Expression of Bcl-2 increases intracellular glutathione by inhibiting methionine-dependent GSH efflux.

Overexpression of Bcl-2 and related anti-apoptotic gene products has been shown to increase the intracellular concentration of the antioxidant tripeptide glutathione in neuronal and hematopoietic cells. A similar examination of HeLa cells that stably overexpress Bcl-2 (Bcl-2/HeLa) demonstrated that the reduced form of glutathione (GSH) was increased by 60% compared to control cells (80 nmol GSH/mg protein compared to 50 nmol GSH/mg). Expression of gamma-glutamylcysteine synthetase, the rate limiting enzyme for glutathione synthesis was found to be independent of Bcl-2 overexpression, as determined by Northern blot analysis and immunoprecipitation of [35-S]-labeled enzyme. Bcl-2 overexpression did not alter the rate of GSH biosynthesis, measured under steady state conditions. Thus, the increase in GSH concentration was not the result of increased synthesis. Two activities have been described which govern efflux of reduced glutathione (GSH), RsGshT known as the sinusoidal transporter and RcGshT, known as the canalicular transporter. Both are low affinity, bidirectional, ATP and Na-independent. Consistent with expression of sinusoidal activity, DTT was found to stimulate GSH efflux while the amino acid methionine inhibited efflux in both HeLa and Bcl-2/HeLa cells. However, methionine-dependent inhibition of efflux was found to be significantly increased by expression of Bcl-2. To test the prediction that the increase in GSH observed in Bcl-2/HeLa cells was mediated by methionine; Bcl-2/HeLa cells were cultured for 24 hrs in methionine-free growth medium. Under these conditions, the GSH concentration of the Bcl-2/HeLa cells dropped to the level observed in HeLa cells (50 nmol GSH/mg protein). These studies suggest that overexpression of Bcl-2 increases GSH levels by altering methionine-dependent GSH efflux, an activity associated in HeLa cells with expression of the RsGshT transporter.

Antitumor effect of a farnesyl protein transferase inhibitor in mammary and lymphoid tumors overexpressing N-ras in transgenic mice.

We tested the antineoplastic effect of the farnesyltransferase inhibitor L-744,832 in mammary and lymphoid tumors overexpressing the N-ras proto-oncogene in transgenic mice. Mice bearing mammary tumors were randomly assigned to receive daily 40 mg/kg s.c. injections of this compound (experimental group, n = 6) or vehicle (control group, n = 6) per day for 5.5 weeks. Treatment with the compound significantly reduced the mammary tumor mean growth rate in the experimental group (-0.7 mm3/day), as compared with the control group (+28.2 mm3/day; P < 0.001). There was a significant difference in lymphoma incidence at the end of the treatment between the experimental (0 of 6) and the control (3 of 6) groups (P < 0.05). Therefore, this compound is effective in treating in vivo mammary carcinomas and lymphomas in which an activated N-Ras pathway drives tumorigenesis. The number of apoptotic figures in mammary tumors was significantly higher (P = 0.04) in the experimental (14.7 +/- 8.1) than it was in the control (5.7 +/- 3.5) group, indicating that apoptotic induction could contribute to the mechanism of antitumor activity of this compound. We analyzed the level of processing of N-Ras and H-Ras after immunoprecipitation and Western blotting of protein extracts obtained from mammary tumors treated with L-744,832 or vehicle, either in vivo or in vitro (after primary culture of the same tumors), and from several in vitro treated control cell lines. In all compound-treated mammary tumors and cell lines, H-Ras was mostly unprocessed (more so after in vitro than after in vivo treatment), whereas N-Ras remained mostly processed. Both H-Ras and N-Ras remained fully processed in all vehicle-treated samples. These findings are consistent with a less intense antineoplastic effect of the treatment with the compound in our N-ras model than the effect previously reported for the same compound in H-ras transgenics. In addition, the finding that, in compound-treated mammary tumors, the N-Ras protein remains mainly processed suggests that, in our model, other proteins in addition to Ras may be a target for the compound. Our results and the previous findings of frequent N-ras activation in human hematopoietic malignancies support a role for L-744,832 in the treatment of lymphomas and of mammary carcinomas with an activated N-Ras pathway, as well as the testing of a farnesyl protein transferase inhibitor in humans to establish its clinical relevance.

Opposing effects of intracerebroventricularly injected norepinephrine on oxytocin and vasopressin neurons in the paraventricular nucleus of the rat.

