An ingenol ester, milliamin, induced phenotypic changes in human thymic ALL cell line HPB-ALL. These alterations included morphological changes, increased sRBC-rosetting, the induction of phenotypic changes of differentiation antigens, i.e. a decrease in OKT4-, OKT6-, OKT9-, OKT10- and Leu3a-positive cells and an increase in Leu2a-positive cells, growth inhibition and decreased DNA synthesis. These phenotypic changes were qualitatively and quantitatively similar to those induced by other polyol diterpens with tigliane and daphnane hydrocarbon skeletons such as TPA and mezereine, respectively.
The 20,000 dalton light chain (L2) was isolated from rabbit and chicken striated muscle myosins, and the Ca2+-induced conformational changes of these proteins were investigated. 1) The reaction of thiol groups of L2 with dithiobisnitrobenzoic acid (DTNB), 2) measurements of the UV difference absorption spectrum, 3) measurements of Stokes radius (Rs) by gel filtration and 4) measurements of the ESR spectrum of L2 whose cysteine or tyrosine residues were spin-labeled were used for the structural studies. The effect of Ca2+ on phosphorylated L2 was also investigated. The long axis of chicken L2 was calculated as 136A from the Stokes radius, suggesting that the L2 is an asymmetrical molecule. After the addition of Ca2+ the long axis was reduced to 104 A. The same effect of Ca2+ has been reported with rabbit L2 (Alexis, N.M. & Gratzer, W.B. (1978) Biochemistry 17, 2319-2325). Besides this large shape change, the addition of Ca2+ to L2 induced environmental changes around tyrosine residues and also changes in the reactivity of cysteine residues with DTNB. Ca2+ is supposed to be bound to the N-terminal region of the molecule, while the tyrosine and cysteine residues are located at the C-terminal region, which is probably sterically remote from the N-terminal region. The reason for the remote effect of Ca2+ may be related to the structural rigidity of the L2 molecule. The functions of two properties of L2, Ca2+-binding and phosphorylation, are discussed in relation to muscle contraction.
In order to investigate the pathogenesis of juvenile chronic myelogenous leukaemia (J-CML) we examined the biosynthetic rates of G gamma and A gamma globin chains in the erythropoietic bursts from the bone marrow of a patient with J-CML. Globin chains were labelled with 14C-labelled amino acids, separated by isoelectric focusing and quantitated by fluorography. The synthesis of gamma-chains in the erythropoietic bursts comprised 89.0% of the total non-alpha-chains. The G gamma:A gamma ratio was 0.67, which is within the ratios obtained in newborns. Furthermore, individual erythropoietic bursts contained varying ratios of both gamma and beta chains and all revealed more G gamma than A gamma chain synthesis. The relative proportions of G gamma and total gamma chain biosynthesis in 62 separate erythropoietic bursts were 0.69 +/- 0.06 and 0.86 +/- 0.06, respectively. Cumulative frequency distributions of individual bursts differing in the ratios of gamma/(gamma + beta) and G gamma/(G gamma + A gamma) approached normal frequency distributions. These results suggest that the levels of Hb F in J-CML are controlled by qualitative changes in a single population of erythropoietic precursors, in which normal switching of the G gamma:A gamma ratio has not occurred, rather than by the abnormal proliferation of an F-cell clone.
An indole alkaloid, teleocidin, induced phenotypic differentiation in the thymic ALL cell line HPB-ALL. Within 30 min of seeding in the presence of teleocidin, the cells formed a smooth round shape. Upon 5-day exposure to teleocidin, most of the cells became smaller and reminiscent of large or atypical lymphocytes. A thymic antigen stained with monoclonal antibody OKT6 was remarkably reduced while Leu2a-positive cells were increased in the treated cells. Upon teleocidin-induced phenotypic differentiation, the growth rate of cells was inhibited, their ability to incorporate DNA via [3H]-labelled precursors was reduced, their ability to bind sRBC rosettes was increased. These changes were paralleled to that induced by 12-o-tetradecanoyl-phorbol-13-acetate.

Iron metabolism in rats with acute turpentine-induced inflammation was evaluated. In acute inflammation, reduced plasma iron and total iron-binding capacity values, shortened plasma iron disappearance time and lower plasma iron turnover were observed. The administration of 59Fe chondroitin ferrous sulfate in order to evaluate the reticuloendothelial (RE) function revealed a significantly increased 59Fe retention in the liver and lower incorporation into red blood cells. Radioactivity in hepatic RE cells was higher in acute inflammation than in control. These results suggest the possibility of a block in the transfer of iron from RE cells to the plasma iron pool during acute inflammation.
