Comparative studies on the uridine-5'-diphosphate-galactose: glycoprotein galactosyltransferase activity in rat liver and Zajdela ascitic hepatoma.

We report here data on the galactosyltransferase activity in serum, liver and Zajdela ascitic hempatoma cells and about the rate of galactose attachment to appropriate acceptors by beta(1-4, beta(1-3) and alpha(1-3) linkages in liver and hepatoma cell homogenates. It was established that in serum of Zajdela bearing rats, in host liver and in Zajdela ascitic cells, galactosyltransferase activity towards ovomucoid was elevated in comparison with control serum and liver. The activity towards low molecular acceptor (N-acetylglucosamine) in liver and hepatoma was nearly the same. The predominant isomer synthesized in both tissues was beta 1-4-galactose-N-acetylglucosamine. When asialofetuin was used as acceptor the activity of alpha(1-3) galactosyltransferase was measured. In Zajdela ascitic hepatoma cells alpha(1-3) galactosyltransferase activity was 3 times higher than that in liver. The elevated ability of the tumor cells to attach galactose residues with alpha(1-3) linkage to Gal beta 1-4GlcNAc-R sequence should be considered as a characteristic feature of these malignant cells.

[Uterine sonography as a method for the early detection and screening of endometrial carcinoma in women in the postmenopause with and without diabetes mellitus].

An attempt was made to examine the efficiency of the uterus sonography as a method for screening of endometrial cancer in postmenopausal women with or without diabetes mellitus for ten years period. 150 patients with diabetes mellitus and 150 patients from control group without diabetes mellitus were subjected to progesterone ampule 250 mg im. After bleeding occurred there were performed uterus sonographies before the abrasion probatoria, after the abrasion probatoria and on the 6th and 12th months. The performed abrasion probatoria showed full correspondence with exception of 3 cases. In 3 patients was found polipus endometrill, in one--atypical glandular hyperplasia, one case with carcinoma in situ and two cases with endometrial cancer.

Partial purification and characterization of cytidine-5'-monophosphosialate synthase from rainbow trout liver.

Trout liver is a rich source of sialate cytidylyltransferase activity. Three procedures are described by which the enzyme was enriched between 67- and 647-fold with high specific activities varying between 0.67 and 1.88 U/mg protein. In the simplest procedure studied, 100,000 x g supernatant of liver homogenate was chromatographed on Q-Sepharose and beta-[3-(2-aminoethylthio)propyl]-N- acetylneuraminic acid as affinity matrix, leading to an enzyme preparation (0.67 U/mg protein) well suited for the synthesis of CMP-N-acetylneuraminic acid. The synthase has a molecular mass of 160 kDa, a temperature optimum of 28 degrees C, a pH-optimum of 9.3 and exhibits Km-values for CTP, N-acetylneuraminic acid and N-glycoloylneuraminic acid of 1.7 mM, 2.1 mM and 2.9 mM, respectively. It is inactive with N-acetyl-9-O-acetylneuraminic acid. The enzyme is inhibited by CMP, CDP and 2'-deoxy-CTP. The sialic acid fraction of trout liver after hydrolysis is composed by N-acetylneuraminic acid (86%), N-acetyl-9-O-acetylneuraminic acid (12%) and N-acetyl-9-O-lactoylneuraminic acid (2%).

[The early detection and screening of endometrial carcinoma by the progesterone test and uterine sonography].

An attempt was made to examine the efficiency of the progesterone test and uterus sonography for ten-years period. The progesterone test was used for 621 patients, most of them from the University Obstetrics and Gynaecological Hospital in the town of Varna. Department of Obstetrics and Gynaecology as well as at the National Oncological Centre, Gynaecological Clinic--Sofia during the last few years. There have been found out deep knowledge in epidemiology, etiology and the present possibilities for early diagnostics of the endometrial cancer. All patients of a total number of 621 were in the postmenopausal period and received Progesterone as one-time medical administration. In 44 women bleeding occurred, and 31 of them agreed to undergo abrasio probatoria. 354 out of these 621 patients were examined by ultrasound B-picture after about 6 months to 1 year. 31 cases needed to be cleared-out additionally. The abrasio probatoria done later showed full correspondence to the ultrasound images with the exception of 6 cases. In 15 cases was found polyps endometrial, in 9 cases--atypical glandular hyperplasia, in one case--carcinoma in situ and in 6 cases--early endometrial cancer.

[Artificial abortions in Bulgaria and their prevention].

Authors go into one extremely important problem for the family, woman, natality and family planning in Bulgaria. Scarcely during the last years it can be observed a slight reduction of the frequency of induced abortions. The average age of women having an abortion is high--about 30, which shows, that induced abortion is still a method of regulation fertility in intergenetic interval. The rate of those who have induced abortion under the age of 19 in Bulgaria is 7.8 to 53.3%, in Czechoslovakia, 6.7%, in DDR--50.0%, Greece--68%, Hungary--8.9%, Sweden--15.8%. The rate of done again induced abortion is higher, that is a result of bad preventing activities during the first abortion. Similar is the tendency of growing number of unmarried women having an induced abortion, as it is in the other countries: from 16.4% in 1980 to 21.7% in 1987. Bulgaria is on the third place in Europe according to the absolute number of dead cases after abortion (maternity death, after Rumania and USSR). It can be mentioned that in Bulgaria women do not use the most modern and safest devices, medicines for interruption of pregnancy.

Influence of demetallization of hydrogenated sunflower oil on changes in the forms of heavy metals and their pro-oxidative effects.

