Basal lamina formation by epidermal cells in cell culture.

The formation of basal lamina in epidermal cell cultures was studied by electron microscopy. Epidermal cells were cultured on plastic dishes which were either uncoated or coated with collagen gel. The cells grew and stratified on the plastic substratum as well as on the collagen gel. While the cultures on the plastic surface showed no occurrence of hemidesmosomes and basal lamina at the interface with the plastic substratum, the cultures on the surface of the collagen gel showed development of hemidesmosomes leading to the focal occurrence of basal lamina at the interface of the collagen gel and the basal plasma membrane of basal cells. Thereafter, basal lamina occurred under the interhemidesmosomal areas in addition to the hemidesmosomal areas of the basal plasma membrane, forming a continuous layer by joining.

Selective localization and growth of Bifidobacterium bifidum in mouse tumors following intravenous administration.

A strain of domestic bacteria, Bifidobacterium bifidum (Lac B), which is nonpathogenic and anerobic, selectively localized and proliferated in several types of mouse tumors following i.v. administration. None of the same bacilli could be detected in the tissues of healthy organs such as the liver, spleen, kidney, lung, blood, bone marrow, and muscle 48 or 96 hr after i.v. administration into tumor-bearing mice. Proliferation of Lac B in the tumor was artifically stimulated by i.p. administration of DDD-H-2s mice of a synthesized disaccharide, lactulose (4-O-beta-D-galactopyranosyl-D-fructofuranose), a sugar which is not metabolized by mammalian tissue cells. Lac B, which survices and proliferates selectively in the tumor following i.v. administration into the tumor-bearing host, should aid in diagnosis and selective therapy for cancer.

True histiocytic lymphoma.

An 86-year-old man had true histiocytic lymphoma of the skin. Diagnosis was established by the following findings: (1) typical histological features, (2) lack of receptors on tumor cells for immunological surface markers, and (3) active phagocytic function. Electron microscopic studies indicated cytoplasmic multivesicular bodies in the tumor cells. Multivesicular bodies have been reported in histiocytic proliferative disorders and may be of diagnostic value in cases of true histiocytic lymphoma.

Potentiation of anticancer effect of carboquone in vivo by glucose pretreatment.

Anticancer effect of carboquone was notably potentiated in tumor-bearing mice when they were pretreated with 5% glucose solution in the system of blood flow-interrupting hyperthermic chemotherapy. It was further found by quantitative analysis that the potentiation was much greater than increase in the general toxicity of the drug by administration of glucose. Mitomycin-C, on the other hand, revealed no such marked effect though it is considerably similar to carboquone in chemical structure.

Hepatic mitochondrial cytochrome P-450: isolation and functional characterization.

A CO-binding heme protein was solubilized and partially purified from the inner membrane fraction of rat liver mitochondria by a modification of a method [Imai, Y. & Sato, R. (1974) Biochem. Biophys. Res. Commun. 60, 8-14] developed to purify cytochrome P-450 from liver microsomes. The partially purified preparation contained protoheme and its spectral properties are characteristic of the heme proteins of the cytochrome P-450 family. The isolated cytochrome P-450 preparation could reconstitute a CO-sensitive, NADPH-dependent 26-hydroxylation activity for 5beta-cholestane-3alpha,7alpha,-12alpha-triol when supplemented with NADPH-adrenodoxin reductase and adrenodoxin, both purified from bovine adrenocortical mitochondria. Unlike a cytochrome P-450 purified from liver microsomes of drug-untreated rats, however, the liver mitochondrial cytochrome P-450 could not catalyze NADPH-dependent benzphetamine N-demethylation in the presence of adrenodoxin reductase and adrenodoxin or function with the purified microsomal NADPH-cytochrome c reductase plus Emulgen 913 as an electron-donating system. It is concluded that the rat liver inner mitochondrial membrane houses a species of cytochrome P-450 functional in 5beta-cholestane-3alpha,7alpha,12alpha-triol 26-hydroxylation.
