A novel human cultured cell line, P39/Tsugane, was established from leukemic cells in the peripheral blood of a 69-year-old male with overt leukemia following myelodysplastic syndrome (MDS). P39/Tsugane cells were characterized by blastic appearance, presence of NaF-sensitive alpha-naphthyl butylate esterase activity, Fc gamma-receptor, C3-receptor, capacity to phagocytize sensitized erythrocytes, and reactivity with monoclonal antibodies such as OKT4, My4, VIMD5, MCS-2 and My7. These data indicate that P39/Tsugane cells are of myelomonocytoid nature. P39/Tsugane had a hypodiploid chromosome constitution with a gain of a consistent marker, 6q+, the presence of less consistent markers 9q+ and rcp(14;16), and random and non-random losses of autosomes: in accordance with the reported cytogenetic profiles of MDS, a representative karyotype of the present cell line is 45,XY,+del(6)(q15),9q+, t(14;16)-(q24;q21),-16,-17. P39/Tsugane cells were transplantable intraperitoneally into nude mice, and produced abdominal tumors and hemorrhagic ascites. These results indicate that P39/Tsugane is the first cultured cell line of myelomonocytoid nature to be derived from overt leukemia following MDS. Therefore, P39/Tsugane cells should be useful for studies on the differentiation of leukemia cells, the pathogenesis of MDS and in vitro-in vivo experimental chemotherapy.
Six non-glucose polyols--mannose, fructose, 1-deoxyglucose, mannitol, glucitol, and inositol--were identified and evaluated in human serum and cerebrospinal fluid by gas-liquid chromatography and by gas-liquid chromatography/mass spectrometry. Concentrations of fructose, mannose, and inositol in the serum of healthy persons or children without metabolic diseases varied with age, as already reported for 1-deoxyglucose. Fructose, inositol, and glucitol concentrations in cerebrospinal fluid significantly exceeded those in serum. The method described here for determining polyols and for evaluating polyol patterns in serum, as well as the resulting data on children and healthy subjects, should be useful in investigations of the clinical and physiological significance of polyols.

Nitrite reductase from Clostridium perfringens was purified by chromatographies on DEAE-cellulose, DEAE-Sephadex, Sephadex G-150, and hydroxylapatite and by isoelectric focussing to a homogeneous state, showing essentially a single protein band in disc gel electrophoresis and a single immuno-precipitation line in double diffusion against antiserum obtained from immunized rabbits. The reductase was induced in the presence of nitrate. It had a molecular weight of 54,000 and showed no absorption peak in the visible region. The pH optimum was 6.2 and Km for nitrite was 5 mM. Ferredoxin, as well as viologen dyes, was found to be an electron donor. The product of nitrite reduction was hydroxylamine. This reductase was inhibited by o-phenanthroline and azide but not by cyanide or diethyldithiocarbamate.
Repair polymerases participating in unscheduled DNA synthesis in isolated liver nuclei, bleomycin-treated permeable cells and in ultraviolet-irradiated living cells were studied using two specific inhibitors of DNA polymerases, aphidicolin and 2', 3'-dideoxythymidine-5'-triphosphate. Unscheduled, i.e., repair, DNA synthesis in rat liver nuclei, and in bleomycin-treated permeable SR-C3H/He and XC cells was mostly attributed to DNA polymerase beta. Unscheduled DNA synthesis in human liver nuclei, bleomycin-treated permeable HeLa and HEp-2 cells, and in ultraviolet-irradiated HeLa, HEp-2 and XC cells was partially inhibited by the polymerase alpha-specific inhibitor, aphidicolin. The results suggested that both DNA polymerase alpha and beta participated in unscheduled DNA synthesis, though the respective degrees of participation differed depending on cell type and the nature and degree of DNA damage.
Gas exchange was measured in eight patients undergoing high-frequency ventilation (HFV) during intrathoracic operations for lung cancer. HFV facilitated the operation because the exposed lung moved only slightly to ventilate and stayed in a less expanded state. Intermittent positive-pressure breathing (IPPB) (control) was switched to HFV 20 minutes after the pleural cavity was opened. HFV either was conducted alone (HFV alone) or was superimposed on a hypoventilation equal to the dead space (HFV on VD). The driving pressure and frequency of HFV were set at 0.5 kg/cm2 using 3, 6, and 12 Hz with an FI02 of 0.50. The only statistically significant difference in arterial PO2 between IPPB and HFV occurred when the HFV alone was used at 12 Hz. Significant differences in arterial PCO2 values existed between IPPB and HFV alone when the HFV was at 6 and 12 Hz, and between HFV alone and HFV on VD at 12 Hz. The arterial pH values were lower at 6 and 12 Hz in HFV alone and were significantly different from pH values during IPPB. Acidosis was due to the respiratory component of HFV only. The metabolic component remained unchanged. It was concluded that the gas exchange was adequately maintained at 3 Hz during HFV alone. The intrathoracic operation was facilitated by less movement as the frequency increased, although the lung volume tended to expand. The gas exchange, particularly the elimination of carbon dioxide, worsened during the use of HFV alone. However, HFV on VD continued to maintain an adequate gas exchange and facilitate the operation.
Twenty liver cancer patients, including 9 hepatocellular carcinoma and 11 with metastatic liver cancer, were treated by intra-arteral one-shot chemotherapy. Alterations in blood coagulation and fibrinolysis were observed serially after one-shot chemotherapy by testing the levels of PT, APTT, FDP, fibrinogen, AT III, alpha 2-macroglobulin, and plasminogen. APTT was prolonged, FDP increased, Fbg increased after a transicent decrease, and AT III, alpha 2-M, and plasminogen decreased. The peaks of these alterations occurred within 7 days after the one-shot treatment; recovery was observed after about weeks. The more advanced the cancer, the greater were the alterations.

