin, erythroagglutinating phytohemagglutinin and Ricinus communis agglutinin. These results suggest that the two enzymes possess similar peptide moieties and degree of sialylation, but differ with respect to other aspects of their heterosaccharide moieties.

An immunologic test for fecal occult blood by counter immunoelectrophoresis. Higher sensitivity and higher positive reactions in colorectal cancer than single radial immunodiffusion and hemoccult test.

A new immunologic fecal occult blood test has been developed employing counter immunoelectrophoresis (CIEP) with anti-human hemoglobin Ao antiserum (anti-HbAo). Minimum detectable hemoglobin in hemolysate was 7, 9, and 7 micrograms/ml by CIEP, single radial immunodiffusion (SRID), and Hemoccult test (SmithKline Beckman), respectively. The values in stool were 0.5, 1.0, 3.0 mg hemoglobin/g stool in the same order. CIEP was more sensitive than SRID in detecting hemoglobin both in hemolysate and in feces. Of 38 stools from the patients with colorectal cancer, 29 (76%) gave positive tests in CIEP, whereas 22 (58%) (P less than 0.005), and 18 (47%) (P less than 0.05) gave positive tests in SRID and Hemoccult test, respectively. In every stage and at every site of cancer, CIEP showed the highest positive tests. CIEP did not give any false-positive reaction in specimens from 20 patients without gastrointestinal lesion or from 34 asymptomatic healthy controls without diet restriction, whereas Hemoccult test gave one and three positive reactions in these groups. These results clearly indicate that CIEP is specific and more sensitive to human hemoglobin than any other previous methods. CIEP is expected to have a potential for mass screening of colorectal cancer.

Phenotype and gene frequencies of acid phosphatase (s-AcP) in the human parotid saliva.

Genetic polymorphism of the human parotid salivary acid phosphatase (s-AcP) in the Japanese population is described. The use of polyacrylamide gel isoelectric focusing electrophoresis with the pH range of 4.0-6.5 enabled us to discern three variant patterns controlled by two codominant alleles at the single autosomal locus. The two alleles were designated s-AcP:A and s-AcP:a, and the gene frequencies calculated from 183 Japanese subjects were s-AcP:A = 0.2268 +/- 0.022, s-AcP:a = 0.7732 +/- 0.022, respectively. The distribution of phenotypes fitted the Hardy-Weinberg equilibrium.

Identification of dipeptidyl peptidase IV as a protein shared by the plasma membrane of hepatocytes and liver biomatrix.

The histotypic organization of liver parenchyma involves specific intercellular contacts and interaction of hepatocytes with supporting biomatrix. Evidence from this laboratory identified a peptide (Hep105, apparent Mr 105 000) that is shared by the plasma membrane of rat hepatocytes and rat liver biomatrix. This report identifies Hep105 as a peptide component of dipeptidyl peptidase IV (DPPIV; EC 3.4.14.-). A monoclonal antibody (MAb 236.3) was shown to immunoprecipitate DPPIV from non-ionic detergent extracts of surface-labeled 125I hepatocytes. The immunoprecipitate contained two 125I-labeled peptides: Hep105 and a peptide of apparent Mr 150000 (Hep150). Proteolysis of 125I-labeled Hep105 and Hep150 by Staphylococcus aureus V8 protease yielded essentially identical patterns of 125I-labeled peptide degradation products, indicating that Hep105 and Hep150 are structurally related. Only Hep150 exhibited DPPIV activity on transblot analysis, an observation that is consistent with the interpretation that it is the monomeric form of the enzyme. Heating (100 degrees C, 5 min) of purified Hep150 in the presence of sodium dodecylsulfate (SDS) resulted in its conversion to Hep105 and the disappearance of any demonstrable enzymatic activity. 3H-labeled diisopropyl fluorophosphate was incorporated into Hep105, indicating that Hep105 contains the active site for DPPIV. Purified rat liver biomatrix was shown to possess significant DPPIV activity. Taken together, these data indicate that Hep105 s a peptide component of DPPIV.

