Induction of leukemic infiltration by allogeneic transfer of HTLV-I-transformed T cells in rabbits.

An HTLV-I-transformed T-cell line from a (B/J x Chbb:HM)F1 rabbit was intravenously inoculated at a dose of 1 x 10(8) cells into B/J and Chbb:HM adult rabbits. All B/J rabbits died or became moribund 6-13 days later, whereas all Chbb:HM rabbits survived without disease symptoms. Marked leukocytosis was observed in Chbb:HM rabbits but not in B/J rabbits, the leukocyte counts in the peripheral blood of the former being 10-30 times the normal level 10 days post-inoculation with most cells being of normal lymphoid appearance. Pathological examination of dead B/J animals revealed leukemic infiltration of abnormal lymphocytes in major organs, the infiltrate being composed of medium-sized lymphoid cells with pan T-cell marker, convoluted vesicular nuclei and frequent mitotic figures. These results indicate that allogeneic transfer of HTLV-I-transformed cells can induce leukemic infiltration under appropriate conditions. Similar leukemic infiltration was observed in major organs of random-bred rabbits 72 h after a similar inoculation of the same cell line. Such adult T-cell leukemia-like disease induced in random-bred rabbits may be useful in studying the mechanism and the prevention of leukemic infiltration.

Lower fat deposition and energy utilization of growing rats fed diets containing sorbose.

1. Growing rats were fed diets containing graded levels (0, 100, 200 and 300 g/kg diet) of sorbose for 6 weeks. Protein, fat and energy deposition were determined by carcass analysis. 2. The values for growth, serum insulin level, digestible energy (DE), metabolizable energy (ME) and fat and energy deposition declined with the increment of dietary sorbose. 3. The efficiency of protein utilization (protein retained/protein consumed) was hardly affected by dietary sorbose. The DE and ME of sorbose per se was calculated as 14.09 and 12.35 kJ/g respectively. The efficiency of energy utilization (energy retained/ME intake) decreased with the increase of dietary sorbose, although sorbose had an ME. 4. The relative weights of gastro-intestinal tract and liver were positively associated with dietary sorbose level, although the reverse was true for the amount of stomach content, being heavier with higher dietary sorbose. 5. It is suggested that dietary sorbose, as a sweetener as well as a bulky agent, seems to be a suitable sugar for the obese and diabetic with special reference to lower body fat and energy deposition without reducing protein utilization.

Inhibitory effect of glycosides like saponin from soybean on the infectivity of HIV in vitro.

Soybean saponins isolated from soybean seeds were investigated for their antiviral activity on HIV in vitro, using an HTLV-I-carrying cell line, MT-4. Saponin B1 completely inhibited HIV-induced cytopathic effects and virus-specific antigen expression 6 days after infection at concentrations greater than 0.5 mg/ml. Saponin B2 also inhibited HIV infection, although less potently. Both saponin B1 and B2 had no direct effect on the reverse transcriptase activity of HIV. Saponin B1 also inhibited HIV-induced cell fusion in the MOLT-4 cell system. The results of this study suggest that soybean saponins, especially saponin B1, have inhibitory activity against HIV infection.

Enhancement of human immunodeficiency virus (HIV) replication in human monocytes by low titres of anti-HIV antibodies in vitro.

Low concentrations of serum obtained from a patient with acquired immunodeficiency syndrome (AIDS) enhanced the replication of human immunodeficiency virus type 1 (HIV-1) in a particular subclone of the CD-4-positive monocytoid cell line U937 clone 2. Cells of this subclone have a high expression of Fc receptors and a considerable degree of Fc-mediated phagocytic activity. IgG purified from the serum was also able to enhance the replication. These results indicate that low concentrations of human anti-HIV antibody may enhance HIV replication on human monocyte-macrophages. Furthermore, two mouse IgG1 monoclonal antibodies against gp120, the envelope glycoprotein of HIV-1, also induced enhancement at low concentrations. The binding of radiolabelled gp120 to the cells was increased at the same low concentrations. Antibodies against envelope glycoproteins may cause enhancement of HIV infection. Both normal and enhanced replication of HIV were completely inhibited by the masking of the binding site of CD4 molecules with F(ab')2 fragments of anti-CD4 antibody. Moreover, CD4-positive, Fc gamma RI-negative K562 cells and mouse macrophages failed to show any infection in the presence of antibody. These results suggest that CD4 molecules on the cell surface are necessary to cause enhancement of infection of HIV on monocyte-macrophages.

