Interchromosomal asynchrony of DNA replication in polytene chromosomes of Drosophila pseudoobscura.

Analysis of 3H-thymidine autoradiograms of late third instar larval salivary glands of Drosophila pseudoobscura revealed a unique example of asynchrony of replication in the autosome complement. The two autosomal arms, 2 and 3, show similar labeling pattern during the initial phases, DD to 3C, and thereafter, the chromosome 3 has fewer labeled sites than chromosome 2 until the most terminal pattern, 1D. Detailed sitewise analysis of 3H-thymidine labeling shows that while nearly 54% of the sites examined in chromosome 2 have a labeling frequency greater than 50%, only 13% of all sites in chromosome 3 have labeling frequency at that range. The number of labeled sites on chromosome 3 plotted against that on chromosome 2 shows a hyperbolic profile rather than a linear relationship. The silver grain ratio of the 2nd to 3rd increases from 1.5 to 3.1 through different stages of the cycle. These results suggest that both chromosomes start replication simultaneously but the third chromosome appears to complete the replication earlier than the second. These data open up the possibility of separate control mechanisms for the initiation and termination of DNA replication in polytene chromosomes.

Antithymocyte globulin in renal transplant recipients. Report of a prospective randomized controlled trial.

Horse antihuman thymocyte globulin (HAHTG) combined with prednisone and azathioprine (lmuran) was used as immunosuppressive therapy in a randomized controlled sutdy in 50 renal allograft recipients. Side effects of HAHTG administration given intravenously were mostly mild. In the treated group, four patients out of 26 died of infectious complications, whereas in the control group, three patients out of 24 died of infectious complications (chi2 = .01,P greater than .05). The graft survival at 18 months was ten of 24 in the control group and ten of 26 in the treated group (chi2 = 1.26, P greater than .05). Cumulative graft survival was 58.3% in the control group and 38.1% in the treated group at 18 months. However, if we consider the people who died with a functioning graft not as graft failure but as if they left the study, then the cumulative graft survival is 64.5% in the control group and 65.9% in the treated group. Thus, the mortality from infective causes and graft survival were not significantly different between the two groups. Hence, we draw the conclusion that use of HAHTG did not exert a beneficial effect on the ultimate outcome.

Early severe renal allograft rejection.

An early, severe form of renal allograft rejection, occuring after an initial day or more of good function and within the first week after transplantation, is an increasingly recognized phenomenon. This type of rejection occurred in 17 of 187 (9%) postmortem-donor renal transplants. It could only be reversed in four grafts, and two of these failed because of recurrent rejection within four months. No clinical observations were helpful in determining which cases were likely to respond to antirejection therapy. Of the 17 patients, four died. Because of the poor graft survival and the high mortality, attempts to treat early severe rejection may not be warranted.

Protective effect of allopurinol in renal ischemia.

We have confirmed earlier findings that treatment with allopurinol offers some protection to the kidney from ischemic damage caused by total interruption of renal circulation. With this evidence, a clinical trial is warranted, particularly when periods of warm ischemia are unavoidable.

Hyperactivity and faster replicating property of the two arms of the male X of Drosophila pseudoobscura.

The two arms of the X chromosome of Drosophila pseudoobscura have different phylogenetic origin, the XL being homologous to the X and the XR homologous to the 3L of D. melanogaster. The replicative and transcriptive activities of the two arms have been examined in order to understand how such phylogenetically different components of the X contribute toward the chromosomal basis of dosage compensation. The 3H-uridine labelled autoradiograms of the polytene chromosomes of larval salivary glands reveal that the intensity of labelling in the XL and XR of the male is not significantly different from that in the two arms of the female, respectively. The number of grains on the two arms plotted against the grain number on an autosome follows a linear regression, and neither slope in the male is significantly different from its counterpart in the female. The 3H-thymidine autoradiograms show that in all phases of replication, viz. initial, middle and terminal, both arms of the X chromosome in the male are advanced by one step in the cycle. Results, therefore, suggest that both arms of the X of D. pseudoobscura, are hyperactive and faster replicating in the male. Such a situation might arise due to a primary signal coming from autosomally located regulators controlling the super-operon structure of the X chromosomal genes.

Photoreactivating property of the Vibrio cholerae cell system.

Ultraviolet-inactivated chlora phage PL163/10 underwent a maximum of abot 36% of photoreactivation within the host V. cholerae cells. UV-inactivated but non-infected V. cholerae cells also underwent the same degree of photoreactivation.

The high incidence of persistent secondary hyperparathyroidism after renal homotransplantation.

Sixty-eight patients were observed for a period of up to seven years after transplantation. Serum parathyroid hormone levels were measured in 41 patients; 17 patients had persistent hypercalcemia for a period of from one to seven years. Serum parathyroid hormone levels were elevated in seven of these 17 patients. Serum parathyroid hormone was elevated in 11 of the other 24 patients with normocalcemia. The function of the kidney was not affected either by the state of hypercalcemia or persistent hyperparathyroidism. Aseptic necrosis was significantly present in those of the hypercalcemic group. It is suggested that persistent hypercalcemia is an important precipitating, if not the main, cause of an aseptic necrotic condition of the bones.

Molecular weight analysis of the polypeptides of cholera phage PL 163/10 by polyacrylamide gel electrophoresis.

Cholera phage PL 163/10, belonging to Mukerjee's Group I, was purified by alternate cycles of low and high speed centrifugation. Gel electrophoresis revealed the presence of four polypeptide chains of respective molecular weights of 10,370+/-515 (A), 30,000+/-1,303 (B), 40,000+/-1,049(C) and 64,000+/-2,433 (D) daltons. Electrophoresis of the sample alkylated with iodoacetic acid resolved the presence of only one polypeptide chain of an average molecular weight of 10,310+/-565 daltons. The polypeptides B, C and D could be interpreted as trimer, tetramer and hexamer respectively of the polypeptide A, which was the basic structural protein of this phage.

Chromosomal basis of dosage compensation in Drosophila VIII. Faster replication and hyperactivity of both arms of the X-chromosome in males of Drosophila pseudoobscura and their possible significance.

3H-thymidine and 3H-uridine labeling patterns of the X-chromosome arms of Drosophila pseudoobscura have been examined autoradiographically. Results show that in all phases of replication, namely, initial, middle and terminal, both arms of the X-chromosome in the male are advanced by one or two steps of 3H-thymidine labeling in comparison with the autosomes, and both arms in the female show more or less similar labeling profile as the autosomes. Both the arms in the male show pale stainability and enlarged width ratio, as reported in other species. The 3H-uridine labeling patterns also reveal that both arms in the male incorporate twice as much precursor as the individual X in the female. Results, therefore, suggest that both arms of the X in D. pseudoobscura are faster replicating and hyperactive in the male, although it is considered that XL is homologous to the X and XR to part of the third chromosome of D. melanogaster.
