Cytogenetics of juvenile type chronic granulocytic leukemia.

Cytogenetic examination of bone marrow cells from three patients with juvenile chronic granulocytic leukemia (CGL) showed 46,XX,3p-,11p+ ,t(11p:3p) in one case, 45XY,-E in another, and 45X(X),-C/47,XX,+G in the third. The case with the translocated chromosome originally presented like an acute lymphocytic leukemia (ALL). TAn overt clinical picture of juvenile CGL emerged two and a half years later. Serial study of this case revealed no cytogenetic abnormalities until two years after the diagnosis of ALL, when the translocated chromosome was first observed. Unlike the Ph1 chromosome in adult type CGL, chromosomal abnormalities in juvenile CGL lack specificity, resembling ALL in this respect.

Comparison of metabolic alterations in hypothalamic and high fat diet-induced obesity.

The heterogeneous nature of the experimental obesities induced by ventromedial hypothalamic (VMH) lesion and high fat diet (HFD) have been demonstrated by comparing VMH-lesioned and sham-operated rats fed a HFD or low fat diet (LFD). VMH rats had increased fat mass serum insulin and serum triglycerides but lower serum glucagon and smaller salivary glands than sham-operated animals. The body weight of HFD obese rats was intermediate between VMH and sham-operated animals on the LDF. Liver and fat pad weights showed effects of lesions and diet. Diet did not affect plasma glucagon or insulin. Pair-feeding VMH rats with sham-operated rats prevented weight gain but did not prevent the increase in insulin and fall in glucagon. Studies of insulin secretion from isolated perifused islets showed that basal and both phases of stimulated secretion were significantly increased in VMH groups. The changes in plasma insulin, plasma glucagon, and salivary gland weight in VMH groups are interpreted as showing decreased activity of the sympathetic nervous system following VMH lesions.

Pulmonary capillary permeability to HRP in dogs: a physiological and morphological study.

Pulmonary capillary permeability was studied in 10 normal anesthetized dogs using horseradish peroxidase (HRP) as tracer. Physiological studies involved measurement of left ventricular, pulmonary arterial, and wedge pressures, and of right lymphatic duct (RLD) and thoracic duct lymph flows. Plasma and interstitial oncotic pressures were calculated from protein measurements. After 1 h of lymph collection, HRP was injected intravenously at three total dose levels, low (20-30 mg/kg), medium (55 mg/kg), and high (110-120 mg/kg) as a bolus followed by smaller doses over 1.5-3 h. HRP was measured colorimetrically in plasma and lymph and appeared in the first RLD lymph sample collected at all dose levels. Electron microscopic studies to examine the passage of HRP from plasma to RLD, while less sensitive in that HRP was seen only at the high dose, showed that it crossed pulmonary capillaries under normal physiological conditions. Freeze-cleaving studies suggest this may have occurred through pathways in the interendothelial space caused by discontinuities in the junctional strands of tight junctions.

The effects of subdiaphragmatic vagotomy in rats with ventromedial hypothalamic obesity.

The present study investigated the effects of subdiaphragmatic vagotomy in rats with ventromedial hypothalamic (VMH) lesions and obesity. Vagotomy or sham-vagotomy was performed two weeks after VMH lesions and rats were observed for 4 more weeks. Complete vagotomy reversed the VMH obesity, lowered serum insulin, lowered basal gastric acid and blocked the secretion of gastric acid after stimulation of the cervical vagus. Pair-feeding of VMH-lesioned rats without vagotomy to the food intake of vagotomized animals also reversed the obesity, lowered serum insulin and lowered basal acid secretion, but it did not prevent the rise in acid after vagal stimulation. These results suggest that subdiaphragmatic vagotomy reversed the obesity of VMH lesioned rats primarily by decreasing food intake. However, there was a positive correlation (r = .70) between the level of serum insulin and basal gastric acid in VMH lesioned rats which remained significant when the effects of food intake were held constant (partial correlation coefficient = 0.449). This supports the possibility that ventromedial hypothalamic injury is followed by enhanced vagal activity and that the vagus may play an important part in the hyperinsulinemia of VMH obesity.

The electrical induction of callus formation and external skeletal fixation using methyl methacrylate for delayed union of open tibial fracture with segmental loss.

A case of delayed union of an open tibial fracture with segmental loss is presented in which electricity was applied to the bone defect through two screws that were connected to a methy methacrylate skeletal fixation device. The fracture developed electrically induced callus formation around the cathode, callus extended to the anode, and solid bony union ensued. External skeletal fixation using methyl methacrylate is convenient and is a suitable adjunct to electrical stimulation of fracture healing.

Tonofilaments in normal human bronchial epithelium and in squamous cell carcinoma.

Tonofilaments in the cells of squamous cell carcinoma of human bronchial epithelium were observed and compared to those in normal bronchial epithelium in 2 patients. In the normal tissue, only small numbers of tonofilaments were observed, and these were found only in the basal cells of the epithelium. In the tumor cells, the quantity of filaments increased considerably. They appeared in the cytoplasm both as branching and anastomosing bundles (tonofibrils) and also as single filaments about 40 A in thickness. These bundles were particularly well developed in the core of narrow cytoplasmic processes of tumor cells. The thickness and appearance of single tonofilaments were similar to those of the cytoplasmic microfilaments also observed in this study. It is suggested that the development of tonofilament bundles in the cytoplasmic processes of tumor cells may provide direct motive force for the deformation of the shape of the cell and potential invasiveness of these tumor cells.