Our previous study shows that intracerebroventricularly (i.c.v.) injected norepinephrine (NE) had different effects on the discharge of different firing patterns of magnocellular neurons in the paraventricular nucleus (PVN). In the present study we further classified antidromically identified magnocellular neurons into two groups: vasopressin (VP) and oxytocin (OT) secreting neurons, and found that of all 48 cases of magnocellular neurons, NE had mainly excitatory effects on 36 cases of putatively OT-secreting neurons, and inhibitory effects on 12 cases of VP-secreting neurons. The third ventricular injected NE had almost the same effect on two types of neurons as that of lateral ventricular injection, partly ruling out the possibility that the lateral ventricularly injected NE may have acted indirectly on the magnocellular neurons in PVN. The results show that different mechanisms may be involved in mediating the effect of i.c.v. injected NE on VP- and OT-secreting neurons in the PVN.

Immunogenicity of DNA vaccines expressing human immunodeficiency virus type 1 envelope glycoprotein with and without deletions in the V1/2 and V3 regions.

DNA vaccines that express the human immunodeficiency virus type 1 HXB-2 envelope glycoprotein (Env) with or without deletions of the major variable regions V1/V2 and V3 were tested for the ability to raise enzyme-linked immunosorbent assay (ELISA) and neutralizing antibody in New Zealand White (NZW) rabbits. Three forms of the Envs were examined: gp120, the surface (SU) receptor-binding domain; gp140, the entire extracellular domain of Env; and gp160, the complete form of Env. For the forms of Env containing the variable regions, the gp120-expressing DNA plasmid was more immunogenic than the gp140- or gp160-expressing DNA plasmids. Removing the V1/2 and V3 variable regions increased the immunogenicity of the gp140- and gp160-expressing DNAs. Deletion of the variable regions also resulted in antibody responses against determinants that were not presented by the forms of Env containing the variable regions. Despite the improved immunogenicity, removing the V1/V2 and V3 domains did not improve the ability of Env to raise neutralizing antibodies. These results suggest that increasing the exposure of internal structures of Env that include the CD4-binding site does not necessarily result in the generation of better neutralizing antibody.

Insulin does not promote rat mammary carcinogenesis.

Indirect evidence from both epidemiological studies and animal experiments suggests that insulin may promote breast cancer development. In this study, we directly tested for a promoting effect of insulin on mammary carcinogenesis in Sprague-Dawley rats. Fifty day-old female rats received an i.p. injection of 37.5 mg/kg methylnitrosourea (MNU). Five days later, the animals were randomized into two groups. One group received insulin injections five times/week until the time of death, while the other control group received similar injections of normal saline. Over the course of 26 weeks following MNU treatment, the mammary tumour incidence in the insulin-treated group did not differ significantly from the saline-treated controls. Furthermore, the number of tumours per tumour-bearing rat did not differ between groups. Our results demonstrate that insulin is not a promoter of mammary carcinogenesis in this model.

Relative fitness of three organophosphate-resistant strains of Culex pipiens pallens (Diptera: Culicidae).

Effects of dipterex, temephos, and chlorpyrifos resistance on the relative fitness of Culex pipiens pallens (L.) were evaluated by examining developmental and reproductive characteristics. Age-specific life tables of Dip-R- (dipterex), Tmo-R- (temephos), and Chl-R- (chlorpyrifos) resistant strains were compared with the susceptible strain to determine relative fitness based on population trend indices (I). The 3 organophosphate-resistant strains possessed reproductive and developmental disadvantages involving fecundity and survival relative compared with the susceptible strain, with relative fitness values of 0.465, 0.520, and 0.501, respectively. These results are discussed in relation to the reversal from resistance to susceptibility in natural populations of mosquitoes.

Immunization of the female genital tract with a DNA-based vaccine.

Vaccines are being sought for contraception and the prevention of sexually transmitted diseases. However, progress is slow in this area largely because of lack of information on induction of protective immune responses in genital tract mucosa. In this study, we investigated whether in vivo transfection with a model DNA-based antigen delivered by gene gun technology would induce an antibody response detectable in vaginal secretions. Female rats were immunized with plasmids encoding human growth hormone (HGH) under the control of a cytomegalovirus promoter (pCMV/HGH) via vaginal mucosa (V), Peyer's patch (PP), and/or abdominal skin (S) routes. Localization of HGH in the target tissues demonstrated that all three sites can be transfected in vivo with pCMV/HGH. Vaginal tissues expressed roughly the same level of plasmid as skin. Antibodies to HGH were detectable in serum and vaginal secretions in rats immunized with pCMV/HGH. In the rats primed and boosted vaginally, vaginal immunoglobulin A (IgA) and IgG antibody titers to HGH were sustained for at least 14 weeks, whereas rats immunized via other routes and protocols (S/V, S/S, PP/PP, or PP/V) did not consistently sustain significant vaginal antibody titers beyond week 6. DNA-based immunizations administered by the gene gun may be an effective method of inducing local immunity in the female genital tract.

