Cheiro-oral syndrome following midbrain haemorrhage.

Cheiro-oral syndrome, a sensory disturbance of the corner of the mouth and the palm of the hand due to a lesion in the midbrain has never been reported previously. We describe a 67-year-old woman with cheiro-oral syndrome following midbrain haemorrhage confirmed by computed tomographic (CT) scan. A CT scan and neurological findings showed that the syndrome was due to a lesion in the medial lemniscus of left midbrain. Consequently, not only parietal lobe lesions and limited lesions in the inferior medial portion of the posterolateral ventral nucleus (VPL) and lateral part of the posteromedial ventral nucleus (VPM), but also damage to a restricted group of fibres reaching these nuclei, may cause cheiro-oral syndrome.

Impaired granulocyte chemotaxis and increased circulating immune complexes in Kawasaki disease.

Our study was carried out to clarify the changes in granulocyte functions and circulating immune complexes in 32 children with Kawasaki disease. Patients were divided into two groups, those with or without coronary aneurysm. In the group with coronary aneurysm, impairment of both granulocyte chemotaxis and phagocytosis was found, together with higher circulating immune complexes and normal intracellular killing activity. In the group without coronary aneurysm, impaired phagocytosis was observed, with normal granulocyte chemotaxis, circulating immune complexes, and intracellular killing activity. No correlation was observed between granulocyte chemotaxis and circulating immune complexes. Impairment of granulocyte chemotaxis and circulating immune complexes may yield pertinent information as to the degree of severity of vasculitis in Kawasaki disease.

Sex difference in subunit composition of hepatic glutathione S-transferase in rats.

The activities of hepatic cytosolic glutathione S-transferases (GSTs) towards 1,2-dichloro-4-nitrobenzene in male rats were higher than those in females, however, the enzyme activities towards 1-chloro-2,4-dinitrobenzene were not significantly different between the two sexes. SDS-PAGE analysis of GSTs purified from male and female rat hepatic cytosols by affinity column chromatography showed that there was a significant difference in the subunit composition between the two sexes. With regard to the several isozymes of GSTs in male and female rats, isozymes with basic and neutral/acidic isoelectric points were separated into seven molecular species by chromatofocusing. These sex differences in the quantitative proportions of GST isozymes were also confirmed by immunotitration using anti-GST-BL and -AC antibodies. On the other hand, glutathione peroxidase (GSH-Px) activities in rat hepatic cytosol towards hydrogen peroxide and cumene hydroperoxide were markedly higher in females than in males. Of the two types of GSH-Px, selenoenzyme (Se-GSH-Px) and the Se-independent enzyme (non-Se-GSH-Px), the former was found to be mainly responsible for the sex difference in the enzyme activities. Moreover, the GSH-Px activity of GSTs, non-Se-GSH-Px, was also higher in females than that in males. Since GST isozymes of the BL type are known to possess GSH-Px activity towards cumene hydroperoxide, the increased activities of non-Se-GSH-Px in the female hepatic cytosol seemed to be mainly due to the increased transferase activities of the isozymes, GST-L2 and -BL.

The relationship of skin color, UVB-induced erythema, and melanogenesis.

The relationship between skin color, delayed erythema, and delayed tanning (DT) elicited by a single exposure of UVB was investigated. Both constitutive and facultative pigmentation were determined by skin reflectance using a melanometer. Skin reflectance using visible light was well correlated to the minimal immediate pigment darkening dose elicited by UVA irradiation, which may relate to epidermal melanin content, a determinant of skin color. Minimal erythemal dose (MED) was well correlated to skin color, but there was less correlation between minimal melanogenic dose and skin color or the MED, since melanogenesis is controlled by genetic factors. DT also correlated to the dose of UVB in terms of MED. A coefficient of the regression line of DT may suggest the tanning capacity of skin. The possibility of detecting mild photosensitivity in individuals from a regression line of the MED on skin color is suggested.

The effects of M-protein fraction of hemolytic streptococci on embryonic heart in the rat.

Wistar rats were administered M-protein fraction (MP) of group A type 12 hemolytic streptococci on day 9 of gestation. When 9 mg/kg of MP was injected, the incidence of malformations was 11.8%, and the incidence of cardiovascular malformations was 7.9%. The main malformations were ventricular septal defect, microphthalmia, and hydrocephaly. Light and electron microscopic studies revealed no myocarditis nor endocarditis in either the maternal or fetal heart. Fluorescent antibody technique demonstrated no immunological cross-reaction between MP and rat heart tissue. The results suggest that MP is slightly teratogenic in the rat.

