Laser microsurgery in cell and developmental biology.

New applications of laser microbeam irradiation to cell and developmental biology include a new instrument with a tunable wavelength (217- to 800-nanometer) laser microbeam and a wide range of energies and exposure durations (down to 25 X 10(-12) second). Laser microbeams can be used for microirradiation of selected nucleolar genetic regions and for laser microdissection of mitotic and cytoplasmic organelles. They are also used to disrupt the developing neurosensory appendages of the cricket and the imaginal discs of Drosophila.

Serotonin uptake by isolated adipose capillary endothelium.

The uptake of serotinin (5-hydroxytryptamine, 5-HT) by endothelial cells was studied in freshly isolated capillary and other microvessel (< 50 micrometers, inner diameter) endothelium obtained from rat epididymal fat pads. Endothelial cells incubated with [3H]-5-HT removed 60 pmol of [3H]-5-HT per mg of protein at 1 hr, an apparent Km of 3 X 10(-7) M and a Vmax of 20 pmol/mg of protein, measured at 15 min, were obtained. Uptake of 5-HT (10(-6) of M) was inhibited by ouabain, selected metabolic inhibitors (iodoacetate, 2--4 dinitrophenol and sodium azide), 4 degrees C, tryptamine (10(-5) M and the 5-HT antagonists, fluoxetine and imipramine (10(-5) M and 10(-4) M, respectively). At concentrations of 5-HT greater than Km value, uptake appears to be principally by nonfacilitative diffusion rather than by carrier-mediated transport.

Sister chromatid exchange and chromosome aberration rates in a group of psoriatics before and after a course of PUVA treatment.

Blood was cultured and chromosome preparations were examined for sister chromatid exchanges and chromosome aberrations in eighteen patients receiving photochemotherapy with 8-MOP and UV-A for the treatment of psoriasis, both before starting treatment and again 6 months later. The UV-A was administered both by means of low output (irradiance: 1.1--2.25 mW/cm2) and high output (irradiance: 6.1--7.6 mW/cm2) UV-A radiation cubicles. Irrespective of the intensity of irradiation or the total dose of UV-A administered there was no evidence of chromosome damage in lymphocytes following treatment.

A study of faculty attitudes toward Ohio State's three-year medical program.

Three-year curricula, once a proliferating innovation in medical education, are now rapidly waning with the fading of capitation support and the easing of physician manpower shortages. In this article the authors examine faculty opinion of a three-year medical program at the Ohio State University. Using a Likert attitude-scaling technique, the authors developed a questionnaire and administered it to full-time faculty members. Five issues upon which faculty opinion was found to be most diverse include: adequacy of content in a three-year program, time available for faculty to do research, work demands on faculty, basic medical knowledge of three-year students, and quality of post-M.D. positions obtained by three-year graduates. Overall, three general deficiencies in the three-year curriculum, as perceived by faculty, emerged: students have limited instruction in basic medical knowledge and skills; unnecessary emotional, physical, and economic demands are imposed; and neither student learning nor quality of teaching is enhanced.
