Serological relationships among some Pichia species.

Antigenic analyses of five species of the genus Pichia were carried out for taxonomic study by the slide agglutination method using monospecific and absorbed antisera and the agglutinin absorption technique. Comparative studies were also performed with a few strains of each of the same species and their classifications are discussed with respect to the antigenic structures and the patterns of proton magnetic resonance (PMR) spectra of their cell wall polysaccharides. ichia delftensis and Pichia zaruensis possessed thermostable antigens 1,2,5 and 11, and the former had also thermoabile antigen m. Both species were closely related to Candida krusei. Pichia toletana possessed thermostable antigens 1,2,5,11,17 and 49. Pichia bovis contained thermostable antigens 1,2,14,15,16,20 and 21, and it was related to most species of the genus Hansenula, although assimilation of potassium nitrate was negative. Finally, Pichia etchellsii possessed thermostable antigens 1,2,3,4,9 and 14, and was closely related to Pichia vini. Patterns of PMR spectra of mannans of these species also supported their serological relationships. Therfore, P. delftensis, P. zaruensis and P. etchellsii are considered to be the synonyms of Pichia fluxuum, Pichia dispora and P. vini respectively, although P. toletanan and P. bovis are independent species.

Hepatocyte growth factor and c-Met/hepatocyte growth factor receptor in pulmonary adenocarcinomas: an evaluation of their expression as prognostic markers.

The expression of hepatocyte growth factor (HGF) and its receptor, c-Met, was investigated immunohistologically in tissue specimens of patients with a pulmonary adenocarcinoma. The prognosis was significantly worse in the HGF-positive or c-Met-positive patients than in the negative patients. Multivariate analysis showed that c-Met had a significant effect on the prognosis, whereas HGF did not. Our findings suggest that HGF and c-Met play an important role in tumor progression and that c-Met can be a useful prognostic marker for pulmonary adenocarcinomas.

Transforming growth factor-beta isoforms expressions in pulmonary adenocarcinomas as prognostic markers: an immunohistological study of one hundred and twenty patients.

The transforming growth factor (TGF)-beta1, TGF-beta2 and TGF-beta3 expressions were immunohistologically studied in tissues specimens from 120 patients with a pulmonary adenocarcinoma. The results of a univariate analysis indicated that the expression of each of the TGF-beta isoforms correlated with the prognosis. Of the three prognostic factors examined using Cox's stepwise regression model, the TGF-beta1 isoform was found to correlate with survival. These findings suggest that TGF-beta isoforms play an important role in tumor progression and that the TGF-beta1 isoform is a useful prognostic marker for pulmonary adenocarcinomas.

An in vitro chemosensitivity test for gastric cancer using collagen gel droplet embedded culture.

Collagen gel droplet embedded culture drug sensitivity test (CD-DST) was applied to 33 patients with gastric cancer (30 primary tumors, 12 metastatic tumors and 25 biopsy specimens). Evaluable rates by CD-DST were 80% for primary tumors, 75% for metastatic tumors and 72% for biopsy specimens. Chemosensitivities of primary tumors were: 5-fluorouracil 25%, mitomycin C 17%, cisplatin 13%, adriamycin 17%, etoposide 21%. Chemosensitivities of metastatic tumors were lower than those of primary tumors. In 4 out of 6 patients who had measurable lesions, clinical responses to chemotherapy were predictable by CD-DST.

An individual patient data meta-analysis of long supported adjuvant chemotherapy with oral carmofur in patients with curatively resected colorectal cancer.

To reappraise the benefits of the long supported chemotherapy with carmofur, a meta-analysis based on individual patient data from the three clinical trials was performed by pooling 614 patients from three trials, there is a statistically significant survival benefit (2p=0.032) and disease-free survival (DFS) benefit (2p=0.021) for carmofur; and a highly significant advantage for carmofur in DFS (2p=0.0004) and in survival (2p=0.004) in Dukes' C patients. This IPD meta-analysis strongly suggested an effect of oral carmofur in a long supported chemotherapy for curatively resected colorectal carcinoma.

Endoscopic polypectomy of the large bowel: management of cancer-bearing polyps.