Applicability of secondary ion mass spectrometry (SIMS) for structural analysis of oligoribonucleotides was studied. For this purpose a number of oligoribonucleotides having various base sequences, chain lengths, and internucleotide linkages were prepared, and their positive ion SIMS spectra were recorded without derivatization. The soft ionization produced ions possessing relatively low internal energies which underwent little fragmentation, so that both pseudo-molecular and sequence (fragment) ions were observed, the former being more abundant than the latter. Nevertheless, sufficient information can be obtained to elucidate the structure of oligoribonucleotides. Thus SIMS can be used as a useful method for structural analysis of a wide variety of oligoribonucleotides.

The case report here was documented clinically, cytologically and histologically over a period of 5 years. A 49-year-old man was admitted to our hospital in September, 1975 because of a painful lump over the sternum and a diagnosis of solitary well-differentiated plasmocytoma (BJ-lambda) was made. The tumor promptly subsided following chemotherapy. In July, 1980 as it had in June, 1979, the sternal tumor relapsed; morphologically it was a poorly differentiated plasmocytoma. It promptly subsided following the start of chemotherapy and/or radiation therapy. In February, 1981, an abdominal tumor and ascites developed, morphologically it was plasma cell sarcoma composed of giant and bizarre tumor cells of plasma cell origin as demonstrated by immunoelectron microscopy. In April, 1981, the patient died of cachexia. At autopsy, multiple tumor formations consisting of giant and bizarre myeloma cells were found in the abdominal cavity, liver, retroperitoneum, diaphragm, left adrenal gland, mediastinum and lymph nodes. There was no infiltration of myeloma cells into bones other than the sternum.
Among 49 myeloma cases studied, 31 lesions were found to develop into extramedullary tumors in 8 cases and 22 lesions were found to develop into skeletal tumors in 14 cases. The locations of the extramedullary tumors were the lymph nodes, subcutaneous tissue, liver, kidney, stomach, and pancreas. The location of the skeletal tumors was the ribs, thoracic spines, and skull. These tumors were diagnosed as plasmacytomas by biopsy and/or touch- or aspiration cytology. The biological and clinical characteristics of the tumor-forming type compared with the non-tumor-forming type, were their occurrence in younger patients, a higher incidence of Bence-Jones proteinuria, more marked bone lesions and almost normal renal function. Concerning the monoclonal immunoglobulin (M-component) of the tumor-forming type, there were more of the Bence-Jones type and fewer of the IgA type than in the non-tumor-forming type. No relationship between tumor-formation and prognosis was seen.
