Structural studies on the carbohydrate portion of human antithrombin III.

Human antithrombin III has been shown to contain four identical N-glycosidically linked carbohydrate chain per molecule. These carbohydrate chains have been investigated by sugar and methylation analysis before and after removal of N-acetylneuraminic acid residues. The chains have been further investigated by Smith degradation, trifluoroacetolytic degradation, and degradation after chromium trioxide oxidation. As a result of these studies, the following structures is proposed for the carbohydrate chains in human antithrombin III: (formula: see text). NeuNAc, N-acetylneuraminic acid; Galp, galactopyranose; GlcNAcp, 2-acetamido-2-deoxyglucopyranose; Manp, mannopyranose are the abbreviations used in the structure.

A new method for degradation of the protein part of glycoproteins: isolation of the carbohydrate chains of asialofetuin.

A new method has been developed for degrading the protein part of several glycoproteins, whilst leaving the carbohydrate portion virtually intact apart from partial degradation at the reducing end. The method is based upon stabilization of the glycosidic linkages of the sugar residues by trifluoroacetyl groups and subsequent cleavage of the peptide bonds by transamidation. The two reactions are carried out in a mixture of trifluoroacetic anhydride and trifluoroacetic acid. After O- and N-detrifluoroacetylation, the carbohydrate portion can be isolated and re-N-acetylated. The applicability of the method is demonstrated by the isolation from asialofetuin of the carbohydrate chains that are attached by N- and O-glycosyl links.

Effect of oral zinc intake on delta-aminolaevulinic acid dehydratase in red blood cells.

Earlier results regarding the in vitro and in vivo effects of zinc on delta-aminolaevulinic acid dehydratase (ALAD) activity in red blood cells were confirmed in healthy human subjects after oral intake of zinc for 12 weeks. In one group of seven healthy adults, oral intake of zinc for 12 weeks. In one group of seven healthy adults, oral intake of zinc, as the sulphate salt (2.07 mmol zinc/day) for 6 weeks, resulted in a 44% increase in the activity of ALAD in erythrocytes. Plasma zinc levels also increased during the experimental period and reached a maximum of 29 mumol after 6 weeks and remained constant thereafter. In another group, the intake of zinc in a lower dose, (0.69 mmol zinc/day) for 12 weeks, showed a similar tendency, although the increase in the enzyme activity, as well as the plasma zinc levels, was relatively much less. The plasma copper levels decreased significantly in the second group after the zinc intake and reached a low value of 11 mumol at the end of the experimental period. The Cu:Zn ratio also decreased considerably towore valuable indicator of zinc status than plasma zinc levels alone.

Immunochemistry of Salmonella O-antigens. Specificity and cross-reactivity of factor O9 serum and of antibodies against tyvelose (Formula: see text) mannose coupled to bovine serum albumin.

The octasaccharide Galp (Formula: see text) Rhap, the synthesized disaccharides methyl 3-O-a-tyvelopyranosyl-a-D-mannopyranoside, methyl 3-O-a-tyvelopyranosyl-beta-D-mannopyranoside and methyl alpha-tyveloside, in order of decreasing effectiveness, inhibited the precipitation of S. typhi T2 alkali-treated lipopolysaccharide by O-factor 9 serum. On a molar basis the relative inhibiting activities of the glycosides were by O-factor 9 serum. On a molar basis the relative inhibiting activities of the glycosides were 100:22:8:2. With rabbit aniserum raised against 3-O-a-tyvelopyranosyl-D-mannopyranosyl covalently linked to bovine serum albumin the relative inhibitory activities of the four glycosides were 11:100:26:3. These data establish that the 3-O-a-tyvelopyranosyl-a-D-mannopyranosyl structure is immunodominant in the Salmonella O-antigen 9. The specificity of the antigen-antibody interaction was high: glycosides in which the tyvelose (3,6-dideoxy-D-arabino-hexose) residue had been replaced by abequose (3,6-dideoxy-D-xylo-hexose) or paratose (3,6-dideoxy-K-ribo-hexose), had less than one fiftieth of the activity of the most active inhibitor in either of the two precipitation systems used. Moreover, the results show that 3-O-a-tyvelopyranosyl-D-mannopyranosyl coupled to bovine serum albumin elicits O-antibodies of higher specificity than those obtained by absorption of antibacterial immune serum.