Studies of the neutralizing activity and avidity of anti-human immunodeficiency virus type 1 Env antibody elicited by DNA priming and protein boosting.

DNA vaccination is an effective means of eliciting strong antibody responses to a number of viral antigens. However, DNA immunization alone has not generated persistent, high-titer antibody and neutralizing antibody responses to human immunodeficiency virus type 1 (HIV-1) envelope glycoprotein (Env). We have previously reported that DNA-primed anti-Env antibody responses can be augmented by boosting with Env-expressing recombinant vaccinia viruses. We report here that recombinant Env protein provides a more effective boost of DNA-initiated antibody responses. In rabbits primed with Env-expressing plasmids, protein boosting increased titer, persistence, neutralizing activity, and avidity of anti-Env responses. While titers increased rapidly after boosting, avidity and neutralizing activity matured more slowly over a 6-month period following protein boosting. DNA priming and protein immunization with HIV-1 HXB-2 Env elicited neutralizing antibody for T cell line-adapted, but not primary isolate, viruses. The most effective neutralizing antibody responses were observed after priming with plasmids which expressed noninfectious virus-like particles. In contrast to immunizations with HIV-1 Env, DNA immunizations with the influenza virus hemagglutinin glycoprotein did not require a protein boost to achieve high-titer antibody with good avidity and persistence.

YakA, a protein kinase required for the transition from growth to development in Dictyostelium.

When Dictyostelium cells starve they arrest their growth and induce the expression of genes necessary for development. We have identified and characterized a protein kinase, YakA, that is essential for the proper regulation of both events. Amino acid sequence and functional similarities indicate that YakA is a homolog of Yak1p, a growth-regulating protein kinase in S. cerevisiae. Purified YakA expressed in E. coli is able to phosphorylate myelin basic protein. YakA-null cells are smaller and their cell cycle is accelerated relative to wild-type cells. When starved, YakA-null cells fail to decrease the expression of the growth-stage gene cprD, and do not induce the expression of genes required for the earliest stages of development. YakA mRNA levels increase during exponential growth and reach a maximum at the point of starvation, consistent with a role in mediating starvation responses. YakA mRNA also accumulates when cells are grown in medium conditioned by cells grown to high density, suggesting that yakA expression is under the control of an extracellular signal that accumulates during growth. Expression of yakA from a conditional promoter causes cell-cycle arrest in nutrient-rich medium and promotes developmental events, such as the expression of genes required for cAMP signaling. YakA appears to regulate the transition from growth to development in Dictyostelium.

Isometric muscle strength in alcoholic and nonalcoholic liver-transplantation candidates.

Intensity of isometric muscle contractions was measured in alcoholic subjects with cirrhosis (N = 42), nonalcoholic subjects with cirrhosis (N = 33), and normal controls (N = 31). Muscle strength and endurance were comparable in the alcoholic and nonalcoholic cirrhotic subjects for all variables. Both cirrhotic groups were inferior to normal controls for all variables. The quantity x frequency (Q x F) index reported for the period during peak alcohol consumption correlated with 6 muscle-force variables, accounting for 9-20% of the variance. Alcoholic and nonalcoholic cirrhotic subjects did not differ in Quality of Life Inventory (QOLI) scales. Significant correlations, however, were found for the alcoholic cirrhotic subjects but not for the nonalcoholic cirrhotic subjects between quality-of-life indices and muscle strength and endurance. Muscle weakness is thus differentially associated with quality of life in alcoholic cirrhotic subjects as compared with nonalcoholic subjects with cirrhosis even though level of strength and endurance in the two groups is comparable.

[Glutathione S-transferase M1, T1 genotypes and the risk of esophageal cancer: a case-control study].

To examine the association between genetic polymorphisms of glutathione S-transferase (GST) M1 and T1 and susceptibility to esophageal cancer, a multiplex polymerase chain reaction method was used to detect the presence or absence of the GSTM1 and GSTT1 genes in genomic DNA isolated from surgically removed esophageal tissues or scraped esophageal cells from cases with cancer (n = 45), cases with severe hyperplasia (n = 45), and sex/age matched normal controls (n = 45) from a high risk area, Linxian, China. Results showed that the frequency of the GSTM1-null genotype in cancer cases (44.4%) or hyperplasia cases (44.4%) was not significantly different from that in controls (46.7%). Similarly, no statistically significant differences were observed in the frequency of GSTT1-null genotype in cancer cases (40.0%) or hyperplasia cases (37.8%) when compared with the controlled population (51.1%). However, the frequency of combined GSTM1-nonnull and GSTT1-nonnull genotypes in cases with cancer (40.0%) and cases with hyperplasia (37.8%) showed a significant increase compared to that in controls (22.2%). Persons with both GSTM1 and GSTT1 positive genotypes had 4-fold risk in developing esophageal cancer (odds ratio, OR = 4.20; 95% confidence interval, CI = 1.23-14.36) and 2.6-fold risk for hyperplasia (OR = 2.64; 95% CI = 0.84-8.30), respectively. These results suggest that combined GSTM1-nonnull and GSTT1-nonnull genotypes may act as risk factor in the development of esophageal cancer in Linxian population.

