Serum, erythrocyte, and liver levels of total corrinoids, cobalamin (vitamin B12), and cobalamin analogues were determined by differential radioassay in 27 patients with alcoholism. Compared with normal subjects, liver content of total corrinoids and cobalamin in alcoholics was low. Conversely, serum total corrinoids and cobalamin were high. Compared with normal, levels of erythrocyte cobalamin analogue in alcoholics were elevated, but levels of cobalamin were not. Analogues in liver represented a similar percentage of total corrinoids in alcoholics as in normals. The data confirm prior work suggesting that, in alcoholism and in liver disease, cobalamin depletion in tissues may be masked by normal to high serum cobalamin and analogue levels. The failure of damaged liver to take up from the serum cobalamin and analogues, compounded by release of these compounds and their binders from damaged liver into the serum, can account for these findings.
The Shinkansen Super-Express trains of the Japanese National Railways, the so-called bullet trains, run from early morning until late at night. Accordingly the car-cleaners of the Shinkansen must do late night shift work. A self-administered questionnaire survey including items about health and work conditions was given to 246 workers at Osaka Station, who were then divided into 3 groups: Group A consisted of 102 night workers working 5 successive days with 2 days off, Group B consisted of 124 alternate-day 24-hour workers working 3 days a week, while Group C included 20 day-workers working 6 days a week. A subjective fatigue inventory was administered before and after work for a week to 20 workers selected from each of three groups, group A, B above group D whose shift was 2 times 2 successive night shifts with one day off, and one holiday a week. The results of the questionnaire survey revealed that the rates of complaints such as gastroenteric disorders and general fatigue were the highest in group A. The results of the subjective fatigue inventory show a tendency for the number of complaints in group A and B to increase significantly after the last work shift of the week, but not in the case of group D. The authors demonstrated that there was a close relationship between patterns of night-shift work and various kinds of health problems, and concluded that to insert a day off on the third day of the group D shift was effective in reducing the work load.
The angiotensin converting enzyme (ACE) activity in the plasma collected from normotensive and hypertensive patients was measured chromatographically by two different methods using hippuryl-histidyl-leucine (HHL-ACE) and synthetic angiotensin I (AI-ACE) as substrates. A single peak was observed in the elution profiles of HHL-ACE on both Sephadex G 150 column and DEAE Sephadex A 50 cellulose column chromatographies. A single peak of AI-ACE appeared on the gel-filtration. However, several peaks of AI-ACE were seen on DEAE Sephadex A 50 cellulose column chromatography, suggesting that there are various types of ACE which hydrolyze angiotensin I but not HHL.

Analogues of vitamin B12 which appear to be noncobalamin corrinoids appear to be present in human red cells, liver, and brain. Their sources, nature, and effects require study, particularly with reference to their positive and/or negative effects on vitamin B12 metabolism. In normal persons, they are concentrated in liver, with only small quantities in red cells and still smaller quantities in brain. Their concentrations in disease states will be of interest, particularly in persons with varying degrees of neurological damage associated with deficiencies in vitamin B12 or analogue metabolism.

Of 16 human malignant tumor cell lines established in our laboratory, seven lines, including three gastric cancer cell lines derived from patients with cancer, were found to carry carcinoembryonic antigen on their cell surface, as determined by radioimmunoassay and indirect immunofluorescence. The existence of antibodies (IgG class) against the gastric cancer cell lines (HPE-GAC-T, -2, -3) and lung cancer cell line (HPL-Ad-K) in the sera of patients with gastrointestinal cancer (incidence 70.8%) was demonstrated by indirect immunofluorescence. In nonmalignant cases and healthy controls, the incidence was 7.7 and 3.2%, respectively. Specificity of the antibodies detected in the sera of patients with gastric cancer was examined by absorption and blocking test methods of immunofluorescence. Even though there was an apparent heterogeneity of the specificity among the antibodies, the detection of such antibodies may be a feasible and practical approach to a clinical diagnosis of malignancy.
Two restriction endonucleases with new sequence specificities have been isolated from Acetobacter aceti IFO 3281 and Bacillus aneurinolyticus IAM 1077 and named AatII and BanII, respectively. Based on analysis of the sequences around the restriction sites, the recognition sequences and cleavage sites of these endonucleases were deduced as below: (formula; see text)

Two new restriction endonucleases have been isolated from Flavobacterium okeanokoites IFO12536 and Micrococcus luteus IFO12992 and named FokI and MluI, respectively. Based on analysis of the sequences around the restriction sites, the recognition sequences and cleavage sites of these endonucleases were deduced as below: FokI: (formula: see text). MluI introduces double-strand cleavages at unique sequences that are completely two-fold rotationally symmetric like most type II restriction endonucleases. FokI belongs to a class of restriction endonucleases that recognize specific but asymmetric nucleotide sequences and introduce staggered cleavages at appointed positions away from the recognition sequences.
