Glucocorticoid receptors and terminal deoxynucleotidyl transferase activities in leukemic cells.

Glucocorticoid (GC) receptor and terminal deoxynucleotidyl transferase (TdT) activities were studied in leukemia cells to investigate their diagnostic and therapeutic implications. Among cell lines with T-cell character, higher GC-receptor and TdT activities were found in T-ALL (HPB-ALL and ALL-Ichikawa) than in cells from adult pleomorphic T-cell leukemia (HPB-MLT). HPB-Null with pre-B cell-character exhibited moderate GC receptor but low TdT activity; Raji cells and CCRF-SB, derived from B-cell Burkitt lymphoma and B-ALL, respectively, manifested low GC receptor and no TdT activity. The highest GC receptor activity was demonstrated in null-cell ALL, followed, in order, by juvenile T-ALL, adult pleomorphic T-cell leukemia, and AML. Other kinds of lymphoid and monocytic leukemias exhibited low GC receptor and no TdT activity. Although low GC receptor and negative TdT were demonstrated in cells from seven out of nine patients under CML blastic crisis, the last patient had cells with positive TdT and GC receptor activity.

Specific cytosol binding proteins for 25(OH)D3 and 1.25(OH)2D3 in human breast cancers.

Binding proteins for 1.25 (OH) 2D3 were investigated in thirty breast cancers. Human breast cancer was shown to contain specific, high affinity cytosol binding proteins for 1.25 (OH) 2D3 and 25 (OH) D3. The binding protein for 1.25 (OH) 2D3 sedimented at 3.7 S and the binding protein for 25 (OH) D3 at about 6.0 S on sucrose density gradient analysis containing 0.3 M KCl and 1 mM dithiothreitol in buffer. Kd for 1.25 (OH) 2D3 were from 0.1 x 10(-11) M to 7.1 x 10(-11) M measured by Scatchard plots. Competition binding studies indicated that the relative specificity of the binding protein for 1.25 (OH) 2D3 much greater than 25 (OH) D3 greater than 1 alpha (OH) D3, 24,25 (OH)2D3 greater than D3 much greater than Estradiol-17 beta. 1.25 (OH) 2D3 receptor-positive was detected in twenty-eight out of thirty breast cancers.

Radioimmunoassay for 11-deoxycortisol using iodine-labeled tracer.

A simple and sensitive radioimmunoassay for 11-deoxycortisol was developed. The antiserum produced in rabbits by immunizing with a complex of 11-deoxycortisol-3-oxime and bovine serum albumin (BSA) has little cross-reactivity with other endogenous steroids. The immunoassay procedure requires only one-step ethanol denaturation of binding proteins in plasma and extraction by an organic solvent can be omitted. Furthermore, use of 125I-labeled tracer significantly simplify the counting procedure. The method is sensitive enough to detect 1 microng/100 ml of 11-deoxycortisol. Plasma 11-deoxycortisol levels measured by this method after the administration of a single dose of metyrapone ranged from 5.0 to 19.2 microng/100 ml, whereas they were 0 to 4.0 microng/100 ml in hypopituitary patients. It is concluded that this simple method is useful for the routine assay of plasma 11-deoxycortisol as a parameter of the metyrapone tests.

A case of alcaptonuria with fatal cardiovascular disturbance.

A case of alcaptonuria combined with aortic insufficiency was found in a 28-year-old male. The patient was palpitating at admission. The daily excretion of homogentisic acid was 2.0-6.0 g. Electrocardiography indicated atrial fibrillation and left ventricular hypertrophy with a ST-T change and right axis deviation. Cartilage tissues in the knee-joints showed no pigmentation. Vertebral X-ray revealed no calcification. The patient's history disclosed a family intermarriage in his grandparents. The patient's mother noticed the presence of black stains on diapers in his infancy and brown pigmentation on the skin and sclera in childhood. No kin had similar symptoms.

An extremely basic monoclonal IgG in an aged apoplectic patient with prolonged bacterial infection.

A case with prolonged bacterial infection accompanied by an abnormal serum protein which migrated in the post-gamma region on electrophoresis is presented. The abnormal protein was identified as IgG with gamma-type light chain moiety. The patient suffered from prolonged pneumonia and cholecystitis, Bone marrow aspiration and skeletal x-rays did not indicate multiple myeloma.
