[Study of cefroxadine in pediatrics regarding clinical efficacy and serum levels (author's transl)].

Basic and clinical evaluations of cefroxadine were carried out in children, and the following results were obtained. 1. Cefroxadine 20 mg/kg was administered to 9 children with heart disease for the prophylaxis against infections before undergoing cardiocatheterization and cardioangiography, and serum levels were determined. Peak levels reached after 30 minutes in 4 of the 9 cases, with a mean peak level of 22.5 mcg/ml and after 1 hour in 5 cases, with a mean peak level of 16.2 mcg/ml. Half life was 3.1 hours in the former group in a 6-hour blood sampling (1.04 hours in a 2-hour sampling) while in the latter group it was 1.37 hours. 2. Clinical responses were evaluated in 56 children comprising 23 cases of pharyngitis, 8 of tonsillitis, 13 of scarlet fever, 10 of urinary tract infections and 2 of impetigo. Fifty of these cases had excellent and good responses showing a efficacy rate of 89.3%. 3. From 42 of the cases, 43 strains were isolated as causative organisms. Major organisms included 27 strains of S. pyogenes, 9 of E. coli and 3 of S. aureus. As for bacteriological responses, all strains were eradicated. 4. No severe side effects were observed except for diarrhea of 1 cases and eosinophilia of 2 cases. Furthermore, no children refused to take cefroxadine dry syrup.

Systemic hyalinosis or juvenile hyaline fibromatosis. Ultrastructural and biochemical study of cultured skin fibroblasts.

Biochemical examinations of a hyalinized skin tumor of systemic hyalinosis (juvenile hyaline fibromatosis) revealed an increase in chrondroitin sulfate synthesis by skin fibroblasts with an enlarged, rough surfaced endoplasmic reticulum as well as an increase in chondroitin sulfate content. Thse results indicate that systemic hyalinosis is a connective tissue disorder characterized by abnormal GAG synthesis of fibroblasts.

Studies on amyotrophic lateral sclerosis by neutron activation analysis--2. Comparative study of analytical results on Guam PD, Japanese ALS and Alzheimer disease cases.

Metal analysis of calcium, manganese, aluminum and copper in CNS tissue samples of degenerative CNS disease cases (six Japanese ALS, three Japanese Alzheimer disease, four Guam PD, one Guam ALS) using neutron activation analysis, was conducted with following results: Five of six Japanese ALS cases, two of three Japanese Alzheimer disease cases and all of four Guam PD cases showed a high content of calcium and aluminum in CNS tissue with a significant positive correlation between calcium and aluminum and/or between calcium and manganese. These findings suggest a possible process of metal-induced soft tissue calcification with interaction of other di- and/or trivalent cations such as aluminum, manganese in CNS tissue of these degenerative CNS diseases.

Growth stimulative effect of parathyroid hormone, calcitonin and N6,O2'-dibutyryl adenosine 3';5'-cyclic monophosphoric acid on chick embryonic cartilage cultivated in a chemically defined medium.

PTH (0.5 unit/ml) and CT (0.5 unit/ml) strongly stimulated the in vitro growth of the chick embryo femur in terms of elongation and dry weight increase as well as an increase in the protein, hexosamine, hydroxyproline, DNA and RNA content of the femur. A synergistic effect was observed when PTH and CT were simultaneously added to the medium, suggesting that the mechanism of the growth stimulating effect of one of these hormones might be different from that of the other. The exposure of PTH to the femur caused an increase in cyclic AMP content. The direct addition of dibutyryl cyclic AMP (1 mM) to the medium mimicked the effect of PTH on the cartilage. PTH, therefore, seems to stimulate the growth of cartilage via the increase in cyclic AMP content of the femur. CT had no effect on the cyclic AMP content of the femur.

Selective activation of diaphyseal chondrocytes by parathyroid hormone, calcitonin and N6,O2-dibutyryl adenosine 3',5'-cyclic monophosphoric acid in proteoglycan synthesis of chick embryonic femur cultivated in vitro.

We have found that the developmental stage of chondrocytes in the chick embryo femur is sensitive to the action of PTH, CT and dibutyryl cyclic AMP in stimulating proteoglycan synthesis as estimated by the incorporation of radioactive inorganic sulfate. PTH treatment of the cartilage resulted in dramatic stimulation of the proteoglycan synthesis in the diaphysis, which is composed of rather old non-dividing hypertrophied chondrocytes with much lower proteoglycan-synthesizing activity than younger dividing cells in the epiphysis and metaphysis. No or slight stimulation was observed the epiphysis and metaphysis. Dibutyryl cyclic AMP produced much the same effect. CT also stimulated the proteoglycan synthesis only in the diaphysis but the stimulation was less potent (133% of control) than PTH (650% of control) or dibutyryl cyclic AMP (625% of control). The hormones and dibutyryl cyclic AMP, therefore, seem to activate the proteoglycan synthesis specifically in rather old chondrocytes of the diaphysis. A synergistic effect of PTH and CT suggesting a different stimulating mechanism was observed in the diaphysis.

[Alpha particle excited X-ray fluorescence analysis for trace elements in cervical spinal cords of amyotrophic lateral sclerosis (author's transl)].

The mean contents of trace elements in anterior gray horn section of cervical spinal cords of six amyotrophic lateral sclerosis (ALS) cases were relatively determined against those of six control cases by alpha-particle excited X-ray fluorescence analysis. The anterior gray horn section of cervical spinal cord samples were excited by 1.6 MeV alpha-particle beam of 2 mm diameter accelerated with a Van de Graaff accelerator, and characteristic X-ray spectra were measured with alpha Si(Li) detector. From the peak area on the X-ray spectra, the relative mean contents of the trace elements in cervical spinal cords of ALS and control cases were determined. As a result, the X-ray peaks of Al, Si, P, S, Cl, K, Ca, Ti, V, Mn, Fe, Cu and Zn were detected. The contents of Al, Si, P, Ca, Ti, V, Mn and Fe in ALS cases were higher than those in control cases. The contents of S, Cl, K, Cu and Zn in ALS and control cases were equal to each other within standard deviation. The precipitation mechanisms of Al, Si, P, Ca, Ti, V, Mn and Fe into cervical spinal cord of ALS cases are discussed on the basis of the previous studies.

Study on trace impurity elements in highly purified iron and aluminum by neutron activation analysis.

Activation analysis was carried out on the purified iron and aluminum to study the behavior of trace impurities on purification process, and to estimate the purity of the purified samples. Thermal neutrons with high Cd ratio through D2O column or graphite column were used for the activation of samples to reduce the interference effects by fast neutrons, the reactions, (n, p) or (n, a), from matrix elements. On the zone refined aluminum, the segregation of impurity elements was cleary observed. In the case of iron, decrease of impurity content was confirmed on superposing the various purification processes.

A preliminary study of chemotherapeutic treatment for bladder tumors.

Our objective was to develop new methods of chemotherapeutic treatment for bladder tumors. Once or twice a week under cystoscopy 10 to 20 mg, doxorubicin hydrochloride were administered directly into the tumors. This procedure was repeated until the size of the tumors was reduced or had disappeared completely under cystoscopic study. A new type of flexible needle was fabricated for this purpose. In 9 patients studied the total success rate was 78%. No significant side effects were seen in any of the patients.