Induction of twinning in dairy or crossbred heifers by ipsilateral frozen embryo transfer.

Induction of twinning by ipsilateral nonsurgical transfer of frozen two or demi-embryos was attempted in 129 virgin dairy or crossbred heifers that had been kept under a stable in our station (group-1), mountain field (group-2) and private farm (group-3) conditions. Ninety seven heifers (75%) were diagnosed pregnant by rectal palpation at 35 to 60 days of gestation; 86 heifers produced 37 sets of twins and 49 single calves. Pregnancy rate of group-1 was lower compared to those of groups-2 and -3 (63%, 88%, 78%, respectively). Abortion and mortality rates of group-3 were significantly lower (P less than 0.05) than those of groups-1 and -2 (8% and 6%, 12% and 16%, 18% and 24%). Twin calves had a lower birth weight (P less than 0.05) than singles. But there was no significant difference in weight between singles and twin calves at 270-330 days of age. Dystocia and difficult delivery were not observed in females producing twins. The incidence of retained placentas in twin calving cows (17%) was higher than that of single calving cows (2%). The interval to conceive postpartum was longer in twin calving cows (95 +/- 41 days) than in single calving cows (87 +/- 26 days). Gestation length was 5.0 days shorter for twin calving cows (P less than 0.05).

[Treatment of suture-thread stones in the kidney by percutaneous nephrolithotripsy. Report of a case].

Percutaneous nephrolithotripsy was performed successfully to the stones in the left kidney, in which pyelolithotomy had been done three years earlier. There were foreign body stones with silk sutures used in the previous surgery as the nuclei. All the stones were removed, and a stricture in the renal pelvis was liberated endoscopically. No reports of endoscopic management to foreign body stones in a kidney are found in the Japanese literature. A few ways of the treatment are discussed.

A new bioassay for human granulocyte colony-stimulating factor (hG-CSF) using murine myeloblastic NFS-60 cells as targets and estimation of its levels in sera from normal healthy persons and patients with infectious and hematological disorders.

[3H]thymidine uptake by NFS-60 cells in microcultures was found to increase in a linear fashion with the increasing doses of purified recombinant human granulocyte colony-stimulating factor (rhG-CSF). Such increases were found neither with rhG-CSF samples pretreated with rabbit anti-rhG-CSF serum nor with other human colony-stimulating factors such as granulocyte-macrophage colony-stimulating factor (hGM-CSF) or macrophage colony-stimulating factor (hM-CSF). Based on these findings, sera from normal persons and patients with severe infections or various hematological disorders were tested after dialysis using this system in order to determine whether G-CSF levels in sera can be estimated or not. In ten normal persons, five patients with acute myelogenous leukemia (AML M1, M2, and M3), five with myelodysplastic syndrome, and four with chronic myelogenous leukemia, no increases in [3H]thymidine uptake were found within the dose range of 0.4 microliters to 50 microliters. In contrast, linear dose responses parallel to a G-CSF standard curve were observed in one patient with a severe bacterial infection, four with aplastic anemia, two with acute myelomonocytic leukemia (AMMoL) (M4), and two with idiopathic neutropenia tested. From the standard curve, the probable levels of G-CSF were calculated as follows: approximately 200 pg/ml with infection, 130-220 pg/ml with aplastic anemia, 150 and 200 pg/ml with AMMoL, and 1120 and 1200 pg/ml with idiopathic neutropenia. The activities of sera were reduced by the anti-rhG-CSF serum pretreatment in the same way as documented in the case of rhG-CSF. Furthermore, the level in a patient with a severe infection became undetectable soon after elimination of the infection and blood neutrophil counts had returned to normal. These findings indicate that the microbioassay system will be useful for measuring circulating G-CSF levels which would fluctuate in accord with requirements for stimulating neutrophil production or with abnormal production of hG-CSF.

[Eosinophil kinetics].

The kinetics of 111In-oxine labeled eosinophils were studied in 3 cases of reactive eosinophilia and were compared with those of neutrophils in patients with CML and neutrophilia. The disappearance curve of the labeled eosinophils consists of two exponential components. There was a slightly increase of radioactivity between these two components, suggesting the presence of recirculation. The disappearance rate of eosinophils was faster than that of neutrophils in CML and neutrophilia. In the cases of eosinophilia and CML, the marginal pool was larger than the circulating pool. In CML, the marginal pool was the largest among these three disorders. The granulocyte turnover rate in eosinophilia was less than in CML or chronic neutrophilia. It is suggested that the migration of eosinophils is less active than that of neutrophils.