[Clinical application of PPC for nonspecific chronic prostatitis].

The clinical effectiveness of PPC, amino acid preparation, on nonspecific chronic prostatitis was evaluated by the double-blind test method. A mixture of two pollen extracts which has been widely employed for the treatment of chronic prostatitis was used as a control. Neither antibiotics nor anti-inflammatory drug was administered during the investigation. A total of 76 cases was reported from six facilities but 14 of them were excluded or dropped out. In 32 cases in the PPC group, subjective symptoms such as discomfort after urination, discomfort in the perineal region, pollakisuria, sense of residual urine and dysuria were improved in 50.0 approximately 61.9% and 70.0 approximately 83.3% after 2- and 4-week administrations, respectively. Similar results regarding each subjective symptom were obtained in 30 cases of the control group and there was no significant difference between the two groups. In the overall clinical effectiveness concerning subjective symptoms, the effective rate including excellently, moderately and slightly effective was 81.3% with PPC and 83.3% with control drug. However when the effective rate was limited to excellently and moderately effective, PPC was slightly superior to the control drug (50.5% versus 36.7%). In regard to prostatic tenderness by the digital examination, both PPC and control drug induced relatively good improvement after 2-week administration. The degree of improvement, however, was not increased in the subsequent 2 weeks. Sclerotic change of the prostate was poorly recovered in both groups even after 4-week treatment. Swelling of the prostate was alleviated in 12.5% of the control group after 2 weeks and in 31.8% after 4 weeks.(ABSTRACT TRUNCATED AT 250 WORDS)

Role of N-acetyl-D-galactosamine residue on B151K12-derived T cell-replacing factor (B151-TRF) molecule in B cell-receptor binding and -stimulating activity.

The role of sugar moiety on T cell-replacing factor molecule derived from a monoclonal T cell hybridoma B151K12 (B151-TRF) was analyzed with respect to the interaction with receptor on B cells. The induction of B cell differentiation into Ig-secreting cells by B151-TRF was specifically inhibited by addition of N-acetyl-D-galactosamine (GalNAc) to culture. Such inhibition appeared to be attributed to the interference of GalNAc in the interaction of TRF with its receptor, because absorption of TRF activity with B cells was notably inhibited by the presence of GalNAc. To substantiate this point further, we established binding assay of B151-TRF molecule to the receptor on B cells by using 125I-labeled TRF fraction enriched by reversed-phase high-performance liquid chromatography and gel filtration. The results revealed that the binding of 125I-TRF molecule to the B cells was almost completely blocked by GalNAc. Moreover, the existence of GalNAc residue(s) on B151-TRF molecule was evidenced by the facts that 1) the TRF activity was eluted from lectin gels with specificity for GalNAc as revealed by the functional assay, and 2) the 125I-TRF molecule specifically bound to such lectin gels. Thus, the GalNAc residue(s) on B151-TRF molecule plays an important role in binding of TRF molecule to the receptor and in the stimulation of B cells. The molecular properties of B cell-stimulatory B151-TRF and its mode of interaction with corresponding receptor on B cells were discussed in the context of B151-TRF as a glycosylated lymphokine molecule and B151-TRF receptor as a carbohydrate-binding protein (animal lectin).

Formation of ester forms of riboflavin in the pituitary gland of rat.

We studied the formation of ester forms of riboflavin in the pituitary gland of rats using 14C-riboflavin labeling and a gel filtration technique. The active formation of ester fraction of riboflavin suggests that two synthesizing enzymes of ester forms, flavokinase and FAD pyrophosphorylase, exist in this organ. A minor peak containing a lesser amount of radioactivity contained protein as measured by UV absorption at 280 nm was found when supernatant from incubated homogenate was chromatographed on a Sephadex G-15 column. This suggests the presence of a riboflavin-binding protein; the amount of 14C-riboflavin bound to pituitary proteins was expressed as the binding capacity of riboflavin. No changes were found in the binding capacity or esterification of riboflavin in the pituitary gland of rats treated with ACTH, dexamethasone phosphate, or subjected to liver impairment by CC14 injection. However, esterification was increased at 24 and 48 h after adrenalectomy. We posit that the riboflavin turnover in the pituitary gland may not be subject to marked changes in order to maintain the function of this organ. In aged rats, esterification was lower than in young rats suggesting that the turnover of pituitary riboflavin is related to senescent phenomena.
