Light scattering changes of edetic acid-treated, washed rat platelets: effect of adenosine diphosphate and calcium ion in relation to chilling.

The effe-ts of adenosine diphosphate (ADP) and Ca2+ on platelet morphology were studied in edetic acid (EDTA)-treated, washed platelets of rat by means of light scattering measurement at right angles. ADP caused a transient change of light scattering, which was independent of concentration above 5 muM and of pH between 4 and 10. When the platelets were pretreated with ADP or chilled persistently, Ca2+ also caused another transient change of light scattering. The effect of Ca2+ was abolsihed at pH below 6.5. The velocity of both ADP- and Ca2+-induced response was dependnet on temperature, and the magnitude was reduced by metabolic inhibitors and SH-blocking agents. Adenosine triphosphate (ATP) competitively inhibited the ADP-induced response, but not the Ca2+-induced one. The response to Ca2+ of the control and chilled platelets was compared by scanning electron microgram. Apparent similarity of the effect of chilling on membrane structure to that of ADP was suggested.

Potent alpha-adrenoceptor action of soterenol on vascular and other smooth muscle.

Treatment with soterenol (0.3-300 mug/kg, i.v.) increased the mean arterial blood pressure and decreased heart rate in rabbits. Soterenol caused a dose-dependent contraction of rabbit aorta and portal vein, guinea-pig vas deferens and cat nicitating membrane which was inhibited by phentolamine. Other beta-agonists such as isoproterenol, salbutamol and terbutaline did not exhibit similar effects. Soterenol had potent stimulant action on the alpha-receptor of the smooth muscle.

Calcium flux and binding in the aortic smooth muscle from the spontaneously hypertensive rat.

There was no significant difference in the tissue calcium content between spontaneously hypertensive rat (SHR) and normotensive rat aortae. 45Ca uptake was significantly greater, whereas 45Ca influx was significantly less in SHR than in control aortae. There was no difference in 45Ca efflux in SHR and control tissues. 45Ca influx in both SHR and control tissues was increased by potassium, not increased by norepinephrine and inhibited by Co++, Mn++ and La+++ (all 0.5 mM). Both SHR and control aortae were equally inhibited by Mn++ and Co++ (0.1 mM). There was no difference in the 45Ca uptake by microsome fraction from both SHR and control tissue. These data indicate some differences in the translocation of Ca++ at the cellular level in SHR and control aortae.

The epidemiological study on the correlation between serum lipids and cerebro-cardiovascular disease.

1. There were significant correlation between hyperlipidemia and obesity, hypertension, abnormal ECG and abnormal eyeground. 2. The incidence of cerebral hemorrhage was closely associated with hypertension but not with hyperlipidemia. 3. It was proved that hypertension with both hyper-Ch and hyper-TG was highly related to the development of cerebral infarction. 4. It seemed that the incidence of myocardial infarction and angina pectoris was related to hypertension with hyper-Ch. 5. Therefore, the present study suggested that the control of hypercholesterolemia and hypertriglyceridemia was useful for the prevention of cerebro-cardiovascular disease.

Coil planet centrifugation and its application to the observation of altered membrane properties of erythrocytes in hepatobiliary disorders.

A coil planet centrifuge (CPC) is an apparatus aiming to obtain a centrifugal force exerting on particles suspended in a fluid in a coiled polyethylene tube (coil) which makes self-rotation around its axis and revolution around the principal axis simultaneously. If a linear osmotic gradient is prepared in the coil and a small amount of blood is applied to it and subjected to centrifugation, one is able to observe the membrane properties of erythrocytes in terms of the shift of hemolysis band appearing in the coil with high accuracy and precision. Coil planet centrifugation was applied to the observation of altered membrane properties of erythrocytes in hepatobiliary diseases. Remarkable shifts of end-point of the hemolysis band going in parallel with the severity of damage were visualized. Study as to its cause revealed that increased free cholesterol content of erythrocyte membrane which was the result of plasma cholesterol Esterstürz due to inhibition of lecithin-cholesterol acyltransferase by stagnant bile acids was responsible for this phenomenon. Elevation of serum bile acid concentration and increased serum leucine-aminopeptidase were closely correlated to the shift of hemolysis end-point toward the low osmolal side. CPC may be useful for detection and chasing the clinical course of patients with hepatobiliary disorders.

[Bilateral giant carotid aneurysm in the cavernous sinus with a carotid-cavernous fistula on one side, case report (author's transl)].

Since Sir Gilbert Blane's report of a case in the year 1800, bilateral aneurysms of the internal carotid artery within the cavernous sinus of nontraumatic origin are extremely rare. In reviewing literatures, only 22 cases have been described, including the present case (Table 1). We are reporting our own case in which carotid angiograms revealed the presence of bilateral giant aneurysms with a carotid-cavernous fistula on one side, and discussed briefly with reference to its clinical features, radiological observations and treatment. (case report) M.R, a woman aged 78, was admitted on May 15, 1973, with a complaint of sudden onset of right supra- and periorbital headache and a loud bruit about 2 months previously. She became aware of ptosis of the right eyelid and prominence of the right eyeball. The carotid arteriogram showed bilateral giant aneurysms in the region of the cavernous sinus (Fig 2, Fig. 3). A carotid cavernous fistula due to the ruptured giant aneurysm was seen on the right and poor filling of the right anterior and middle cerebral artery was also observed (Fig. 2). An attempt was made to close the fistula by using propelling method (polyurethane foam emboli after Ohta's technique) with great success (Fig. 4). No special treatment was done to the left giant aneurysm. She is now in good condition 7 months after discharge.