[The comprehensive treatment of varicosis of lower extremity with chronic ulcer of leg].

From Oct. 1993 to Dec. 1995, nineteen refractory cases with varicosis and chronic ulcer of lower limb were treated. The average age of these patients was sixty-eight, the disease history was more than 20 years. The size of the ulcer of the leg ranged from the minimum of 10 cm x 8 cm to the maximum of 30 cm x 15 cm. All of them had once received saphenectomy and split skin graft without ulcer healing before they were admitted in our department. Both venography and ultrasonography showed superficial venous valve incompetence. The following comprehensive treatment was adopted. Firstly, myoplasty around popliteal vein was done. Secondly, phlebexairesis and phleborrhaphy were done for the variciform veins through minor incision. Then through debridement of the ulcer was performed. Delayed split skin graft was exerted one week later. The result showed that all the cases were successful: the ulcer was healed and there was no recurrence of varicosis.

The influence of stress on edentulous maxillary bone with different artificial anterior tooth arrangements by three-dimensional photoelasticity.

OBJECTIVE: To investigate the influence of stress on edentulous maxillary bone different artificial anterior tooth arrangements, with the goal to establish theoretical evidence for optimal anterior tooth arrangements in complete denture restoration. MATERIALS AND METHODS: Three-dimensional photoelasticity experimental stress analysis was conducted. Shear stresses were measured and principal stresses were separated into six different models of maxillary central incisor arrangements. RESULTS: The value of maximum stresses and the absolute values of principal stresses and positive stresses were obtained. The distribution curve of shear stresses and principal stresses was also shown. CONCLUSIONS: The horizontal distance between the incisive papilla center and the incisal margin of the maxillary central incisor should be no more than 8 mm, if the anterior mandibular arch is larger than the maxillary arch. The principal stress is a better indicator of stress on edentulous maxillary bone when load is applied at the protrusive occlusal position.

[The effect of tangential excision on the local IL-8 release and wound inflammatory response in deep-partial thickness burn].

OBJECTIVE: The aim of this study is to investigate the effect of tangential excision on the local IL-8 release and wound inflammatory response. METHODS: The tissue specimens, which were taken from 7 patients with deep partial thickness burn before and after operation, were both cultured and histopathologically examined. The level of IL-8 in the culture medium was assayed, meanwhile local inflammatory response of tissue samples was evaluated. RESULTS: The result showed that the level of IL-8 is much lower in the postoperation wounds, while in the non-operation wounds the neutrophil infiltration was marked, with enlargement in necrotic area and disappearance of skin appendages. CONCLUSION: The findings suggested that tangential excision could reduce the local production of IL-8 and decrease local inflammatory response, which is helpful to prevent the progressive necrosis of burn wound.

[Clinical significance of pulmonary valvular resistance in children determined by color Doppler echocardiography].

Pulmonary valvular resistance (PVR) of 40 children with pulmonary valvular stenosis was determined by color Doppler echocardiography before and after percutaneous balloon pulmonary valvuloplasty as well as during their follow-up. The correlation coefficient of PVR between Doppler method and catheterization method was 0.85 ( P <0.01) and the correlation coefficient between PVR determined by former method and pulmonary valvular area determined by the latter was -0.80 ( P <0.01). This study indicates that color Doppler echocardiography can take the place of catheterization to determine PVR in assessing the severity of pulmonary valvular stenosis in children.

[The value of color Doppler ultrasonography in diagnosing aneurysm].

Using Doppler ultrasound, we detected 59 patients with aneurysm, 54 cases were confirmed by angiography or operation or both. Among them, 33 cases (63%) showed true aneurysm, 18 cases (33%) pseudoaneurysm, and 2 cases (4%) superficial congenital arterio-venous aneurysm. Eight cases were associated with dissected stripping. One case was falsely diagnosed by ultrasonography. The diagnostic accuracy was 98%. The results suggest that color Doppler ultrasonography is useful in detecting aneurysm.
