Effect of neuroactive peptides on labeled 5-hydroxytryptamine release from rat spinal slices in vitro.

Effects of neuroactive peptides on the release of labeled 5-hydroxytryptamine (5-HT) from preloaded rat spinal cord slices were investigated. The 5-HT release was significantly stimulated by somatostatin (10-50 microM) and substance P (10-50 microM), but not by neurotensin (50 microM), beta-endorphin (30 microM) and methionine-enkephalin (met-enk) (100 microM). Somatostatin-stimulated 5-HT release was markedly inhibited by gamma-aminobutyric acid (GABA) (30 microM), but not by baclofen (30 microM) and met-enk (100 microM). Substance P-stimulated 5-HT release was strongly inhibited by GABA (30 microM) and baclofen (30 microM), but not by met-enk (100 microM). High concentrations (20 mM) of potassium also stimulated 5-HT release. The high potassium-stimulated 5-HT release was not affected by GABA (30 microM) and met-enk (100 microM). These results suggested further evidence on the important role of somatostatin and substance P as modulators of serotonergic neurones.

[Anaplastic transformation of well differentiated carcinoma of the thyroid].

Four cases of thyroid cancer with anaplastic transformation are reported. The anaplastic transformed cancers were found as a metastatic lesion 3-37 years after the resection of well-differentiated thyroid cancers. In one case, anaplastic cancer was mixed with well-differentiated cancer in an old metastatic focus but not present in the partially removed hyalinized primary lesion. The role of radiation therapy in anaplastic transformation was discussed.

Induction of hepatic glutathione transferase and suppression of 7,12-dimethylbenz[a]anthracene-induced chromosome aberrations in rat bone marrow cells by Sudan III and related azo dyes.

Noninbred Long-Evans rats fed the reduced form of glutathione (GSH) 2 hours before injection with 7,12-dimethylbenz[a]anthracene [(DMBA) CAS: 57-97-6)] showed a significant suppression of DMBA-induced chromosome aberrations (CA) in bone marrow cells. However, rats given injections of ethyl maleate [(EM) CAS: 141-05-9; (z)-2-butenedioic acid diethyl ester] 0.5 or 2 hours before being killed showed a remarkable fall in the hepatic GSH level. Furthermore, the administration of EM and bromosulfophthalein [CAS: 71-67-0; 3,3'-(4,5,6,7-tetrabromo-3-oxo-1(3H)-isobenzofuranylidene) bis(6-hydroxy)benzenesulfonic acid disodium salt] shortly before the DMBA injection inhibited the suppressive effects of Sudan III on DMBA-induced CA. A clear positive correlation was found between the capacity of Sudan III and related azo dyes to protect against DMBA-induced CA in bone marrow cells and glutathione transferase (GST) activity toward 1-chloro-2,4-dinitrobenzene in the liver cytosol induced by these azo dyes. DMBA activation with hepatic S-9 obtained from rats treated by polychlorinated biphenyls (PCB), phenobarbital, and Sudan III in the Ames system was high in this order as was also the case for the cytochrome P450 content. The addition of GSH to the Ames system containing S-9 from PCB-treated rats resulted in a substantial loss in the mutagenicity of DMBA. The present results suggest that GST and GSH play important roles in DMBA inactivation in rats previously administered Sudan III.

[Thermotherapy for cancer of the urinary bladder in combination with tegafur and picibanil--with special reference to the serum bladder and bladder cancer tissue concentrations of tegafur in a perfusion fluid].