The effect of removing heavy metals from hydrogenated sunflower oil using a method based on changes in the contents of the various forms of the heavy metals copper, iron, zinc and nickel and their respective pro-oxidative effects was studied. A high degree of removal of dissociated and bounded ions was observed for hydrogenated oil, while there was no change in the contents of the co-ordination form. The pro-oxidative effect of the dissociated ions and of the bound metal ions in hydrogenated oil decreased nine-fold and seven-fold, respectively, compared with that of the non-hydrogenated oil, as a result of which the oxidation stability of hydrogenated oil increased two-fold. Comparative studies confirmed as appropriate the use of wash waters and auxiliary solutions that do not contain heavy metals for the processing of the hydrogenate after the classical technology.

[The surgical treatment and results in patients with leukoplakia vulvae].

For period from 1976 to 1988 in the Clinic of Gynecology were treated 78 patients (women--31-74 years old) with leukoplakia vulvae. 61 patients (77.2%) were subjected to classical simple vulvectomy. 18 patients (22.8%) were subjected to simple vulvectomy according to Abitbol. Per primam was closed operative sore at 12 patients, operated by classical vulvectomy and at 8 patients, operated according to Abitbol. Per secundam was closed operative sore at 49 women from first group and at 10 from second group. Stenosis vulvae was shown at 4 women (5%) and at 7 (8.9%) was developed recurrence, who was applied to accomplish a repeated treatment. Good results were educated from vulvectomy according to Abitbol. This vulvectomy was recommended from authors.

[Cardiotocographic control during labor].

The authors discuss different aspects of intrapartum foetal monitoring. The scientific basis of foetal intrapartum foetal monitoring are pointed out. The authors present three groups of cardiotocographic signs which should alarm the obstetricians: warning signs, threatening signs and clear signs for foetal distress. The authors discuss also the specific characteristics of the cardiotocographic record of premature fetus during pregnancy and labour. It is pointed out that the intrapartum foetal monitoring and the correct interpretation of the cardiotocographic record lead to better perinatal results.

[The posttherapy survival of patients with invasive cancer of the vulva].

The treatment results and five-years survival are studied in 157 patients with invasive vulvar cancer treated for the period of 1979-1988 in Gynaecological Clinic of the National Oncological Center Institute in Sofia. The general five years survival is 59.2%. The highest is the survival of the patients with first clinical stage--79.55%. The best treatment results are by patients treated with radical vulvectomy and regional lymph node dissection, without histological founded metastases in the extirpated lymph nodes--69.24% five years survival. The authors show some better results because of the better early diagnosis of the disease and the improved therapeutical methods.

gamma-Glutamyltranspeptidase and dipeptidyl peptidase IV activity in the serum of normal and hepatoma-bearing chickens and in the plasma membranes from liver and hepatoma Mc-29.

Investigations on the activity of gamma-glutamyltranspeptidase (GGT) and dipeptidyl peptidase IV (DPP IV) in the serum of healthy chickens and those bearing hepatoma Mc-29, and in liver and hepatoma plasma membranes were carried out. There was no difference in the serum enzyme activities of control and tumor-bearing chickens but the activity of GGT was twice higher and that of DPP IV 20 times lower in hepatoma plasma membranes than in chicken liver plasma membranes. Using thin-layer analytical isoelectric focusing in agarose gels it was established that the pI range of GGT from host serum and hepatoma plasma membranes was shifted to more acidic values. This could be interpreted as a specific feature for this enzyme considered as a tumor marker.

[The antenatal diagnosis of the sex of the fetus by the cytological determination of sex chromatin].

Forty women between 17 and 23 weeks' gestation were included in the study, whose interruption of pregnancy was performed by medical and social indications. The method of Aburel-Atanasov was used as in 10 ml of the removed amniotic fluid sex chromatin was examined cytologically on cellular smears. Staining and determination of sex chromatin was made by the method of Dokumov. The fetus of male sex, when there was no sex chromatin, but the sex was female, when it was present in various percentage of cells. The genuine sex of the fetus was determined after interruption of pregnancy. Coincidence of cytological and genuine sex of the fetus was found in 36 cases (92.31%), but the sex was determined not exactly in 3 cases (7.69%). There was no sufficient material for preparation of high-grade cytological smears in one case. 14 out of 40 fetuses were of female sex (with sex chromatin in 28.57% of cells on the average) and 25 of male sex. The significance and advantages of the method in comparison with other methods for determination of fetal sex at the same term of pregnancy are discussed.

[A modified Sugiura operation].

This is the first report in Bulgaria of the application of the modified method of Sugiura. The operation is less traumatic than the original one, the postoperative mortality is essentially reduced, patient survival to 7 years is increased and there is no encephalopathy. This operation is technically easier to perform than splenorectal anastomoses and may be performed both as planned and emergency intervention. Two personal observations on two patients operated by the modified technique of Sugiura are reported. The results were very good.

[Our initial experience with Shouldice's hernioplasty].

For the first time in Bulgaria the authors herniotomy of inguinal hernias by the method of Shouldice. This method is anatomo-physiological and is pathogenetically well-grounded. Shouldice's method is considered the method of choice in inguinal hernias.

[A modified Sugiura operation].

The modified method of Sugiura is reported for the first time in Bulgaria. The operation is less traumatic than the original one, the postoperative case fatality rate is lower, patient survival rate up to 7 years is high and there is no encephalopathy. This operative technique is more practicable than splenorenal anastomosis and may be applied both for planned and for emergency operations. Two patients have been operated by the authors, using the modified technique of Sugiura, with very good results.
