In vitro disruption of ATP dependent active transport following treatment with aldrin and its epoxy analog dieldrin in a fresh water teleost, Labeo rohita.

Tissue homogenates of brain, gill, liver and kidney of Labeo rohita were subjected in vitro to the various concentrations as 5.00, 1.66, 0.55, 0.18 and 0.06 mu M of 2 organochlorine pesticides aldrin and dieldrin and the disruption of ATP dependent active transport (involving ATPase) was studied. The inhibition of total ATPase, Mg2+ and Na+, K+-dependent ATPase by both the pesticides was quite similar and there was an increased inhibition with increased concentration of the toxicant, however, at the lowest concentrations of both the pesticides stimulation was observed in all tissues except gill Mg2+ ATPase where insignificant inhibition was observed after dieldrin treatment. Inhibition of ATPase system was noted maximum in brain followed by gill, kidney and liver of the experimental fish, however in each tissue maximum inhibition of ATPase was noted after dieldrin treatment than that of th e aldrin treatment. Therefore, the observed action of these pesticides might be related to the ability of the compounds to alter the cellular membrane configuration by binding with the lipid portion of the membrane and this blocking the movement of substances by active transport.

Adenosine Triphosphatases↗

Adenosine triphosphatase activity in few tissues of a fresh water teleost, Channa gachua following in vivo exposure to endosulfan.

Fresh water teleost fish, Channa gachua, were chronically exposed to various sublethal concentrations of endosulfan (0.00534, 0.00355, 0.00213 and 0.00174 mg/1) at 25 +/- 4 degrees C for 30 and 60 days respectively. The study shows enzyme inhibitions, greatest in oligomycin-insensitive Mg2+ ATPase in brain, gill and liver tissues, with pronounced effects at highest endosulfan concentration for 60 days exposure. However, in kidney the highest inhibition was of oligomycin-sensitive (mitochondrial) Mg2+ ATPase. The study indicates that endosulfan interferes with energy metabolism in vivo. The marked sensitivity of mitochondrial Mg2+ ATPase to endosulfan is suggestive of the potential for endosulfan to interfere markedly with various energy requiring processes in the fish body.

Seasonal histological changes in the gonads of two teleost fishes, Notopterus notopterus (Pallas) and Colisa fasciatus (Bloch. Schn.).

1. The reproductive cycle of the gonads of the Notopterus notopterus and Colisa fasciatus have been studied in the present account. 2. In the ovary of both the fishes same developmental stages have been observed. Ovarian cycle is distinguished in the 4 phases depending upon their changes in nucleus. Before spawning the enlargement of the ovary is due to the accumulation of the yolk in the egg. 3. Similar accounts of 4 important stages in the maturation of testis are given. A detailed description is given to gametogenesis in both species, and correspond closely with the macroscopic field stages. The tubules of testis in both the fishes are divided by the connective tissue strands. Spermatogonia present in these tubules play main part in spermatogenesis. 4. The spawning in both the fishes occurs during late May to August.

Comparative study of morpho-histology of gonads of three teleost fishes.

1. In the present paper comparative study of morphohistology of gonads of 3 teleost fishes has been made. 2. The gonads are unpaired in Notopterus notopterus and paired in Nandus nandus and Colisa fasciatus. 3. The testis in Notopterus notopterus is flat structure and is situated ventrolaterally. In Nandus nandus the testes are paired elongated and slightly flattened structures while they are paired threadlike structures in Colisa fasciatus. 4. Histologically the testis consists of lobules filled with various stages of germ cells, such as primary and secondary spermatocytes, spermatids and sperms. 5. The testis can be differentiated into anterior and middle functional regions and the posterior non=functional region in the case of Notopterus notopterus while in Nandus nandus and Colisa fasciatus no such differentiation could be found. 6. The ovary is somewhat oval unpaired structure in Notopterus notopterus. In Nandus nandus ovaries are paired, elongated organ taper anteriorly while the ovaries are paired and triangular in Colisa fasciatus. 7. The histological structure of the ovary in all the 3 fishes represents the generalised teleostean structure. 8. The cystovarian condition of gonads are present in all the 3 fishes, Notoperus notopterus, Colisa fasciatus and Nandus nandus.

Histochemical mapping of certain enzymes in few fresh water teleosts. I. Acid phosphatase.

A detailed investigation of the distribution pattern of acid phosphatase in the different parts of alimentary canal and associated glands of Colisa fasciatus, Macrognathus aculeatus, Notopterus notopterus and Nandus nandus has been made. Though this enzyme shows its hydrolytic activity in all the parts of the digestive system yet its intense activity has been noted in the intestine, pyloric caeca, liver and pancreas of all the 4 fishes. Mucosal and submucosal layers of all the parts of the alimentary canal are the main seat of localization of this enzyme.

