Urophonographic studies of the lower urinary tract: a new approach to urodynamics.

The detection and recording systems for urethral sounds during micturition were developed. This procedure was termed as "urophonography" and its recording as "urophonogram". Classification of urophonograms was undertaken on the basis of analyzing normal healthy male volunteers, and patients with benign prostatic hyperplasia. Four types of urophonogram were demonstrated. Urophonography was useful for investigation of dysfunctional voiding, and lower urinary tract obstruction. Urophonography will be a new approach to urodynamic investigations.

The synthesis of metallothionein-like protein containing zinc in liver of rat after administration of calcium and calcitonin.

Subcutaneous (s.c.) and intraperitoneal (i.p.) injection and oral administration of calcium (Ca) into rats increased Ca content of liver compared with control rats. When liver cytosol was filtered through a Sephadex G-75 column, zinc (Zn) content of metallothionein (MT)-like protein fraction was several times higher in Ca treatment than in control. Zn and copper (Cu) content of high-molecular (HM) and moderate-molecular (MM) fractions and Cu content of MT fraction was unaffected by Ca treatment. Zn in MT fraction showed two peaks in the direct-connection method of high-performance liquid chromatography to atomic absorption spectrophotometer (AAS). The retention time of these two peaks agreed with those of Zn or cadmium (Cd) in MT fraction of liver cytosol obtained from Zn- or Cd-administered rat. These results show that MT-like protein containing Zn is induced by Ca. In conditions showing a slight increase in liver Ca and a significant decrease in serum Ca by synthetic [Asu1,7] eel calcitonin (CT) injection, gel filtration of liver cytosol obtained from CT-treated rats showed a higher content of Zn and a higher radioactivity of [3H]cystine than that from control injection. This suggests that CT causes an increase in liver Ca and results in induction of MT-like protein containing Zn by Ca.

Leptomeningeal cyst of the posterior fossa. Case report.

A patient with a posttraumatic leptomeningeal cyst is described. The cyst simulated an occipital encephalocele and communicated with an epidural cerebrospinal fluid collection in the posterior fossa.

[Pharmacokinetics of astromicin intravenous drip infusion in patients with renal disorders].

Astromicin (ASTM), a new aminoglycoside antibiotic, was administered to 7 patients with renal disorders. Concentrations of ASTM in blood were determined for pharmacokinetic analysis. ASTM was administered by intravenous drip infusion over 1 hour at a dose of 200 mg to each of 6 patients and at a dose of 100 mg to 1 patient. Renal function was observed by the clearance of intrinsic creatinine (Ccr) as the indicator. Concentrations of ASTM in blood became higher and retention times longer as degrees of the loss of renal function were larger. Although ASTM is proved to be one of drugs with the highest degree of safety compared with other existing aminoglycoside antibiotics, it should be administered with care to patients with renal disorders.

A new method for drug application using electrolysis of water.

A micropipette for drug application using the electrolysis of water is described. The drug solution is applied from the tip of a micropipette by the pressure of the hydrogen and oxygen gas that are produced by electrolysis of water in the micropipette. The amount of the drug applied is proportioned to the applied current and time of electrolysis. This method is simple, reliable and effective for the application of any kind of drug.

Elastase--does this enzyme play any role in bladder cancer invasion?

Elastase activity in bladder cancer tissue and normal bladder epithelium was assayed with succinyl trialanine paranitroanilide (Suc-Ala3-NA), a synthetic substrate of elastase. Elastase in the tissue extract liberated paranitroanilide (NA) from Suc-Ala3-NA and coexistant endopeptidases produced alanine-NA and dialanine-NA. These products were detected by high performance liquid chromatography. All activities of elastase and endopeptidases in bladder cancer tissues from 20 patients are significantly higher than those in 17 normal bladder epithelia. In 20 bladder cancer cases evaluated, the relation between the activities of elastase and endopeptidases and the background factors of each bladder cancer case was examined. There was a significantly different activity of elastase between the superficial bladder cancer group (less than or equal to T1) and the invasive group (T2 less than or equal to). Elastase is assumed to play an important role in cancer invasion.

[Results of a survey on the daily activities of patients following ureterosigmoidostomy].

Results of a questionnaire study on the daily life of patients undergoing ureterosigmoidostomy are reported. Nine patients who survived for more than 1 year after the operation were subjected to this study. At the time of their regular checkup, they were asked questions including the frequency of febrile attack, degree of daily work, bathing, complaints, frequency of urination and incontinence and feeling towards ureterosigmoidostomy. These patients had neither serious complications nor complaints. All except for 1 patient were satisfied and were enjoying daily life after ureterosigmoidostomy.

Neuronal activity in the medial orbitofrontal cortex of the behaving monkey: modulation by glucose and satiety.

To investigate neuronal responses to interoceptive information, single neuron activity of the orbitofrontal cortex (OBF) of the behaving monkey was recorded during glucose injection, natural feeding and an operant bar press feeding task. Intravenous glucose injection had almost no effect on rates of spontaneous firing, but tended to attenuate neuronal responses during the bar press and reward periods. In about half of the neurons tested, the spontaneous firing rate changed for a relatively long period after the animal ate to satiety. The results suggest that blood glucose concentration is a modulatory factor in neuronal processing for feeding, but other interoceptive information generated by satiety strongly affects the activity of OBF neurons.

Monkey orbitofrontal neuron activity during emotional and feeding behaviors.

Long term, single neuron activity was recorded in the monkey orbitofrontal cortex (OBF) through chronic platinum-iridium microwire electrodes. Tonic as well as phasic changes in neuronal activity occurred upon presentation of palatable (food) and aversive stimuli during bar pressing for food. Lesions in this region are known to induce changes in emotional behavior. Therefore, the results suggest that the OBF neurons are involved in processing emotion.

A telemetry system for single neuronal discharge recording from behaving monkey.

A telemetry system for neuronal discharge recording from behaving monkeys positioned in a primate chair is described. Using FM telemetry system single neuronal activity was recorded from the monkey brain through five teflon-coated platinum-iridium microwire electrodes (25 mu in dia.). This system permits stable long term neuronal discharge recording during feeding behavior with minimal artifacts even during mastication. The system has a broadcasting distance of more than 30 m. The frequency response and the S/N ratio obtained from this system is almost comparable with that of the usual direct wire procedures, and the elimination of artifacts is superior. This report describes the procedure for constructing the microelectrode assembly and FM telemetry system.