Latamoxef (LMOX) is a new antibiotic synthesized by Shionogi Research Laboratory. Chemically LMOX is especially unique with a sulfur atom replacing the oxygen atom in the 1 position of the conventional cephalosporin nucleus, and in addition, this antibiotic has a cephamycin-like structure. The antibacterial activity of LMOX shows high potency against Gram-negative bacteria, but tends to be weak against Gram-positive bacteria. The tissue levels of LMOX in humans after intravenous injection of 1 g were examined. The levels in uterine and adnexa uteri tissue at 1 hour after administration were 25.4 and 27.4 micrograms/g respectively. LMOX was administered to 147 cases in infections of obstetric and gynecological field. The clinical effect according to disease was 94.6% for intrauterine infections, 95.0% for adnexitis, 87.0% intrapelvic infections, and 100% for external genital organ infections, making a total of 92.5%. The rate of occurrence of side effects or abnormal laboratory findings was similar to or slightly less than that seen with other beta-lactam antibiotics.
Since a first successful A-C bypass using saphenous vein graft was performed in 1970, eighty five patients with severe coronary heart disease were operated on in our clinic. Up to date, A-C bypass was carried out in sixty four cases, and aorto-coronary sinus bypass (Ao-Cs bypass) as an arterialization of the coronary venous system was done in one case in the group of angina pectoris. Three cases among sixty four patients who underwent A-C bypass and a case of Ao-Cs bypass died of low cardiac output syndrome postoperatively. Other two cases died of refractory arrhythmia and cardiomyopathy 6 years after surgery. The remaining fifty nine cases are doing well without any complications during follow up from ranging 2 months to 13 years (mean 3 years 4 months). Recently, no operative death was found during past 8 years with development of surgical technique and myocardial protection. Ventricular function (cardiac index, LVedp, Max LV dp/dt, EF, LVEDVI , double products at rest and during treadmill exercise test) disclosed good results especially in complete revascularization group. Graft patency had a close correlation between a diameter of the coronary artery at the anastomosis site and bypass graft flow after completion of bypass. Subsequently, a good patency rate (84%-86%) was obtained in cases with the coronary artery more than 1.5 mm in diameter. It could be concluded that A-C bypass was useful procedure to improve ventricular function as well as clinical symptoms and longevity of the life.
A circadian rhythm was demonstrated in 10 males and 10 females with respective mean decreases in plasma iron concentration at 18 hr of 62% and 47% of morning values. Ferrokinetic studies performed on 5 normal males and 5 normal females showed a more rapid disappearance rate and lower plasma iron turnover in the evening. Parallel studies were done on 6 normal males in the morning and 4 normal males in the evening of the release of reticuloendothelial iron at 8 and 18 hr after intravenous injection of 59Fe chondroitin ferrous sulfate. The 6-hr release in the morning was 54.1% and in the evening 25.9%. Composite data from morning and evening showed a correlation between plasma iron level and plasma iron turnover (r = 0.76, p less than 0.001). A similar correlation existed between the plasma iron level and the percent of radioiron released from the reticuloendothelial system (r = 0.67, 0.02 less than p less than 0.05). These data are consistent with a fluctuating iron release from the reticuloendothelial cell in normal subjects, which would account for the diurnal variation in plasma iron.

The T-cell bearing Fc receptor (IgG-FcR+ T cell) has been considered as a suppressor or a part of the killer cell, as determined by its function. The population of IgG-FcR+ T cells was determined by Moretta 's method in patients with urinary bladder cancer, urological benign diseases and in normal subjects. The population of IgG-FcR+ T cells in the peripheral lymphocytes of 16 patients with urinary bladder cancer was 20.5 +/- 10.1%, that of 7 patients with urological benign diseases was 9.5 +/- 3.2%, and that of 8 normal subjects was 9.0 +/- 2.1. The population of IgG-FcR+ T cells in the peripheral lymphocytes patients with high stage bladder cancer was significantly higher than that of patients with low stage cancer. In low stage bladder cancer cases, the population of IgG-FcR+ T cells was decreased to the normal range at three weeks after removal of the tumor. But in high stage bladder cancer cases, it was not changed at 3 weeks.
