Evidence for active immunity to morphine in mice.

The serum from mice actively immunized with a morphine immunogen contained antibodies that could bind dihydromorphine. Morphine effects were diminished in these "immunized" mice, and the concentration of morphine in their plasma was altered.

Analgesia

Resistance of noradrenaline in blood vessels to depletion by 6-hydroxydopamine or immunosympathectomy.

1. The degree of the decrease in the noradrenaline concentrations caused by 6-hydroxydopamine or immunosympathectomy was different in different areas of the cardiovascular system.2. In rats or guinea-pigs 6-hydroxydopamine depleted the noradrenaline content of the heart by 90%, of the mesenteric vein by 80% and of the mesenteric artery and aorta by 30-60%. Immunosympathectomy elicited a 70% reduction in the cardiac noradrenaline but only a 50% reduction in the noradrenaline of the blood vessels of the rat.3. The tyrosine hydroxylase activity of the heart, blood vessels, or adrenal glands was not significantly altered 2 weeks after 6-hydroxydopamine. Nor was the monoamine oxidase activity in heart or blood vessels changed.4. The inconsistent ability of both 6-hydroxydopamine and immunosympathectomy to abolish experimental hypertension may be due to the partial persistence of noradrenaline and functional sympathetic nervous system activity in the blood vessels.

Effects of sodium intake on inherited hypertension in the rat.

The effect of sodium chloride intake on systolic blood-pressure was studied in genetic hypertensive Wistar rats. In these animals hypertension can develop on a sodium-"free" diet, but the height to which the blood-pressure rises is related to the sodium intake; increasing the amount of potassium in the diet promotes sodium excretion and ameliorates the hypertension. It is suggested that this form of inherited hypertension may be determined by two different sets of autosomal alleles only one of which is sodium-dependent.

Barbiturates: radioimmunoassay.

The development of a radioimmunoassay for barbiturate is described. The barbiturate is made antigenic by coupling it to a protein, bovine gamma globulin. The radioimmunoassay can measure as little as 5 nanograms of barbiturate.

Animals

Disposition of morphine in man.

The disposition of morphine was investigated by means of radioimmunoassay after a single intravenous dose (10 milligrams per 70 kilograms) was administered to 10 adult normal male subjects who had not received other drugs for 2 weeks preceding the study. A multiphasic decline in serum concentrations of morphine occurred. Detectable blood concentrations of morphine, or of a metabolite, or of both persisted for 48 hours after a single intravenous dose.
