Preparative scale thin-film dialysis apparatus.

A thin-film dialysis apparatus capable of processing large volumes of material is described. Dialysis casing, of defined molecular weight cutoff, is stretched on a compact frame producing a thin-film configuration. The dialysis unit is rotated by a variable speed motor with the direction of rotation periodically alternated by an electronic reversing control. This device utilizes the entire membrane area, eliminates solution stratification, and minimizes viscosity effects; membrane clogging is minimized should gelation or precipitation occur. The solutions are in contact only with the dialysis membrane; the construction materials are Teflon and polymethylmethacrylate (Plexiglas). At a load factor of 0.5 ml/cm of dialysis casing the half-time for solute removal, t1/2, for 8 M urea was 7 +/- 1 min; over 99% of the denaturant was removed in 1.5 h. The following operational factors are discussed: load capacity, rotation rate, membrane, ratio of internal to external solutions and solute contributions.

Ionizing radiation does not alter the antitumor activity of herpes simplex virus vector G207 in subcutaneous tumor models of human and murine prostate cancer.

Viral gene therapy against malignant tumors holds great promise for tumors that are susceptible to the oncolytic activity of viruses. One advantage of oncolytic viral therapy is that it can potentially be combined with other therapies, such as radiotherapy, to obtain an enhanced tumor response. In the case of prostate cancer, herpes simplex virus-mediated therapies have been shown to be highly effective in animal models; however, studies of the efficacy of combined viral and radiation therapy have not yet been reported. In this study, we have combined G207, a multimutated HSV type 1 vector, with external beam radiation therapy of prostate tumors grown subcutaneously in mice. We examined both the human LNCaP tumor in athymic mice and the mouse transgenic TRAMP tumor in either athymic mice or its syngeneic host, C57BL/6 mice. Virus was delivered either intravenously, in the case of LNCaP, or intratumorally, in the case of TRAMP. We found that individually, either G207 or radiation was effective in delaying tumor growth in these models. However, delivering the treatments simultaneously did not produce an enhanced effect.

Chelation therapy in children as treatment of sequelae in severe lead toxicity.

Certain children who experience a toxic lead episode requiring hospitalization in infancy or early childhood will manifest central nervous system dysfunctions, including hyperactivity, as sequelae of this experience. In this study, findings indicate that persistent, higher-than-normal lead levels, dating back to the time of the initial toxicity, may well be a mechanism underlying some of these sequelae. Consequently, some part of these sequelae may be preventable and/or treatable.

In vitro analysis of bromine chemical burns with use of full-thickness human skin.

Parameters of bromine injury were studied in vitro with the use of full-thickness human skin (HS) specimens--discards from various surgical procedures. The morphology of in vitro-treated HS resembled that of in vivo-injured skin. The damage was pronounced in the epidermis (destruction of the stratum corneum, and extensive vacuolation of keratinocytes) and the dermis (collagen coagulation), depending on the bromine concentration, exposure time, and application method. A decreased viability of epidermal cells, assayed by dye exclusion, was observed as well. Permeation parameters of bromine via abdominal HS were determined by quantitating bromine concentration in the donor and receiving compartments. The amount of bromine that permeated HS was inversely related to the concentration gradient applied. This in vitro study suggests that prompt treatment and early medical intervention may be required for successful healing of both severe and mild cases of bromine injuries.

Enchancement of the antiplaque value of antibacterial agents by enamel-conditioning methods. I. Rationale, mechanism, and initial findings.

A method is proposed to induce chemically the incorporation of bacterial agents inot enamel and thus rende this tissue resistant to bacterial colonization. It consists of promoting a slight initial dissolution of enamel, followed immediately by tis reprecipitation. During this latter phase, antibacterial agents included in the test formulations would become trapped in the reprecipitating enamel. That this happens is proved by the chemical detection of antibacterial agents in treated enamel, as well as by the development of a remarkable resistance of treated specimens to grow in vitro plaque.
