Production of macrophage activating factor by human leukemic T cell lines.

Human leukemic T cell lines were tested for their ability to produce a macrophage activating factor. When mouse peritoneal macrophages were cultured for 48 hr in the presence of culture supernatants from cell lines HPB-ALL, CCRF-CEM, or MOLT-4, glucose oxidation via the hexose monophosphate pathway was enhanced by five to seven fold. Culture supernatants from cell line HPB-MLT stimulated the oxidation to a lesser extent. However, cell line CCRF-HSB-2 was essentially inactive as a producer. The active supernatants also stimulated the release of hydrogen peroxide from macrophages, whereas the inactive one did not. Since treatment of the cell lines with 12-o-tetradecanoyl phorbol acetate or phytohemagglutinin had little effect on the production of the factor except HPB-ALL, the cell lines seemed to secrete the factor constitutively. The stimulatory effect was dose-dependent and evident at a concentration as low as a 1/80 dilution. The factor was resistant to heat treatment at 100 C for 20 min, nondialysable and sensitive to protease digestion. The activating factor could be partially purified by anion exchange and gel filtration chromatographies.

Antitumor activity of D-mannosamine in vitro: cytotoxic effect produced by mannosamine in combination with free fatty acids on human leukemia T-cell lines.

Cytotoxic effects of mannosamine and free fatty acids on human malignant T-lymphoid cell lines derived from patients with T-cell leukemia were investigated. The combination of mannosamine and an unsaturated fatty acid (oleate or linoleate) produced more striking cytotoxic effects on malignant lymphoid cells than on normal human lymphocytes. The amino sugars glucosamine or mannosamine in the combination caused a synergistic cytotoxic effect, while the other carbohydrates (N-acetylmannosamine, N-acetylglucosamine, or mannose) had little effect. On the other hand, the effect of saturated fatty acids (palmitate or stearate) in the same system was nil. An unsaturated fatty acid (oleate) caused an increase in lipid fluidity of the surface membrane in MOLT-4 lymphoid cells, which possess higher lipid fluidity in combination with mannosamine, while saturated fatty acids had no effect on the fluidity properties of the membrane lipids (even in the presence of mannosamine). The relationship between mannosamine and unsaturated fatty acids in cytolysis was discussed.

Ontogeny of the third component of complement of Japanese quails.

Ontogeny of quail complement was examined in terms of serum C3 level, cytolytic activity of total complement, functional activity of C3 via the classical pathway (CCP) and an alternative route (ACP), and the distribution of C3-bearing cells. In serum, C3 was detected from 7-day-old embryos by rocket immunoelectrophoresis. Thereafter, its concentration increased rapidly and reached the adult level at 4-5 weeks of age. Total activities of serum complement via CCP and ACP, both of which were assayed by cell lysis, were detected first in 10-day-old embryos, and increased sharply after hatching, reaching the maximum level at 5 weeks of age. Deposition of C3 on the cell surface via CCP and ACP was also clearly demonstrated from 10-day-old embryos by haemagglutination and immunofluorescence techniques, respectively. By an immunofluorescence technique using anti-quail C3 monospecific serum, cells belonging to the reticuloendothelial system were indicated to serve major sites for the synthesis of C3 during ontogenesis.

[Effects of neonatal androgenization on 7,12-dimethylbenz[a] anthracene-induced rat endometrial carcinogenesis].

Effects of neonatal androgenization on 7,12-dimethylbenz[a]anthracene (DMBA)-induced endometrial carcinogenesis in Sprague-Dawley rats are studied. Testosterone propionate (1.25 mg) was injected subcutaneously into 110 2-day-old rats. At 8 weeks of age, a pellet containing 0.2 or 1mg DMBA was introduced into each uterine horn of both neonatally intact control rat (NR) and neonatally androgenized rat (ASR). All rats were killed at 24 or 48 weeks of age. The results were as follows. Vehicle group: No malignant uterine tumor was observed during both 16- and 40-week observation periods in either 122 horns of NR or 98 of ASR. 0.2 mg DMBA treated group: No malignant uterine tumor was detected in either 80 NR or 72 ASR during a 16-week observation period. Although three malignant uterine tumors were found among 72 NR during a 40-week observation period, no malignant uterine tumor was observed in 30 ASR. 1mg DMBA treated group: Although one malignant uterine tumor of 16 NR was found during a 16-week observation period, no malignant uterine tumor was observed in 20 ASR. Four malignant uterine tumors of 168 NR induced by DMBA were histologically 2 squamous cell carcinomas and two sarcomas. As mentioned above, although four malignant uterine tumors were induced by DMBA in 168 NR, no malignant uterine tumor was found in 122 ASR. These results seem to indicate that neonatal androgenization suppressed the induction of malignant rat uterine tumors by DMBA.

