[Influences of serum magnesium on measurements of serum parathyroid hormone].

The amino sequence recognizable parathyroid hormones (PTH) in lower and non-lower magnesemic groups in randomized samples were measured by using various kinds of the well established PTH kits. The levels of carboxy terminal, mid-region and intact PTH in lower magnesemic group were more decreased than these in non-lower magnesemic group. It is likely that the shortage of magnesium in serum makes a suppression of PTH secretion.

[Azygos and hemiazygos connections imaged by two-dimensional, and pulsed and color Doppler echocardiography].

Identification of the azygos or hemiazygos connection is important in the diagnosis of congenital heart disease, especially in cases of the polysplenia syndrome. Although two-dimensional echocardiographic observation of this anomaly has been reported, the direct visualization of drainage sites is still rather difficult. A method for visualizing the azygos or hemiazygos connection by a combined use of two-dimensional and pulsed and color Doppler echocardiography was reported here. Subjects consisted of 10 patients with the polysplenia syndrome, each of whom had been confirmed as having the azygos or hemiazygos connection; five each of the azygos and hemiazygos types. The transducer was placed over the second intercostal space on the right side for the azygos, and on the left side for hemiazygos connection. The two-dimensional echocardiographic plane was adjusted to visualize the long-axis view of the superior vena cava; the right superior vena cava for the azygos and the left superior vena cava for the hemiazygos connection. The mode of the equipment was changed to color Doppler, and an orange-colored blood flow toward the transducer, draining into the superior vena cava was carefully sought. Finally the pulsed Doppler mode was selected and the sampling volume was set in the orange-colored area. The wave form of the pulsed Doppler echocardiogram of the azygos or hemiazygos vein potentially had two peaks per cardiac cycle, and was modified by respiration. We found azygos or hemiazygos veins coursing from the posterior aspect of the right pulmonary artery in seven of 10 cases by two-dimensional echocardiography alone, while we observed all veins by the additional use of color and pulsed Doppler echocardiography.

Preliminary experimental results of gas recycling subsystems except carbon dioxide concentration.

Oxygen concentration and separation is an essential factor for air recycling in a CELSS. Furthermore, if the value of the plant assimilatory quotient is not coincident with that of the animal respiratory quotient, the recovery of O2 from the concentrated CO2 through chemical methods will become necessary to balance the gas contents in a CELSS. Therefore, oxygen concentration and separation equipment using Salcomine and O2 recovery equipment, such as Sabatier and Bosch reactors, were experimentally developed and tested.

Clinical usefulness of serum tripeptide aminopeptidase activity in diagnosing liver diseases.

We examined the clinical usefulness of serum tripeptide aminopeptidase (EC 3.4.11.4) activity in diagnosing liver diseases. We found that increased tripeptide aminopeptidase activity accompanied an elevation in alanine aminotransferase activity (ALT) in the sera of patients with non-cancerous liver diseases and that these enzyme activities were correlated (r = 0.826-0.972). In contrast, an increase in tripeptide aminopeptidase activity did not coincide with an elevation in ALT activity in the sera of patients with cancerous liver diseases. We also observed a correlation in acute hepatitis and in patients following transcatheter arterial embolization of hepatoma throughout their subsequent clinical courses. The difference in serum enzyme activities for cancerous and non-cancerous liver diseases seems to result from changes in enzyme activities in cancerous liver tissues. We found that the ratio of tripeptide aminopeptidase to ALT activity in hepatoma tissues (1.9) was 4 times higher than that in normal liver tissues (0.5).

Immunoaffinity purification and characterization of leucine aminopeptidase from human liver.

Leucine aminopeptidase was purified from human liver cytosol to homogeneity, 1538-fold, with a yield of 84.4% by immunoaffinity chromatography. Increases in the activity and the stability of the enzyme were simultaneously observed during the purification procedure, suggesting the presence of some endogenous inhibitor in cytosol. The specific activity and Km value of the enzyme for L-leucine amide were found to be 58.00 mumol/min/mg of protein and 4.02 mM, respectively, at pH 8.0. The molecular weight of the enzyme was determined to be 360,000 by both polyacrylamide gradient gel electrophoresis and Sephadex G-200 gel filtration. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of native and dimethyl suberimidate cross-linked enzyme indicate that the native enzyme has two subunits of Mr 53,000 (a) and 65,000 (b) and is a hexamer arranged as a trimer of dimers (3 X (a X b)). The optimum pH was 10.5, and the enzyme was stable in the pH range from 7.5-8.5. The enzyme was activated by divalent metal ions, especially by Mg2+ and Mn2+, with no change in Km value. The enzyme was inhibited by metal-chelating agents, indicating it to be a metalloenzyme. Amastatin and bestatin strongly inhibited the enzyme, but leupeptin did not. The enzyme had a broad substrate specificity toward oligopeptides and amino acid amides but had little or no activity toward chromogenic substrates. The enzyme also could hydrolyze natural substrates contained in liver cytosol and accordingly produce many kinds of amino acids commonly found in proteins.