Structural observations on the gills of two fresh water teleost fishes, Notopterus notopterus and Colisa fasciatus.

1. The structural observations of the gills of Notopterus and Colisa fasciatus have been studied in detail. 2. The gills of Notopterus notopterus and Colisa fasciatus consist of 2 regions: a) Gill head region, and b) Gill lamellae region. Gill head region is covered by epithelial layer possessing the mucous and mast cells. Gill head region consists of bony arch, gill rakers and interbranchial septum. 3. Interbranchial septum is reduced in Notopterus notopterus while it extends near half to the length of the filament in Colisa fasciatus. 4. In both fishes, the adductor muscle, afferent and efferent branchial vessels and gill rays are present in the interbranchial septum area. 5. Gill lamellae region consists of primary and secondary lamellae in both the fishes. 6. In Notopterus notopterus the primary lamellae are compressed flat structures possessing gill rays, 1 afferent and 1 efferent branchial vessel, while in Colisa fasciatus 2 efferent vessels are present. 7. Secondary lamellae are arranged parallel to one another on both sides of the primary lamellae. Secondary lamellae consist of basement membrane, epithelial cells, vascular layer and pillar cells. Mucous and mast cells are also present in this region in case of Notopterus notopterus while only mast cells are present in Colisa fasciatus. 8. Mast cells are eosinophilic, highly granulated in both the fishes.

Studies on the morphology of the gills and gill arches with reference to the blood supply in Notopterus motopterus and Colisa fasciatus.

The morphology of the gills, with their blood supply have been described in Notopterus notopterus and Colisa fasciatus in some detail. Gills are curved and perforated on the dorsolateral and ventrolateral wall of the pharynx. The gills consist of 2 rows of filaments which are stacked one above the other to form a space. The gill filaments are smaller on both the ends and larger in middle. The gill filaments are of pink colour as they are supplied with blood. Gill rakers are large in size in Notopterus notopterus while they are small in Colisa fasciatus. 3 pairs of basibranchials are present in Notopterus notopterus which are covered by median membranous bony plate while 2 basibranchials are present in Colisa fasciatus. 3 pairs of hypobranchials are present in both fishes. 5 pairs of ceratobranchials are present in which Vth ceratobranchial bears teeth. 4 pairs of epibranchials are present. 3 pairs of pharyngobranchials are present in which the tip of the IVth pharyngobranchial bears minute teeth in Notopterus notopterus while in Colisa fasciatus IInd and IIIrd pharyngobranchial bear minute ones. One afferent branchial vessel is present in Notopterus notopterus and Colisa fasciatus in each gill like in other teleostean fishes. One efferent branchial vessel is present in each gill of Notopterus notopterus while in Colisa fasciatus 2 efferent are represented in each gill.

Studies on the toxicity of lindane on Colisa fasciatus (part I: TLm measurements and histopathological changes in certain tissues).

1. In the present investigation static bioassay studies and histopathological changes in some tissues (gills, liver and kidney) induced by Lindane 20% E.C. in the fish Colisa fasciatus have been taken under consideration. 2. With the help of bioassay studies, the TLm were noted. The TLm values were 0.87, 0.78, 0.68 and 0.64 mg/l for the temperature range 17 degrees C to 20 degrees C and 0.60, 0.56, 0.46 and 0.41 mg/l for the temperature range 32 degrees C to 35 degrees C at 12, 24, 48 and 96 h respectively. 3. The statistical analysis of different factors such as concentrations, time and temperature range based on the observed data gave the following results: a) At both temperature ranges, survival rates at different concentrations and time are significant at 1% and 5% level. b) The difference in survival rates at different temperature range are insignificant both at 1% and 5pected survival numbers. The standard errors were also calculated for these two lines which gave the insignificant results and there was no much change in the shape of experimental and expected lines. 4. The histopathological changes in the tissues (gills, liver and kidney) shows the following results: a) In the case of the gills there was a loss of different types of cells i.e. respiratory cells and blood cells. Blood vessels were atrophied and the outer membrane of gill lamellae was ruptured. The erosion of the tips of the gill filaments were also observed. b) In the case of liver the outer membrane was ruptured. Some hepatic cells were vacuolated and some completely degenerated. The central areas were much affected. Due to the toxic effect, large splitting of tissues were found inside the liver. c) In the case of kidney the outer epithelium, the parenchymatous cells of renal tubules, cuboidal cells of uriniferous tubules were much affected. Due to the degeneration of the cells large spaces were formed inside the tissue. Much affection was observed in the central areas.