Thermotherapy combined with Tegafur and Picibanil was performed in 32 patients with cancer of the urinary bladder. The thermotherapy was performed with a closed circulation type of thermotherapeutic apparatus manufactured for trial, using a 3-way catheter. The flow quantity of the perfusate was 150 ml/min at a constant temperature of 43 degrees C. The temperature was monitored at the inlet and outlet catheter at the urethral meatus. The thermotherapy was performed for 6 hours once a week. Tegafur and Picibanil were added to the perfusate at the time of thermotherapy. No anesthesia was used. Complete disappearance of tumor occurred in 9 cases. The concentration of Tegafur in the serum, tumor and bladder wall was simultaneously measured. The serum levels after two hours of thermotherapy with 16 gm of dissolved Tegafur showed that FT-207 was 28.837 mcg/ml and 5-FU, 0.028 mcg/ml. After 4 hours the FT-207 was 36.464 mcg/ml and 5-FU, 0.040 mcg/ml. By the 6th hour FT-207 was 39.430 mcg/ml and 5-FU, 0.049 mcg/ml. After 24 hours FT-207 was 11.242 mcg/ml and 5-FU, 0.013 mcg/ml in the serum, while FT-207 was 8.205 mcg/g and 5-FU, 0.274 mcg/g in the bladder tumor, and FT-207 was 17.029 mcg/g and 5-FU, 0.157 mcg/g in the bladder wall. Nausea was observed in 1 case in which a large quantity of Tegafur was absorbed. No other side effects were noted.

[A case of giant prostate cancer].

A 67-year-old patient with stage C giant prostate cancer was treated with a combination of endocrine administration and chemotherapy. After administration of diethylstilbestrol diphosphate (250 mg/D, 16 days), bilateral orchiectomy and subsequent CDDP administration (30 mg/D X 5 days, e. 3 weeks, 4 courses), the primary tumor was reduced by about 90%. Clinical response was evaluated as partial response. Serum acid phosphatase activity, prostatic acid phosphatase, prostate antigen and nuclear DNA histogram served as useful tumor markers and changed in parallel to clinical course.

Establishment and characterization of a human monocytoid leukemia cell line, CTV-1.

A new human monocytoid leukemic cell line, CTV-1, was established from a patient with relapsed acute monoblastic leukemia. The characteristics of this cell line were evaluated by morphologic and cytochemical analyses, electron-microscopy, chromosome study, surface marker analysis and a study of differentiation potential with tumor-promoting agents.

Functional morphology of tonsillar crypts in recurrent tonsillitis.

The cryptal epithelia contained three types of micropores (termed microcrypt) in association with free cells. Type III microcrypts appeared between ordinary epithelial cells with microridges and "specialized epithelial cells". The specialized epithelial cells consist of ciliated cells and non-ciliated cells having secretory granules and pinocytotic vesicles. Horseradish peroxidase (HRP) was taken up by phagocytes which protruded from the microcrypt and the superficial squamous epithelial cells with pinocytotic vesicles of idiopathic hypertrophic tonsils. It is suggested that Type III microcrypts have two contradictory functions. One is an active absorption of exogenous antigens; and the other is an exclusion of exogenous invasions according to mucociliary function. In recurrent tonsillitis, HRP was remarkably detected in the dilated stratified squamous epithelial clefts, epithelial tunnels, subepithelial layers and in the lumen of the high endothelial venules. The occurrence of sclerotic germinal centres in recurrent tonsillitis, and in focal infection, was greater than in idiopathic tonsillar hypertrophy, whereas the incidence in appearance of specialized epithelial cells was less frequent in acute tonsillitis than in recurrent tonsillitis, and in focal infection.

Phorbol ester increases the level of interleukin 2 mRNA in mitogen-stimulated human lymphocytes.

TPA alone did not induce the production of IL 2 in human tonsillar lymphocytes but enhanced the PHA-induced IL 2 production by seven-fold. That the effect of TPA was due to an increase in IL 2 mRNA was demonstrated by examining the amount of IL 2 mRNA translatable in Xenopus laevis oocytes, and by Northern blotting analysis using IL 2 cDNA as a probe. In these ways, it was shown that TPA alone did not induce any significant IL 2 mRNA synthesis, but when added together with PHA it increased the level of IL 2 mRNA by at least 10-fold, as compared with that induced by PHA alone.

[An autopsy case of ovarian clear cell carcinoma 14 years after irradiation for uterine cervical carcinoma].

An autopsy case of a 46-year-old woman who developed right ovarian clear cell carcinoma 14 years after radiation treatment for uterine cervical cancer, is reported. The pathological characteristics of the clear cell carcinoma were solid, tubulo-cystic and papillary structures of clear, granular eosinophilic, hobnail , and vacuolated cells. A Review of the literature on ovarian clear cell carcinoma and multiple cancers is also presented.