Stimulation of superoxide release in neutrophils by 1-oleoyl-2-acetylglycerol incorporated into pH-sensitive liposomes.

Incorporation of 1-oleoyl-2-acetylglycerol (OAG) into multilamellar liposomes composed of egg phosphatidylethanolamine (PE) and arachidonic acid (AA) resulted in a significant enhancement of superoxide release by guinea pig neutrophils when compared to free OAG. OAG incorporated into liposomes containing phosphatidylcholine and arachidonic acid were generally less effective than free OAG. The potency of the liposomes correlates well with the ability of the liposomes to undergo lipid mixing at acidic pH. The enhanced effect of liposome-associated OAG could be related to exposure to an acidic environment in the endosomes/lysosomes once liposomes are endocytosed by neutrophils.

Normal bone marrow adherent cell-conditioned medium corrects the impaired differentiation of cultured mononuclear phagocytes from vitamin D-deficient rats.

There is increasing evidence that 1,25 dihydroxyvitamin D3 acts directly to differentiate and activate cells of mononuclear phagocyte lineage (MNP). However, it seems possible that in bone marrow this hormone may modulate the differentiation of MNP via the factor elaborated by the stromal cells or the macrophages. We tested this hypothesis using two separate culture systems of bone marrow cells from vitamin D-replete (D+) and -deficient (D-) rats. The animals were maintained from the third day of lactation on D+ or D- diet for up to 105 days. In vivo studies showed that the long-term D- state resulted in a depletion of acid phosphatase-positive macrophagelike cells and osteoclasts in bones. After a 14 day incubation period, cultures of D- bone marrow cells contained an increased number of fibroblastoid cell colonies and a reduced number of macrophages attached to fibroblastoid cells, compared with the controls. When D+ bone marrow cells were cultured for 4 days in the presence of supernatants obtained from 14 day cultures of D- bone marrow (marrow adherent cell-conditioned medium, MACCM), the differentiation of MNP into macrophages was inhibited. Impaired differentiation was almost complete in D- bone marrow cell cultures to which D- MACCM was added. These cultures consisted of only 6.6% macrophages compared with 53.9% in the control cultures. However, when D- bone marrow cells were cultured with D+ MACCM, differentiation was restored and 75.4% of the cells were macrophages. These results suggest that a paucity of macrophages and osteoclasts in D- rats is in part, if not entirely, a result of a failure of stromal cells and/or macrophages in the bone marrow to release a factor(s) necessary for differentiation of MNP and osteoclast precursors.

Posterior lingual mandibular bone depression.

An unusual case of posterior lingual mandibular bone depression in a 39-year-old Japanese man is reported. The bilocular type of this condition was composed of 2 large ovoid radiolucent areas which were adjacent to each other. The diagnosis of this condition was made by sialography and tomography. The present case was compared with other unusual cases reported in the Japanese literature.

[A comparative double-blind trial of HSR-902 and butylscopolamine bromide for relieving colic in ureteral stone patients].

The clinical efficacy and safety of HSR-902 (tiquidium bromide, Thiaton) in patients with spastic pain caused by ureteral stones were evaluated in a double blind comparative trial. A daily dose of 30 mg HSR-902 or 60 mg butylscopolamine bromide as a control drug was orally administered for 7 days. With either drug marked improvement of spastic pain was observed. The time to obtain relief from spastic pain and utility rating were significantly more excellent with HSR-902 than with butylscopolamine bromide. With all other parameters used no significant difference was observed between the two drugs. Mild adverse effects such as abdominal discomfort and constipation were observed in 2 of the 87 cases in the butylscopolamine bromide group but none of the 83 cases in the HSR-902 group. Taking efficacy and safety of the treatment into consideration, no significant difference was observed in usefulness between the two drugs, and we were able to confirm the usefulness of HSR-902 for relief from spastic pain caused by ureteral stones.

Osteopathia striata with cranial sclerosis affecting three family members.

Skeletal surveys were performed on a 38-year-old Japanese mother, her son and daughter. The radiographs of both children showed characteristic features of osteopathia striata. However, in the mother, the skull, mandible, and lower extremities were homogeneously sclerotic with no evidence of a striated pattern of sclerosis in her skeleton. Additional features of striated sclerosis of the mandible in patients with osteopathia striata are discussed.