From January 1974 to May 1979, among 2,338 endoscopic examinations of the large bowel, 184 polypectomies were performed. Forty-two cases of familial polyposis coli were excluded from this study. Of the remaining 142 cases, 21 had cancer foci: fourteen were mucosally confined carcinomas, and seven submucosally invading carcinomas. Endoscopic polypectomy was considered a radical treatment for the former and not satisfactory for the latter. Therefore macroscopic type of polyp, location of carcinoma in the polyp, and lymphatic and venous permeation were the main criteria evaluated for the strategy of treatment in cases of submucosally invading carcinoma-bearing polyps. Radical resections are necessary in cases of semipedunculated polyp with carcinoma located near the base, and in cases of pedunculated polyps with carcinoma extending to the stalk. However, as general criteria for the management of submucosally invading carcinoma-bearing polyps have not yet been established, radical resections are indicated in cases which do not fit the generally accepted criteria, in order to attempt a curative treatment. Endoscopic polypectomy therefore seems to be of great value in the early detection of colorectal cancers and their treatment.

A rapid immunoperoxidase method for pathological diagnosis of occult thyroid carcinoma.

The authors report four unusual cases of clinically occult carcinoma of the thyroid. Exploratory excision of osseous metastatic lesions was performed for diagnosis in Patients 1 and 2, and fine needle aspiration of cervical metastatic lesions was used for diagnosis in Patients 3 and 4. A definite diagnosis of thyroid cancer could not be made by hematoxylin and eosin staining in Patient 2, nor by fine needle aspiration in Patient 3. However, the diagnosis of thyroid cancer in both patients was made using the rapid immunoperoxidase method with an antibody against thyroglobulin. The rapid immunoperoxidase method reported in this paper requires only about 5 minutes. Hence, this rapid staining method may be routinely applied to definitive identification of the thyroid as the source of metastatic lesions, prior to surgery.

Vascular endothelial growth factor and its receptor correlate with angiogenesis and survival in pulmonary adenocarcinoma.

The vascular endothelial growth factor (VEGF) and its receptor (Flt) protein expression was immunologically studied individually and in combination with the microvascular density (MVD) determined by the factor 8-related antigen (F8RA) and their prognostic value in 118 patients with a pulmonary adenocarcinoma. The presence of VEGF and Flt was associated with the MVD and prognosis. A reduced expression of both of VEGF and Flt correlated with less tumor angiogenesis and a better prognosis. Our findings suggest that VEGF and Flt expressions play an important role in tumor angiogenesis and that the VEGF and Flt coexpression promotes angiogenesis and metastasis in pulmonary adenocarcinoma.

Pancreatic duct patency after pancreaticogastrostomy: long-term follow-up study.

BACKGROUND/AIMS: To clarify whether the pancreatic duct remains patent during long-term follow-up of patients after pancreaticogastrostomy. In a previous study of pancreaticogastrostomy with post-operative follow up for 3 years after surgery, we found that the orifice of the pancreatic duct was difficult to detect in some patients because of swelling of the gastric mucosa. Previous studies have not examined pancreatic duct patency during long-term follow-up. METHODOLOGY: Between July 1985 and August 1989, 20 patients underwent a pylorus-preserving pancreaticoduodenectomy with reconstruction by pancreaticogastrostomy. Five of these patients were followed up post-operatively for more than 9 years to determine the patency of the pancreatic duct. All pancreatic anastomoses were performed by the telescopic method. RESULTS: All 5 patients were female, with a mean age of 65.4 years (range: 54-75). Median post-operative follow-up was 10.8 years (range: 9-12). The indications for surgery were carcinoma of the ampulla of Vater in 4 patients and chronic pancreatitis in 1 patient. Pancreatic duct patency was confirmed in 4 patients by gastroscopy and pancreatography. However, the anastomotic orifice could not be detected in the remaining patient because of complete coverage by the gastric mucosa. In this patient, pancreatic exocrine and endocrine function deteriorated with dilation of the distal pancreatic duct. The patient underwent a second operation involving dissociation of the pancreatico-gastric anastomosis and resection of about 1 cm of the fibrous, proximal portion of the pancreas. Reconstruction was performed with a Roux-en-Y pancreaticojejunostomy and a mucosa-to-mucosa anastomosis. CONCLUSIONS: Although pancreaticogastrostomy has been applied as a safe and straightforward method for reconstruction after pancreaticoduodenectomy, anastomotic stenosis is a potential late complication of this approach.
