The effect and toxicities of Cis-containing combination chemotherapy were tested in 28 patients with primary lung cancer. All patients were treated with 80 mg/m2 Cisplatinum on the first day and 750 mg ftorafur p.o. every day. In addition to these drugs, patients with squamous cell cancer were treated with continuous subcutaneous infusion of 4 mg/m2 Peplomycin for 5 days and one shot i.v. of 4 mg MMC. Patients with adeno- and large cell cancer were treated with 30 mg/m2 Adriamycin and 4 mg MMC, while patients with small cell cancer were given 150 mg/m2 VP-16 p.o. for 5 days. The following results were obtained. Of 22 evaluable patients, overall response rate was 50%. In each histologic type, response rate was 50% (5/10) for squamous cell carcinoma 50% (4/8) for adenocarcinoma 33% (1/3) for large cell carcinoma and 100% (1/1) for small cell carcinoma. No CR was obtained in this series. Main side effects due to Cisplatinum were nausea, vomiting, loss of appetite, mild leukopenia and thrombocytopenia, mild elevation of serum creatinine and BUN and alopecia, all of which were transient. Interstitial pneumonitis was observed in 40% of patients with squamous cell cancer. Two patients with adenocarcinoma died within 3 weeks after treatment due to embolism of the abdominal aorta and myocardial infarction probably caused by treatment with Adriamycin.
Seven normal volunteers and 11 patients with cerebrovascular dementia were studied about the relations between effect of aging, severity of dementia, cerebral glucose metabolism and metabolic response to verbal stimuli by positron emission tomography (PET) using C-11-glucose administered orally. The diagnosis of cerebrovascular dementia was done according to the Hachinski's ischemic score and X-ray CT findings, and the severity of dementia was classified according to the DR scale originated by Hasegawa. PET scanning started at 10 minutes and ended at 50 minutes after administration of C-11-glucose, and bloods were sampled at every 5-10 minutes during the PET scanning . Stimulation studies were performed after 3 or 7 days from control studies. An educational broadcasting program was used as the verbal stimuli. Regional distribution of glycogenic metabolites (RDGM: mg/100 g brain), which was a semi-quantitation of the pool of glycogenic metabolites mainly amino acids, were calculated. The RDGM values in elder normal subjects were significantly low compared with young normal subjects in frontal cortex (p less than 0.05). The decline in frontal cortex metabolism could have been caused by the morphological changes in the course of aging. In temporal cortex, there was no significance between two groups. RDGM increased significantly respond to the verbal stimuli in frontal and temporal cortex both young and elder normal subjects. The RDGM values in vascular dementias were significantly low (p less than 0.001) compared with elder normal subjects' in frontal and temporal cortex. Significant difference existed between mild and severe dementia in frontal cortex (p less than 0.05).(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of disodium cromoglycate (DSCG) on the action of various bronchoactive agents was investigated using isolated guinea-pig tracheal strips. DSCG attenuated the acetylcholine-induced contractile responses and shifted the dose-response curve of acetylcholine downward. It also attenuated the histamine-, serotonin-, bradykinin- and PGF2 alpha-induced contractile responses dose-dependently. On the other hand, DSCG potentiated the isoprenaline (isoproterenol)-, epinephrine-, PGE2- and salbutamol-induced relaxation responses, and shifted these dose-response curves downward. Such a shift became dominant by increasing the dose of DSCG. The above results suggest that DSCG might have a direct action on bronchial smooth muscle in addition to the inhibition of the release of chemical mediators from mast cells.