Glucose-containing oligosaccharides in the urine of patients with glycogen storage disease type II and type III.

Patients with glycogen storage disease type II and type III were recently found to excrete increased amounts of a glucose-containing tetrasaccharide DGlcp(alpha1 leads to 6)DGlcp(alpha1 leads to 4)DGlcp(alpha1 leads to 4)DGlc [Lennartson, G., Lundblad, A., Sjöblad, S., Svensson, S. and Ockerman, P.A. (1976) Biomed. Mass Spectrom. 3, 51--54]. In addition to this tetrasaccharide, urine from these patients also contains larger oligosaccharides containing only glucose. From urine of patients with glycogen storage disease type II and type III, three and four oligosaccharides respectively have been isolated. Structural studies including sugar analyses, methylation analyses, partial acid hydrolysis and optical rotation revealed that three compounds were present in the urine of both patients. Their proposed structures or partial structures are as follows: DGlcp(alpha1--6)DGlcp(alpha1--6)DGlcp(alpha1--4)DGlcp(alpha1--4)DGlcp(alpha1--4)DGlc, DGlcp(alpha1--4)DGlcp(alpha1--6)DGlcp(alpha1--6)DGlcp(alpha1--4)DGlcp(alpha1--4)DGlc, and DGlcp(alpha1--6)DGlcp(alpha1--4)DGlcp(alpha1--4)DGlcp(alpha1--4)DGlcp(alpha1--6)DGlcp(alpha1--4)DGlcp(alpha1--4)DGlc. A fourth compound has been partially characterized as a branched heptasaccharide with four (1 leads to 4) linkages and two (1 lead to 6) linkages. Glycogen is possibly the origin of these compounds. However, the number of (1 leads to 6) linkages is higher than expected and may indicate a shorter distance between branches in glycogen than has been generally assumed.

Urinary abnormalities in fucosidosis. Characterization of a disaccharide and two glycoasparagines.

The urinary excretion of fucose-containing material was found to be highly increased in a patient with fucosidosis type 2. Three structurally related compounds, a disaccharide and two glycoasparagines, were isolated from the urine. The isolation procedure included ultrafiltration, gel chromatography on Sephadex G-25, preparative zone electrophoresis and paper chromatography. From structural studies including optical rotation, sugar analysis, methylation analysis, ninhydrin degradation, reduction with lithium aluminium hydride and partial hydrolysis, the following structures were deduced: formula (see text), where Fucp is fucopyranose, Manp is mannopyranose, Galcp is galactopyranose, GlcNAcp is 2-acetamido-2-deoxyglucopyranose and Asn is asparagine. The yields of these compounds were 1.7, 40, and 6 mg/l, respectively. The origin of the disaccharide and the two glycoasparagines is probably the core region of glycoprotein carbohydrate chains.

Zinc and copper concentrations in serum, blood and liver in moderate experimental pancreatic insufficiency.

Serum and liver concentrations of zinc and copper were studied in the rat after ligation of the pancreatic duct. Ligation of the bile duct was done close to the duodenum and high up in the hilum of the liver. The bile flow was by-passed into the intestine via a plastic catheter. The surgical method used was found suitable for a study of moderate exocrine pancreatic insufficiency since significant--although markedly reduced--concentrations of amylase and trypsin were estimated in intestinal contents at sacrifice 19 weeks after the operation. The serum zinc level was unaffected by duct ligation except for a slight decrease after 2 weeks. Similarly zinc in whole blood was unaffected. The copper concentration in serum was, however, significantly enhanced during in first 10 weeks. The copper zinc ration was markedly increased in duct ligated rats during the whole period. Zinc and copper concentrations in liver tissue were significantly decreased after 19 weeks. The findings suggest that a decrease function of exocrine pancreas might induce a disturbed balance of zinc and copper in the rat.

Effect of phenylhydrazine on delta-aminolevulinic acid dehydratase in red blood cells.

Delta-aminolevulinic acid dehydratase activity was measured in the blood of rats poisoned by successive injections of phenylhydrazine (1 ml, 35 mmol). After 5 days of successive injections, more than 90% of the circulating blood cells consisted of reticulocytes and the enzyme activity also increased successively and reached a maximum corresponding to the number of reticulocytes. Thus delta-aminolevulinic acid dehydratase activity measured in the peripheral blood is mainly due to the percentage of circulating reticulocytes.