Signet cell carcinoma of the stomach in a patient with acquired immunodeficiency syndrome: a case report.

A case of signet cell carcinoma associated with AIDS is presented. A 50-year old Japanese man with hemophilia A was suffering from human immunodeficiency virus (HIV) infection, the result of multiple injections of clotting factor concentrates. A diagnosis of signet cell carcinoma of the stomach was reached upon endoscopic and histological examinations. Opportunistic infections of esophageal candidiasis and candida septicemia occurred. The patient died of repeated gastrointestinal bleeding and cachexia. Although there is a possibility of the patient having a coincidential carcinoma along with AIDS, the HIV infection, perhaps, had a role in causing signet cell cercinoma.

Regulation of expression and transforming ability of the c-erbB-2 gene.

The human c-erbB-2 gene encodes a growth factor receptor-like protein that is highly homologous to the epidermal growth factor (EGF) receptor. Previous studies have shown that the c-erbB-2 gene is preferentially expressed in fetal epithelial cells, suggesting that the ligand of the gene product controls the growth of these cells. Molecular analysis of the c-erbB-2 promoter region revealed three regulatory systems: they are a typical set of CCAAT and TATA boxes, GC boxes, and Myb binding sites. Binding of the c-myb and B-myb gene products to the promoter region seems to down-regulate the c-erbB-2 mRNA synthesis. Elevated expression of this gene is often associated with adenocarcinomas such as breast cancers and stomach cancers and is correlated with the spread of the former. Involvement of the c-erbB-2 gene in the tumor development is further demonstrated by induction of B-lymphomas in transgenic mice carrying normal or mutated forms of this c-erbB-2 gene. In addition, the transforming potential of the c-erbB-2 gene is closely correlated with elevated tyrosine kinase activity of the gene product, which is negatively regulated by its carboxy-terminal sequence of about 230 amino acid residues.

[Blood cell kinetics in the spleen].

The mean intrasplenic blood cell transit time (STT), splenic blood cell volume (SV) and the slow mixing splenic blood cell volume (SSV) have been measured in normal subjects and patients with congestive and infiltrative splenomegaly. The normal red cell SV was 0.47 +/- 0.14 (mean +/- SD) % of the circulating red cell volume (RCV). In patients with congestive and infiltrative splenomegaly, SV was 9.82 +/- 3.55% and 0.96 +/- 0.71% of RCV, respectively. The SSV was 69.1 +/- 8.4% of the SV in normal subjects, but in patients with splenomegaly, its value was lower than that of normal SSV. The STT was 0.43 +/- 0.11 min. in normal subjects. In the patients with splenomegaly, it prolonged. But, the STT/SV and STT/SSV were rapid in patients with congestive splenomegaly and low in infiltrative splenomegaly. It seemed that SST/SV and STT/SSV were useful to distinguish between congestive and infiltrative splenomegaly. In the intrasplenic granulocyte kinetics, SV was similar to that of red cell. The STT was longer than that of red cell, in normal subjects and patients with splenomegaly. In the intrasplenic platelet kinetics, the ratio of cellular splenic volume to general circulation was the largest in platelets. The STT of platelet was the slowest in blood cells.

[Acute promyelocytic leukemia with a history of RAEB in transformation and the 15/17 translocation].

A 34-year old female was admitted to our clinic because of fever and general fatigue on March 26, 1987. On admission, peripheral blood (PB) revealed pancytopenia. Bone marrow smears revealed 9. 0% of promyelocytic cells with or without Auer rods. Diagnosis of RAEB in transformation was made. Chromosome study of the bone marrow cells showed t(15; 17) in 3 out of 20 cells analysed. After 3 months, the leukemic cells were observed in PB and increased in number. Then the patient showed bleeding tendency and fibrin degradation products (FDP) increased up to 40 micrograms/ml. And the leukemic cells were over 30% in PB at the end of July, 1987. The diagnosis of APL with DIC was made. To our knowledge, this is the first case of APL with a history of MDS with t(15; 17).

[The use of MCNU to a patient of primary myelofibrosis complicated with pericardial effusion and proteinuria].