ECG changes during cerebral angiography. An analysis of 334 patients--942 cerebral angiographies.

We have analyzed HR changes greater than 20% among 334 patients and 942 cerebral angiographies. A tachycardial effect was seen in 14.9% of patients, while a bradycardial effect was seen in 7.1% including two patients having cardiac standstill (0.5%). These two patients were examined without atropine premedication after subarachnoid hemorrhage. Patients under 19 years of age, unpremedicated with atropine sulfate and suffering from subarachnoid hemorrhage secondary to ruptured aneurysm or arteriovenous malformation showed a significantly high incidence of bradycardia. On the other hand, patients with the neoplastic disease and having an initial sinus bradycardia showed a significantly high incidence of a tachycardial effect.

Infectivity titration of hemorrhagic fever with renal syndrome virus: use of immune adherence hemagglutination for detection of virus growth.

Serial dilutions of hemorrhagic fever with renal syndrome viruses were inoculated into Vero-E6 cells in microplates. After 2 weeks of incubation, infected cells were disrupted by freezing and thawing, and virus antigens were detected by immune adherence hemagglutination. The infectivity titers of the virus as determined by this method were in close agreement with those obtained by the immunofluorescent antigen endpoint method. Then, a neutralization method was established. Japanese hemorrhagic fever with renal syndrome isolates, strains SR-11 and TR-352, were found to be distinct from Hantaan virus, strain 76-118, by the neutralization test.

Oncogenecity of an avian erythroblastosis virus in mutant strains of Japanese quails.

The susceptibility of 12 mutant strains of Japanese quails to the R strain of avian erythroblastosis virus (AEV) was examined. Three strains, SBPP, PNN and CWE, showed high susceptibility and developed various types of tumors including erythroblastosis, hemangioma and myeloblastic leukemia. In relatively resistant WE strain, increased incidence and various types of tumors were observed by modification of host conditions. These results indicate pluripotential oncogenicity of AEV in quails as well as partial control of AEV-oncogenesis by genetical background of the host.

[Analysis of the high risk factors of endometrial adenocarcinoma--abnormal excretion in urinary steroids in androgen sterilized rats].

We experimentally produced spontaneous endometrial carcinoma in anovulatory sterilized Sprague-Dawley rats (ASR) after 500 days of age and compared the 24 hour urinary steroid excretion patterns in ASR with those in normal control rats (NR). The results obtained were as follows: The 24 hours urinary 17-ketosteroid, 11-deoxy-17-ketosteroid and 11-oxy-17-ketosteroid levels of 500-day-old ASR were 127.5 micrograms/day, 73.4 micrograms/day and 54.1 micrograms/day which were 1/3, 2/5 and 1/3 of the same aged NR, respectively. Although the 24 hour urinary androsterone, etiocholanolone and dehydroepiandrosterone of 100-day-old ASR were significantly higher than those of the same aged NR, the seven fractionated 17-ketosteroid levels of 500-day-old ASR were lower than those of NR. Moreover, the urinary androsterone, etiocholanolone and 11-ketoetiocholanolone of 500-day-old ASR were significantly lower than those of NR. The total excretion of tetrahydrocortisol, tetrahydrocortisone and tetrahydrocorticosterone of 100- and 500-day-old ASR were 318 micrograms/day and 244 micrograms/day which were slightly higher levels than those of NR. And the urinary tetrahydrocortisol level of ASR was found to be higher than that of NR. The present results suggested that an abnormality in androgen metabolism of ASR relative to NR was one of the high risk factors in endometrial carcinogenesis in ASR.

[A case of myocardial infarction related to total cystectomy and ileal conduit diversion under general anaesthesia].