Microwave surgical treatment of diseases of prostate.

A new transurethral probe for microwave radiation of the prostate has been developed. As a preliminary experiment, sliced ham was radiated with microwaves using this probe in order to evaluate the extent of thermal effect. Using mongrel male dogs, microwave coagulation of the prostate was examined. These animal experiments showed marked destruction of the prostate gland. Furthermore, the safety of this method was confirmed on the basis of results from the experiments. Prostatic bladder neck obstruction also has been treated in 6 patients with benign prostatic hypertrophy or carcinoma of the prostate by this technique. There has been no mortality and also no complications. The results of this preliminary clinical trial have been excellent.

Role of the sympathetic nerve in bladder and urethral sphincter function during the micturition cycle in the dog evaluated by pressure flow EMG study.

The role of the sympathetic nerve on bladder and urethral sphincter function during the whole micturition cycle, including the collecting and emptying phases, was evaluated on 10 decerebrated dogs by pressure flow EMG study. A series of experiments was performed before and after hypogastric nerve transection. In the control condition and after hypogastric nerve transection, reflex micturition with bladder contraction and spasmodic rhythmic sphincter contractions occurred. Urodynamic parameters of the micturition cycle were statistically compared between control and nerve transection experiments. Threshold volume, threshold pressure and opening pressure showed a small but significant change after nerve transection. A decrease in voided volume seems to be secondarily accompanied with a decrease in threshold volume. It seems that adrenergic nerves play a certain role in bladder function during the collecting phase of the micturition cycle.

Effects of autonomic drugs on in vivo recording of electromyograms of canine renal pelvis and ureter.

The effects of the autonomic drugs (noradrenaline, isoproterenol and acetylcholine) and the urine volume change on the pacemaker of ureteral peristalsis were studied by our new method of the in vivo recording of electromyograms (EMGs) of canine renal pelvis. The EMGs of the pelvicalyceal region showed a two phasic slow-rising potential of amplitude of 20 microV and discharge interval of 5 s and was different from propagated waves and therefore considered to be the pacemaker potentials. In the diuretic state the pacemaker potentials kept a constant discharge interval. On the contrary, the discharge interval of the ureter EMG became shorter and finally corresponded to the pacemaker EMG one to one. These results suggest that the urine transport in the diuretic state is controlled by the change of efficiency of the peristaltic propagation and not by the changes of the discharge of the pacemaker itself. Noradrenaline promoted the ureteral peristalsis and isoproterenol inhibited the ureteral peristalsis. These two drugs had no effects on the pacemaker potentials. Acetylcholine had a great variety of the effects on the ureter EMG and the pacemaker EMG separately. These results suggest that the pacemaker of the ureteral peristalsis is controlled under the influence of the parasympathetic system and the ureter is controlled under the influence of both the sympathetic and parasympathetic system.

Effects of the autonomic antagonists on canine urethral responses to autonomic nerve stimulation.

The effects of the autonomic antagonists on urethral activity was studied in female dogs. Urethral responses produced by autonomic nerve stimulation were measured with pressure and motility changes in control conditions and after administration of autonomic antagonists. Phentolamine and guanethidine reduced both hypogastric and pelvic nerve mediated responses. It seems that urethral smooth muscle function is greatly influenced by adrenergic drugs.

Effects of autonomic agonists on in vivo female canine urethral motility.

The response of the urethra to intra-arterially administered autonomic agonists was studied in vivo by measuring the motility and pressure changes in female dogs. Phenylephrine produced shortening along the transverse axis with an increase in urethral pressure in a dose-dependent manner. Isoproterenol produced lengthening along the transverse axis with a decrease in urethral pressure. Both acetylcholine and dimethylphenylpiperazinium did not produce consistent patterns in urethral response. It seems that adrenergic drugs act predominantly on the circular muscle layer and cholinergic drugs act equally on both the circular and longitudinal muscle layers.