A case of primary myelofibrosis complicated with pericardial effusion and proteinuria is described. A 66-year-old female was admitted to our hospital because of abdominal fullness and shortness of breath. On admission, hepatosplenomegaly and pericardial effusion were observed. Blood examination revealed leukoerythroblastic anemia and thrombocytosis with tear drop cells and giant platelets. Bone marrow aspiration was dry tap and its biopsy showed remarkable myelofibrosis. Urinalysis indicated severe proteinuria. Although neutrophilic alkaline phosphatase score was low, no signs of acute blastic crisis of chronic myelogenous leukemia was found. The diagnosis of an atypical type of primary myelofibrosis was obtained. Administration of MCNU was started in August 1987. Hepatosplenomegaly, pericardial effusion and proteinuria were gradually improved after the administration. The etiology of the pericardial effusion and proteinuria were not obvious, however, these facts suggest that these abnormal findings might be related to PMF itself and MCNU was effective to PNF.

[The dynamics of leucocytes and complements in endotoxin induced uveitis].

The dynamics of leukocytes, complement and tumor necrosis factor (TNF) were studied in endotoxin induced uveitis (EIU) in rats. Endotoxin was administered to footpads and the time courses of the following were examined: 1) the number of leukocytes in peripheral blood 2) the number of leukocytes and protein concentration of aqueous humor 3) complement (CH50, C1, C3, C5) in serum and aqueous humor 4) TNF in serum 5) histological changes in the eyes, lungs, liver, and skin. The number of leukocytes in peripheral blood decreased to one third that of controls during three to six hours after endotoxin administration, but increased thereafter along with the number of leukocytes and protein concentration in aqueous humor. More leukocytes were observed than in controls in the small vessels of the iris, lung, liver and skin, some of which attached to the vascular endothelium. Serum C3 increased and serum C5 transiently decreased after endotoxin administration. Serum TNF reached a peak (3500 IU/ml) at 1.5 hours and rapidly decreased subsequently. It is suggested that the adhesion of leukocytes to vascular endothelium and the activation of complement system may play important roles in the development of EIU.

[Bone marrow aplasia without blast crisis in a case of CML of 10-year survival].

A case of chronic myelogenous leukemia (CML) of 10-year survival in described. A 44-year old male was admitted to our hospital because of general malaise, abdominal fullness and fever in February, 1977. On physical examination, giant splenomegaly and hepatomegaly were detected. Peripheral blood examination revealed leukocytosis without hiatus leukemia , normochromic macrocytic anemia and thrombocytosis. NAP rate and score were 16% and 22. Cytogenetic analysis of PB without stimulator revealed 46, XY, Ph1. Then he was diagnosed as having a typical type of Ph1-positive CML. He had been successfully treated over 9 years by intermittent administration of busulfan. However, anemia suddenly progressed in February, 1986 followed by leukopenia and thrombocytopenia. Hemorrhage was not detected by the examination. Though he had been received blood transfusion, the anemia progressed rapidly. He was died of cachexia on 4th of August, 1987. The postmortem examination revealed bone marrow aplasia with no signs of blast crisis nor myelofibrosis. Secondary hemochromatosis was seen in the liver, spleen, pancreas and some other organs.

[Chemotherapy in the treatment of infections--basis and proper use of antibiotics].

There have been changes of pathogenic microorganisms (pathogens) and conditions of diseases in the field of obstetrics-and-gynecologic infections. Infections caused by weak-bacteria, i.e., opportunistic infections, have increased in the bacterial infections of genital organs. Increase of compromised hosts is one of the contributing factors to such increase. On the other hand, advance of chemotherapy plays a major role in the treatment of infections and many antibiotics including beta-lactam antibiotics (penicillins and cephalosporins), new quinolones and others have been introduced in the recent few years. When history of development of antibiotics and changes of pathogens in obstetrics-and-gynecologic infections are reviewed, it is noticeable that significance of gram negative bacilli (including anaerobic bacilli) has increased, and complicated infections (aerobes plus anaerobes) account for two thirds of the infections. Consequently, emphasis has been placed on the development of antibiotics which should be effective for these pathogens, and beta-lactam antibiotics such as penicillins and cephalosporins are the main flow of the recent antibiotics. However, in consideration of potentiated antibacterial activity and pharmacokinetics, new antibiotics of a little different usage and selection from the prevailing ones are in demand. Thus, the contents of chemotherapy have been changed as well as the changes of pathogens. Antibiotics have specific mode of action and point of action respectively and they show respective characteristics in their pharmacokinetics (serum concentration, urinary excretion and transfer into genital tissues).(ABSTRACT TRUNCATED AT 250 WORDS)