Prognosis is extremely poor when an aged patient is attacked by myocardial infarction during or after a major operation. This paper reports that a 63-year-old male patient who had not been specifically diagnosed to have any coronary artery disease before operation recovered from a myocardial infarction which occurred during total cystectomy and ileal conduit diversion under general anesthesia. The continuous electrocardiogram monitoring carried out during general anesthesia enabled us to discover and treat the disease early and save the patient's life. References to the myocardial infarction and the general anesthesia are also discussed.

[Pathological study of endometrial carcinogenesis in androgen-sterilized-rats].

As reported previously, squamous metaplasias, endometrial hyperplasias and cancers of the uterus occurred more frequently in androgen-sterilized-rats (ASR) than in normal rats (NR). This time we pathologically examined the endometrium of ASP in detail and obtained the following results: 1) In NR, aged from 70 to 750 days, no abnormal findings were found. In 61 ASR, aged more than 500 days, were found 8 simple endometrial hyperplasias, 2 atypical endometrial hyperplasias, 15 simple squamous metaplasias, 1 atypical squamous metaplasia, 2 adenocarcinomas, 1 adenosquamous carcinoma and 1 squamous carcinoma. 2) In ASR, proliferative and metaplastic changes coexisted in the endometrium. Two adenocarcinomas were accompanied by metaplasias and atypical hyperplasias, and one squamous carcinoma was accompanied by simple hyperplasias. 3) The endometrial carcinogenesis of ASR was thought to be as follows. In ASR, the endometrial epithelium loses its secretory function, becoming low columnar epithelium. A part of the non-secretory endometrium gains proliferative activity and progresses to metaplastic epithelium or to glandular hyperplasia. The former may develop to atypical metaplasia and squamous carcinoma, the latter to atypical hyperplasia and adenocarcinoma.

Malignant change of dermoid cysts of the ovary. Report on an adenosquamous cell carcinoma and a clear cell carcinoma.

Five cases of malignant change of ovarian dermoid cyst are presented. Two of these cases were of special interest due to the unusual nature of the malignancy. One case (an adenosquamous cell carcinoma) was apparently a malignant change of the columnar epithelium and its squamous metaplasia. The other case (a clear cell carcinoma) appeared to represent coincidental development of two tumors.

[Analysis of high risk factors in endometrial carcinoma--endocrine imbalances and dysfunctional uteri].

We analysed the high risk factors in endometrial adenocarcinoma by using androgen sterilized Sprague-Dawley rats (ASR). The results obtained were as follow: The response of uterine weight and uterine peroxidase induction by E2 administration in neonatal ASR was less than in normal control rats (NR). The mean cytosol E2 receptor content in the uteri in 300- and 550-day-old ASR was less than in NR. The plasma progesterone (P) values for ASR at 100, 300 and 750 days of age were significantly lower than the values for NR. The values for E1, E2 at 300 and 750 days of age were not significantly different between ASR and NR. Therefore, the plasma E1 + E2: P ratios in the ASR were almost 3 times greater than in NR. The plasma E1 : P and E2 : P ratios in 300-day-old ASR were twice as great as in NR. E2 : P in 550-day-old ASR with adenocarcinoma was 2.5 times and 4.6 times greater than in ASR without adenocarcinoma and NR, respectively. The present results suggested that endocrine imbalance (continuous estrogenic stimulation unopposed by P) and a dysfunctional uterus of long duration were the high risk factors in endometrial adenocarcinoma in ASR.

Teleocidin-induced phenotypic changes in thymic ALL cell line, HPB-ALL.

An indole alkaloid, teleocidin, induced phenotypic differentiation in the thymic ALL cell line HPB-ALL. Within 30 min of seeding in the presence of teleocidin, the cells formed a smooth round shape. Upon 5-day exposure to teleocidin, most of the cells became smaller and reminiscent of large or atypical lymphocytes. A thymic antigen stained with monoclonal antibody OKT6 was remarkably reduced while Leu2a-positive cells were increased in the treated cells. Upon teleocidin-induced phenotypic differentiation, the growth rate of cells was inhibited, their ability to incorporate DNA via [3H]-labelled precursors was reduced, their ability to bind sRBC rosettes was increased. These changes were paralleled to that induced by 12-o-tetradecanoyl-phorbol-13-acetate.
