Peripheral lymphocyte count in untreated non-Hodgkin's lymphoma patients and their lymphographic findings.

The relation between grades of foaminess in lumbar, iliac and inguinal nodes on bipedal lymphograms, and peripheral lymphocyte counts was assessed in patients with untreated non-Hodgkin's lymphoma. The lymphocyte counts were widely distributed in and around the normal range in patients with normal lymphographic findings, but in patients with increased grades of foaminess they were depressed to a marked degree. The decrease in peripheral lymphocyte counts in patients revealing no pathological grades of foaminess may be caused by the occult abdominal non-Hodgkin's lymphoma or may suggest the impending extension of the disease in these nodal chains.

Fingerprint inclusions and circular structures in the muscle. Report of a case.

Fingerprint inclusions, circular structures of unknown origin, mitochondrial changes, dilatation of the sarcoplasmic reticulum, rods of Z-line material, elongated T-systems, honeycombs, and myelin figures were present in some damaged muscle fibres. Many damaged fibres showed loss of myofilaments and groups of atrophied fibres occurred. The muscle biopsy was from the left biceps brachii and presented also the changes of polymyositis. The patient had muscle weakness, malignant hypertension, alcoholism, polyarthropathy, and evidence of mild peripheral neuropathy.

Ultrastructural changes in polymyositis.

Muscle biopsies from 32 cases with polymyositis and dermatomyositis were examined by electron microscopy. Most of the changes of the muscle fibres were non-specific and little structural difference was seen in the various clinical types. The muscle fibres sometimes showed rough endoplasmic reticulum, annulate lamellae and prominent Golgi apparatus. Thin and thick filamentous inclusions were present both in the sarcoplasm and nuclei of some muscle fibres. Thick filamentous inclusions were seen in chronic cases and were sometimes associated with annulate lamellae. Regeneration of muscle was often conspicuous. The endothelial cells of the blood vessels were hypertrophied; some cells contained granulotubular inclusions, rod-shaped bodies (Weibel-Palade bodies) and filamentous material. The basement membrane of the vessels was often multi-layered. The cells infiltrating the interstitial tissue included macrophages, lymphocytes, transformed lymphocytes, plasma cells, monocytes, mast cells and only occasional eosinophils and basophils. Lymphocytes, macrophages and a few plasma cells were seen between the plasma and basement membranes of degenerating muscle fibres. Changes were also noted in some intramuscular nerves, motor end-plates and a muscle spindle.

Bilateral popliteal artery entrapment syndrome--report of a case.

A 15-year old female presented with bilateral popliteal artery entrapment syndrome. The left popliteal artery was completely occluded and thromboendarterectomy was performed. Angiography on passive dorsiflexion revealed compression of the right popliteal artery, although it was free of symptoms and of normal caliber. End-to-end anastomosis in the normal anatomical position without division of the medial head of the gastrocnemius muscle was performed on both sides.

Free radical scavenging in protection of human lymphocytes against chromosome aberration formation by gamma-ray irradiation.

Human peripheral blood lymphocytes were exposed to 300 rad gamma-rays, in the presence or absence of radical scavengers, and the change in the frequency of chromosome aberrations was analysed with attention directed to the protection by scavengers against the formation of primary damage leading to chromosome aberrations. The results showed that the damage involved in the formation of exchange-type aberrations was efficiently protected by scavengers, and about 60 per cent of them resulted from indirect action that could be abolished by alcohols. The SH-compounds afforded additional protection. The comparison of protective ability with reaction rates demonstrated that the indirect effect was due to the reaction of OH radicals possibly to DNA as a target molecule, and the involvement of H and eaq-was unlikely. In contrast to the exchange-type aberrations, terminal deletions were not significantly protected, suggesting that the damage leading to the terminal deletion differed in its nature from the leading to the exchange-type aberration.

Chloride-induced contraction in a skinned fiber of toad striated muscle.

Cl- -induced contraction was studied in a skinned fiber dissected from toad sartorius muscle. When K-methanesulphonate in a relaxing solution was replaced isotonically with KCl, the skinned fiber contracted due to 'depolarization'-induced Ca++ release on the SR. Contraction was induced by superfusing the fiber with solutions containing different concentrations of Cl-. In the presence of 2 mM EGTA, the Cl- -induced contraction developed when the relaxing solution was replaced by a Cl- solution which contained up to 20 mM Cl-, while contraction started at 15 mM-Cl- in the presence of 0.5 mM EGTA. The peak tension of the Cl- -induced contraction depended on Cl- concentration without obeying the all-or-none law. When the fiber was treated with 1.5 mM caffeine, the threshold for the Cl- -induced contraction was significantly lowered. In spite of previous application of Cl- at different concentrations below 80 mM, the skinned fiber contracted by sudden increase in Cl- concentration, and the tension versus Cl- concentration curve revealed almost the same shape as that in a fiber equilibrated with Cl- -free solution. This suggests that the SR membrane is repolarized after transient depolarization caused by elevation of Cl- concentration.

Role of connectin in the length-tension relation of skeletal and cardiac muscles.

Resting length-tension relation was studied in glycerinated preparations of frog semitendinosus and atrial muscles. The sarcomere length was measured by using either the light diffraction method or photomicrographs. When the glycerinated preparation was treated with 1 N NaOH solution and subsequently with 2% SDS solution, the resting tension decreased. In glycerinated semitendinosus muscle the resting tension decreased to below 20% of the value before treatment. However, the glycerinated atrial muscle retained more than 50% of its resting tension in spite of such biochemical treatment. The thermoelasticity of the muscle, which was represented by (formula:see text), was determined. Glycerinated semitendinosus and atrial muscles, which were treated with 0.6 M KCl, showed negative beta at all sarcomere lengths. However, when the glycerinated muscle was treated with both alkali (1 N NaOH) and SDS solution, beta at the shorter length became positive (i.e., a rubber-like property). Since connectin survived the biochemical treatment (either NaOH or SDS), it was concluded that the fraction of resting tension, which remained in spite of the biochemical treatments, might be due to the connectin.
