Improved assay for alcohol dehydrogenase activity in serum by centrifugal analysis.

We describe an improved method for determination of alcohol dehydrogenase (EC 1.1.1.1) activity in 60 microL of human serum, based on conversion of ethanol to acetaldehyde with simultaneous reduction of NAD+ in glycine NaOH buffer (pH 9.0) at 37 degrees C in a centrifugal analyzer. The final concentration of NAD+ was 10 mmol/L and ethanol was 20 mmol/L. The dilution curve was linear with enzyme activity up to 200 U/L, and results by this method correlated well with those by a manual method (N Engl J Med 279: 241-248, 1968). Within-run precision (CV) was 0.9 to 8.2% over the range of 4.5 to 88.1 U/L, and day-to-day precision was 5.4 to 5.6%. In sera from 198 healthy individuals, mean alcohol dehydrogenase activity was 1.6 (SD 1.2, range 0-5) U/L. To evaluate the clinical utility of determining alcohol dehydrogenase, we measured the activity of alanine aminotransferase and alcohol dehydrogenase in sera from 470 patients with various diseases in our hospital, and found that results for the two enzymes did not correlate well.

[A newly established melanoma cell line (GAK) with 5-S-cysteinyldopa phenotype].

The production of 5-S-cysteinyldopa by a newly established melanoma cell line GAK is reported. The cell line was derived from a metastatic inguinal lymph node of vulvar malignant melanoma. The cell line grew well without interruption for over 4 years, GAK cells were proved to have melanin granules and tyrosinase activity in their cytoplasma by Masson's staining and dopa reaction, respectively. Melanin granules were ultrastructually identified as melanosomes in various maturing stages. The chromosomal number varied widely and showed aneuploidly, but the modal chromosomal number was stable in the hypotriploid range. GAK cells were transplanted to nude mice and produced tumors resembling the original. Because glucose-6-phosphate dehydrogenase of GAK revealed a type B (slow) mobility pattern on electrophoresis, the possibility of Hela cell contamination could be completely excluded. High performance liquid chromatography revealed "5-S-cysteinyldopa", a new tumor marker of malignant melanoma, in culture media of GAK cells. The cell line described may serve as a representative model system for basic and clinical studies on malignant melanoma.

Survival of patients with superficial bladder cancer after treatment: a 10-year retrospective follow-up study.

The clinical aspects of 64 patients with superficial bladder cancer (Ta, T1, T2 according to the TNM classification, International Union against Cancer or Stage O, A, B1 according to Marshall) admitted to the Tsukuba University Hospital were analyzed retrospectively. Treatment of these patients was mainly by transurethral surgery. The overall 10-year survival rate was 73%. Eight patients died and four of them showed an invasive change, the so-called metamorphosis, during the follow-up period. Patients with papillary, pedunculated tumors had better survival rates than those with papillary, sessile tumors. Patients with tumors less than 1 cm in size had better survival rates than those with tumors 1-3 cm in size. There were no statistical differences in survival rates between patients with single and multiple tumors. Up to seven years from surgery, the patients with T2 tumors had lower survival rates than those with Ta-T1 tumors. The patients with grade I tumors had better survival rates than those with grade II tumors. There were no significant differences in recurrence rates related to the grade of the tumor. It is thought that the prognosis of superficial bladder cancer in our series depended mainly on the shape, size, number, depth and grade of the tumor.

Anti-idiotypic antibodies in a patient with monoclonal rheumatoid factor after pneumococcal bacteremia.

A 51-yr-old Japanese female patient with monoclonal IgM gammopathy with rheumatoid factor activity was admitted because of pneumococcal bacteremia. About 2 wk after admission, her rheumatoid factor activity became undetectable by RAHA test and radioimmunoassay, subsequent to the initial marked elevation. The suppressive capacity of the patient's IgG fraction on the rheumatoid activity of her monoclonal IgM on January 11 was determined. The IgG fraction obtained on February 22 blocked the binding of the rheumatoid factor to rabbit IgG. The suppressive activity in the IgG fraction of February 22 was shown to be localized within the F(ab')2 fragment. Furthermore, the specificity of the suppressive serum factor was shown by the inability to block the binding of SRBC coupled with diazotized phosphorylcholine to anti-pneumococcal antibody. Thus, the marked reduction of rheumatoid factor activity was considered to result from anti-idiotypic antibody transiently appearing in her serum after pneumococcal bacteremia.

Sodium butyrate produces concordant expression of "early placental" alkaline phosphatase, pregnancy-specific beta 1-glycoprotein and human chronic gonadotropin beta-subunit in a newly established uterine cervical cancer cell line (SKG-IIIa).

The production of early and term placental alkaline phosphatase (ALP), human chorionic gonadotropin beta-subunit (beta-hCG) and pregnancy-specific beta 1-glycoprotein (SP1) was confirmed in the newly established uterine cervical cancer call line SKG-IIIa. Treatment of these cells in culture with sodium butyrate caused an increase of all of these oncodevelopmental proteins. On the other hand, prednisolone treatment enhanced only term placental ALP, while it reduced early placental ALP, beta-hCG and SP1. These data suggested that such oncotrophoblastic proteins as early placental ALP, beta-hCG and SP1 were concordantly modulated by butyrate and prednisolone. These findings may support the possibility of the reexpression of sets of development phase-specific genes in cancer cells.

Experimental induction of two inner cell masses in mouse embryos by vinblastine treatment in vitro.

Induction of artificial fission of the inner cell mass in an in vitro embryonal culture system was attempted. Mouse blastocysts were collected from uteri on day 3 of gestation and exposed to vinblastine sulfate after removal of zona pellucida. Embryos in the control group had a single inner cell mass on the trophectoderm and developed to the postblastocyst stage. On the other hand, the inner cell masses of the embryos in experimental groups subdivided into two or more. The present results, therefore, revealed that the vinblastine treatment at the blastocyst stage induced fission of the inner cell mass in mouse embryos. Further studies are planned in improved culture conditions to determine whether each inner cell mass subdivision develops into independent embryos.

Sarcomatous Wilms tumor associated with consumption coagulopathy.

This is a case report of a sarcomatous Wilms tumor complicating disseminated intravascular coagulation with gastrointestinal bleeding, ascites, pleural effusion, and hepatitis after the second course of actinomycin D. A sarcomatous Wilms tumor is believed to have an unfavorable prognosis even if multimodal therapy is given. Arteriovenous fistulae which we created for purposes of chemotherapy and hyperalimentation by microvascular-sleeve anastomosis functioned well for three years in spite of repeated punctures.
